However, their role as Cry receptors in dipteran species remains unknown. Here, we identified the ABC transporter orthologs in the Aedes aegypti genome corresponding to the Cry toxin receptors described in lepidopteran and coleopteran species. Analysis of their transcriptomic regulation in gut tissue, revealed the expression of ABCA2, ABCB1, and ABCC2. Using CRISPR-Cas9, we knocked out (KO) the expression of these three ABC transporter genes and performed in vivo binding assays and bioassays with four mosquitocidal Cry toxins (Cry4Aa, Cry4Ba, Cry11Aa, and Cry11Ba). Histopathological analysis showed a significant reduction of Cry toxins binding to the apical microvilli membrane of gut epithelium, depending on the ABC KO mutation. However, similar larvicidal activity was observed for Cry toxins between ABC KO and wild type strains. Transcriptomic analysis revealed specific upregulation of their corresponding ABC transporter paralogous, suggesting a compensatory mechanism that could mitigate the fitness cost of the ABC KO and potentially contribute to redundant receptor function of Cry toxins. The predominant subtype of avian influenza virus was H9 (accounting for 54.67%). The peak of positive influenza virus detection rates occurred in winter and spring seasons every year. The venue with highest positive rate was poultry slaughtering and processing plants (41.83%), followed by urban and rural live poultry markets (35.48%); among all types of specimens, the highest positive rate was detected in swab specimens from the surfaces of poultry slaughtering or display tables (47.37%), followed by wastewater from poultry washing (45.83%), and surfaces of cages (27.65%).</p><p><strong>Conclusion: </strong>The contamination of avian influenza virus in the poultry environment in Huzhou City is relatively severe, with diverse subtypes. A total of 260 pregnant women with SARS-CoV-2 infection at any pregnancy trimester were enrolled in a prospective follow-up study. Clinical features were recorded between the SARS-CoV-2 infection diagnosis towards delivery and postpartum period. ANOVA and Chi-square/Fisher tests, Bi- and multivariate analyses were performed to verify the effect of predictors on disease outcome, adjusted for the pregestational variables. Data demonstrated that anosmia (64.6%), nasal congestion/discharge (61.5%), headache (60.8%), ageusia (58.5%) and myalgia (58.5%) were the most common symptoms observed amongst pregnant women with non-severe COVID-19. Fever (44.6%) and dyspnea (36.5%) were associated with higher disease severity. Gestational diabetes mellitus (35.8%), systemic arterial hypertension (18.1%), preterm delivery (11.5%) and superimposed preeclampsia (6.2%) were reported as adverse pregnancy outcomes amongst pregnant women with COVID-19. Parturients with acute COVID-19 and pregnant women infected at the 3rd trimester presented more severe or critical outcomes as compared to those infected at 2nd and 1st trimesters. Preterm labor (Odds Ratio = 3.64), acute fetal distress (Prevalence Ratio = 2.40) and Apgar 1st minute score ≤ 7 (Prevalence Ratio = 2.56) were adverse outcomes reported in parturients with acute COVID-19 and those with severe or critical outcomes. Together these findings demonstrated that SARS-CoV-2 infection during pregnancy was associated with relevant maternal and neonatal adverse outcomes. Thus, we conducted a network meta-analysis to provide evidence-based insights for clinical practice.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Methods: &lt;/strong&gt;Databases such as PubMed, Excerpt Medica Database (Embase), Cochrane Library Central Register of Controlled Trials, APA PsycInfo, China National Knowledge Infrastructure Database, Wanfang, and SinoMed were used to retrieve randomized controlled trials (RCTs) on electromagnetic therapy for PSI, with a search deadline of Sep 2024 for each database. The Cochrane bias risk assessment tool was used to evaluate the quality of the included RCTs. Stata was used for network meta-analysis.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Results: &lt;/strong&gt;We included 28 RCTs involving 2353 patients across 12 different treatment regimens. The surface under the cumulative ranking results showed that the ranking of Pittsburgh sleep quality index decline was: cranial electrotherapy stimulation&gt;low frequency repetitive transcranial magnetic stimulation (LF-rTMS)&gt;infra-low frequency repetitive transcranial magnetic stimulation (ILF-rTMS)&gt;fastigial nucleus stimulation (FNS)&gt;transcranial direct current stimulation (tDCS)&gt;low frequency electric stimulation (L-FES)&gt;high frequency repetitive transcranial magnetic stimulation (HF-rTMS)&gt;middle frequency repetitive transcranial magnetic stimulation (MF-rTMS)&gt;sham stimulation (SS)&gt;common treatment (CT); Ranking of Hamilton depression scale decline degree: HF-rTMS &gt; LF-rTMS &gt; tDCS &gt; SS&gt;continuous theta-burst stimulation (cTBS)&gt;MF-rTMS &gt; CT; national Institute of health stroke scale decline ranking: HF-rTMS &gt; LF-rTMS &gt; SS &gt; L-FES&gt;electroencephalographic biomimetic stimulation&gt;CT &gt; cTBS; Clinical total effective rate ranking: LF-rTMS &gt; FNS &gt; ILF-rTMS &gt; L-FES &gt; CT&gt;repetitive transcranial acupuncture stimulation.