In order to asses population fitness and conservation status, genetic diversity among and within population was estimated using ISSR markers.A set of 6 primers were utilized after selection from a primer set which showed clear and reproducible banding patterns, generating a total number of 86 loci. A presence/absence matrix was generated by evaluating the gel electrophoresis banding patterns of PCR products. From this, genetic distances between individuals and populations were calculated, which were used in all subsequent generation of genetic diversity indices, assuming the Hardy-Weinberg equilibrium. Result revealed a low expected heterozygosity for both populations. Further testing by constructing an UPGMA dendrogram and a PCoA analysis confirmed that populations are clearly separated and with levels of genetic diversity that could ensure population survival in time and space.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2541-2547"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"43485627","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The present paper deals with optimization of anthocyanins extraction from Paeonia officinalis L. flowers and their application in cotton eco-dyeing processes. The results on extraction and mathematical models indicate improved extraction by ultrasonication at high solvent/solid ratio (50/1) compared to that by maceration. Among the investigated extraction solvents, the most efficient was 70% ethanol. Cotton dyeing was performed via conventional procedure and ultrasonication, in the presence and absence of classic mordant (copper sulphate) and biomordants (tannic and citric acids). A successful dyeing was obtained according to ATR-FTIR analysis of mordanted dyed samples. The colour properties of dyed cellulosic substrates as determined by CIELAB system showed higher redness values and large positive differences in chroma in samples dyed with peony extract by exhaustion in presence of high concentration of citric acid.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":" ","pages":""},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"46362335","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Dairy products contain nutrients essential for health, of which vitamins could help reduce the risk of certain diseases. Cheese is a highly nutritious food that can play an important role in a wellbalanced diet. The aim of this research was to investigate the vitamin composition of various types of cheese and to evaluate the vitamins intake provided by cheese as part of the daily diet. The determinations were performed on traditional Romanian cheese, meaning cow, sheep, goat and buffalo Telemea and some Kashkaval types purchased from local supermarkets. The samples were subjected to vitamins B1, B2, B12, A and E analysis using spectrofluorometric, spectrophotometric, HPLC and microbiological methods. The results showed a higher content of vitamins B2 and E in Telemea cheese while Kashkaval types were distinguished for vitamin B1, B12 and A. Regarding the vitamin content depending on raw material used for obtaining the different types of cheese, those made from goat milk recorded the highest level of the analyzed vitamins, followed by those from sheep milk, while lowest amounts were found in products","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2375-2383"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"46869410","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The oils were then used in the development of hydrogels based on nanostructured lipid carriers (NLC) with therapeutic effect on varicose veins. Several vegetable oils (pumpkin seed oil, wheat germ oil, evening primrose seed oil, castor seed oil, grape seed and jojoba oil) were analyzed by gas chromatography coupled with mass spectrometry (GC-MS), establishing the profile of saturated and unsaturated fatty acids. Antioxidant activities were determined by the FRAP (Ferric Reducing Antioxidant Power) method and were correlated with the composition ratio of saturated and unsaturated fatty acids. It has been established that pumpkin seed oil and wheat germ oil are most indicated in the development of NLC-based hydrogels.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2370-2374"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"48294657","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Antibacterial, antioxidant and phenolic content of nine medicinal plants (Achillea millefolium, Prunella vulgaris, P. laciniata, Lythrum salicaria, Epilobium angustifolium, E. hirsutum, Pedicularis comosa, Agrimonia eupatoria and Verbena officinalis) were investigated in connection with folkloric usages. Aerial parts of nine plant species were extracted with water and methanol (MeOH). Disc diffusion method was performed to evaluate the antibacterial activity of the extracts against ten pathogenic bacteria. Antioxidant activity was determined by using 2,2-diphenyl-1picrylhydrazil (DPPH) radical photometric assay. Total phenolic and flavonoid content were investigated by using Folin-Ciocaltaeu and aluminum chloride (AlCl3) colorimetric method, respectively. Quantitative analysis of phenolic constituents of nine plants species were performed by high performance liquid chromatography-diode array detector (HPLC-DAD) via chosen ten phenolic standards (gallic acid monohydrate, caffeic acid, rutin hydrate, luteolin-7-O-β-D glucoside, kaempferol, rosmarinic acid, myricetin, quercetin, coumarin and apigenin). E. hirsutum showed the best antibacterial activity against gram-positive bacteria (S auerus, S. epidermidis and S. pyogenes). L. salicaria also demonstrated strong and broad spectrum antibacterial activity. P. laciniata, L. salicaria and E. angustifolium showed potent antioxidant activity. The highest phenolic and flavonoid content was observed with E. hirsutum and A. millefolium, respectively. L. salicaria also had very high amount of total phenol and flavonoid content. HPLC-DAD analysis displayed that P. laciniata and A. eupatoria were the best sources of rosmarinic acid. Furthermore, V. officinalis and P. vulgaris also had remarkable amount of rutin. This study revealed the scientific rationale behind the traditional knowledge of the tested plants. Consistent with traditional usages, the most prominent plants were L. salicaria, E. hirsutum and E. angustifolium in regard to strong antibacterial and antioxidant potentials. These plants may be proper natural sources with potential applications in pharmaceutical and food industry.