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Conclusions: &lt;/strong&gt;Different electromagnetic therapies can effectively improve sleep quality in PSI patients, and the efficacy and safety of LF-rTMS are significant. However, owing to the limitations of this study, the efficacy ranking cannot fully explain the advantages and disadvantages of clinical efficacy. In the future, additional multicentre, large-sample, double-blind, clinical, and randomized controlled trials are required to supplement and demonstrate the results of this study.&lt;/p&gt;&lt;p&gt;&lt;strong&gt;Strengths and limitations of this study: &lt;/strong&gt;This is the first study to conduct network meta-analysis on PSI treatment with different electromagnetic therapies. Simultaneously, we refined the classification based on different frequency patterns of the same therapy, and the results can serve as a reference for clinical workers. Semaglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, has shown potential neuroprotective effects. This study investigates the efficacy of semaglutide in ameliorating cognitive dysfunction in a mouse model of T2DM.</p><p><strong>Methods: </strong>Male C57BL/6J mice were fed a high-fat diet for four weeks and received a single intraperitoneal injection of streptozotocin (150 mg/kg) to induce T2DM. All mice were divided into four groups: control, diabetes control (T2DM), semaglutide treatment (semaglutide, 0.1 mg/kg) and dapagliflozin treatment (dapagliflozin 1 mg/kg). Cognitive function was assessed using the Morris water maze (MWM) test. Histomorphological analysis of hippocampal tissues was performed using H&E and Nissl staining. Immunofluorescence was used to assess LRP1 expression and apoptosis. Biochemical analyses measured oxidative stress markers (SOD, MDA) and inflammatory cytokines (IL-1β, IL-6, TNF-α, CRP).</p><p><strong>Results: </strong>Semaglutide treatment significantly reduced blood glucose levels in diabetic mice. In the MWM test, semaglutide-treated mice showed reduced escape latencies, indicating improved spatial learning and memory. Histomorphological analysis revealed preserved neuronal structure in the hippocampus with reduced neuronal damage and apoptosis in the semaglutide-treated group. Immunofluorescence showed increased LRP1 expression and decreased apoptosis. Biochemical analyses indicated that semaglutide reduced oxidative stress and inflammatory markers, further supporting its neuroprotective effects.</p><p><strong>Conclusions: </strong>Semaglutide effectively ameliorates cognitive dysfunction in T2DM mice, likely through mechanisms involving the reduction of oxidative stress, inflammation, and neuronal apoptosis. These findings suggest that semaglutide has potential as a therapeutic agent for managing diabetes-associated cognitive decline. This overview systematically consolidates existing evidence to assess the clinical viability of physical activity as a scalable supplementary therapy for asthma management.</p><p><strong>Methods: </strong>We searched 12 electronic databases to identify systematic reviews (SRs) from inception until February 12, 2025, concerning the efficacy of physical activity in asthma management. Literature was independently reviewed, data extracted and verified by two researchers. A third author was designated to mediate any disputes concerning screening. The methodological quality of SRs was assessed using the A Measurement Tool to Assess SRs 2 (AMSTAR 2) checklist, and the certainty of evidence for key outcomes was evaluated using the Grading of Recommendations Assessment, Development and Evaluation (GRADE) framework.</p><p><strong>Results: </strong>This study analyzed 34 SRs (published 2000-2024) that included quantitative synthesis involving 113-2280 participants from asthma populations: adults (9 SRs), children (8 SRs), and mixed adult-child cohorts (17 SRs), with disease severity varying from mild to severe. Ten SRs were assessed as moderate to high quality by AMSTAR 2, whereas the other SRs were classified as low or very low quality. We evaluated the quality of evidence for SRs utilizing the GRADE evidence quality assessment framework. Thirteen moderate-quality evidence, and 51 low or very low-quality evidence support the improvement of PA on the outcomes of asthma quality of life, asthma control, lung function, exercise capacity, and respiratory muscle strength.</p><p><strong>Conclusion: </strong>Engagement in physical activity has been demonstrated to markedly enhance asthma-related outcomes. Specific interventions provide targeted advantages in various domains: aerobic exercise enhances AQLQ scores and lung function metrics, including FEV1, FVC, PEF; breathing exercises improve AQLQ, FVC, and PImax; yoga correlates with enhancements in AQLQ, FEV1, and FVC; aquatic exercise results in increased FEV1; and inspiratory muscle training yields improvements in FEV1, FVC, PEF, and PImax. Nonetheless, there exists an imperative requirement for more stringent studies to fortify the existing evidence base. {"title":"The Relationship between students' engagement, grit, and teacher care in Chinese context.","authors":"Xin Wang, Jianguo Xiao","doi":"10.1371/journal.pone.0326485","DOIUrl":"https://doi.org/10.1371/journal.pone.0326485","url":null,"abstract":"<p><p>The significant growth in the field of English as a Foreign Language (EFL) education, along with the introduction of Positive Psychology (PP), has led to an increased emphasis on student engagement within the academic realm. Additionally, the focus on grit in language education highlights its importance in students' success throughout their educational journey. Besides, learners' engagement in academic domains is a noteworthy supporting the achievement of students in higher education. Also, the investigation of favorable communication actions has been examined as an important indicator of EFL learners' engagement and grit. However, the significance of teacher caring in predicting EFL learners' grit and engagement has been underresearched among Chinese EFL learners and this study makes an effort to consider it. To fill this gap, 401 EFL learners from 11 regions of China were requested to respond to three scales, namely engagement, grit, and teacher care. The data were collected and through Structural Equation Modeling (SEM), it was revealed that about 59 percent of changes in the EFL learners' grit can be predicted by their teacher care, and about 67 percent of changes in the EFL students' engagement can be predicted by their teacher care. One such fold, called the β-trefoil, is present in functionally diverse proteins including cytokines involved in the immune system such as interleukin-1. The unrecognizable sequence-level diversity, even among paralogs of interleukin-1 within the same chromosomal locus, suggests the resilience of this fold to mutational onslaught. Furthermore, β-trefoil domain containing-proteins are known to coexist with other domains to achieve functional diversity. In this study, we challenge the reach and limitations of function prediction using fold-fold comparison using β-trefoil fold as an example. We identified proteins containing β-trefoil fold belonging to thirty-two distinct functional classes based on diverse domain architecture and/or functional annotation by mining both the PDB and AlphaFold databases using fold-fold comparison. Among the proteins with novel domain architecture we find β-trefoil along with chitinase, lipase, β-glucosidase, protein kinase, peptidoglycan-binding + peptidase matrixin, glycosyl hydrolases family 3 + PA14 + fibronectin type- III, alpha galactosidase A, PhoD-like phosphatase, insecticidal crystal toxin, trypsin, alginate lyase and two novel structurally uncharacterized domains. We demonstrate that fold-fold comparison can extend function prediction beyond the reach of sequence-based approach and provides an opportunity to discover novel domain architecture associated with known folds. We aimed to evaluate and compare the prevalence of IUGR in Tehran using 3 common standards and identify the associated factors.</p><p><strong>Methods: </strong>Using the Iranian Maternal and Neonatal Network registry, we extracted the data of all singleton live births in Tehran province of Iran in 2018 to eliminate the possible confounding effects of the COVID-19 pandemic. We defined IUGR as having a birth weight less than the 10th percentile for gestational age using 3 standards including World Health Organization's and INTERGROWTH-21st charts and the same population's 10th percentiles. Logistic regression was used to identify the associated factors.</p><p><strong>Results: </strong>There were 187031 singleton live births. The prevalence of IUGR using WHO, INTERGROWTH-21st, and the population's 10th percentile was 11.8, 4.2, and 9.7 percent respectively; Among these, 7681 cases (4.1%) were identified by all 3. Neonatal trisomy 21, maternal addiction, eclampsia/pre-eclampsia, chronic hypertension, history of abortion, Primigravidity, being older than 35 yrs. and parental consanguinity were positively associated with IUGR where mother's gestational diabetes, higher education level, delivering the baby in a private hospital and living in Paakdasht or Shahryar cities were inversely associated with IUGR.</p><p><strong>Conclusion: </strong>The IUGR prevalence highly depended on the standards used ranging from 4.2 to 11.8%, showing a great need for a global consensus.