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2499-2510"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"48493565","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Some ornamental (morphologic and phenologic) features were analyzed and also the anatomic structure of the leaf, the content of photosynthetic pigments and the enzymatic activity of the leaves. The biometrical and phenological determinations indicate a good ecological adaptability of the plants, in accordance with similar reports. The anatomical differences between the three species are not very great, but the stomata in E. stenophillus and E. himalaicus are arranged at the same level as the epidermal surface while in E. robustus they are arranged a little lower, and the assimilating parenchyma layer is more developed in E. stenophillus and E. himalaicus than in E. robustus. At E. robustus and E. himalaicus a larger content of assimilatory pigments and a more reduced enzymatic activity were observed.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2489-2498"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"49391272","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In our study, relative expressions of MALAT1, miR-494-3p and PTEN in HemECs (HemEC) were determined using qRT-PCR methods. MTT assays were used to measure cell viability. The rate of cell apoptosis was assessed using caspase-3 assay. Transfections were performed to mediate lncRNA MALAT1 and miR-494-3p expression in HemEc cells. Also, Bioinformatic analysis and Luciferase reporter were used to predict and validate the bindings between MALAT1 and PTEN, and PTEN and miR-494-3-p. MALAT1 was highly expressed in HemECs. Cell proliferation increased significantly due to MALAT1 overexpression in HemECs while MALAT1 overexpression significantly reduced cell apoptosis in HemECs. On the other hand, contradictory results were observed due to the reduction of MALAT1 in HemEC. We also found that MALAT1 interacts with miR-494-3p/PTEN to mediate cellular functions. Collectively, the results showed that the MALAT1 expression was negatively associated with miR-494-3p and positively matched the PTEN expression. In addition, MALAT1 acted as a ceRNA by binding with miR-494-3p to up-regulate PTEN in HemECs. Our study showed that MALAT1 accelerates the proliferation of HemEC cells by controlling the miR-494-3p/PTEN axis promoting new insight into IH treatment.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2535-2540"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"49488780","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The secondary objective was to investigate the effect of the socioeconomic status on the preoperative COMQ-12 scores in individuals with a similar stage of disease. Methods: This was a cross-sectional study. Patients diagnosed with different stages of (COM) with cholesteatoma, scheduled for the first surgery, filled in the Romanian translated version of the COMQ-12. The patients were divided according to the stage of the disease and the environment of origin: rural versus urban areas. Results: The internal consistency of the Romanian COMQ-12 proved excellent, with a Cronbach’s alpha equal to 0.959. The correlation of the 24 randomly split-half reliability analysis varied from 0.904 to 0.917, with a mean of 0.908. Significantly higher COMQ-12 scores are obtained by patients from urban (mean = 42.14, SD =9.04) as compared to those from rural (mean = 30.44, SD = 5.88) only for those with stage III disease (P-value=0.0292). Conclusions: Romanian version of COMQ-12 proved to be a valid instrument that can be use in clinical practice to provide suitable information in COM patients, which contributes to the development of an international common otology dataset. Patients from rural areas tend to disregard their symptoms, being more predisposed to potential complications related to COM rather than urban patients at the same stage of the disease.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2455-2463"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"46727849","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The implementation of an energy strategy to harness the potential of renewable energy sources (RES) is part of the coordinates of Romania’s medium – and long-term energy development and provides the appropriate framework for the making of decisions on energy alternatives and the inclusion in the Community acquis in the field. In this respect, a study on the biomass potential of Timiş County and on the possibilities of producing unconventional energy from biomass has been carried out. The study is based on research, data collection from the literature, as well as from official documents or official websites, the processing and interpretation of the data and their quantitative and qualitative analysis. It was concluded that biomass is a promising renewable energy source for Romania, both in terms of potential and in terms of usability.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2434-2440"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"42971080","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The response of several apricot hybrids was evaluated over the vegetative period of 2010 by visual monitoring of symptom development and by serological and molecular methods. After two cycles of study, all the replicates of ‘Mari de Cenad’, ‘Traian’ and ‘Tabriz’ showed sharka symptoms while the replicates of ‘Stark Early Orange’ and NJA 2 did not show any symptoms and were not ELISA-positive. The resistant progenitors (‘Stark Early Orange’, NJA17, NJA42 and NJA2) were able to transmit PPV resistance to their descendants, in agreement with previous results (DOSBA et al, 1994, MARTINEZ-GOMEZ et al, 2000; AUDERGON et al, 1994). The genetic control hypotheses for PPV resistance in apricot referenced by different authors considered the resistance allele as dominant. Romanian apricot F1 and F2 progenies evaluated were classified into two groups: susceptible to PPV and resistant to PPV. After that the most important genotypes were grafted onto infected GF 305 (considered as indicator for PPV) in greenhouse conditions. The final step is the introduction and development of the marker-assisted selection (MAS) in PPV resistance F1 and F2 progenies in apricot Romanian genotypes. DNA was isolated from all hybrids and parent genotypes. These samples will be used to implement the molecular markers for identification of genes linked to plant response to PPV infection.","PeriodicalId":21566,"journal":{"name":"Romanian Biotechnological Letters","volume":"26 1","pages":"2428-2433"},"PeriodicalIF":0.0,"publicationDate":"2021-02-02","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"48737665","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
