Antioxidant activity was measured using DPPH photometric assay, and flavonoid content was quantified via the aluminum chloride method. The extract's neuroprotective effect was assessed using the pentylenetetrazole (PTZ)-induced seizure model, along with immunohistochemical (IHC) analysis and hematoxylin & eosin (H&E) staining. Methods The antioxidant potential of the extract was determined through DPPH radical scavenging activity, while flavonoid content was assessed as quercetin equivalents (QE). The in vivo antiepileptic activity was evaluated in mice using the PTZ-induced seizure model. Key biomarkers of oxidative stress, including glutathione (GSH), glutathione-S-transferase (GST), catalase, and lipid peroxidation (LPO), were measured to evaluate the extract's ability to ameliorate PTZ-induced oxidative stress. Results The extract demonstrated notable antioxidant activity, with 72% scavenging activity observed at 900 µg/mL. Flavonoid content was determined to be 39.45 ± 1.84 µg QE/mg. In the PTZ-induced seizure model, Achillea millefolium extract at 400 mg/kg and 600 mg/kg significantly reduced neuroinflammatory markers and oxidative stress, as evidenced by decreased LPO and increased levels of GSH, GST, and catalase. IHC and H&E staining revealed neuroprotective effects, including reduced neuroinflammation and enhanced neuronal survival. Conclusion The ethanolic extract of Achillea millefolium exhibited strong antioxidant and neuroprotective properties, effectively mitigating PTZ-induced seizures, oxidative stress, and inflammatory responses. These findings suggest its potential as a natural alternative for epilepsy treatment.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"20 2","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147332110","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Therefore, there has been increasing attention on the development of anti-alcoholism products. In this study, we evaluated the therapeutic benefits of an anti-alcoholism product (Tengcha–Gegen Plant Solid Beverage, which contains Ampelopsis grossedentata , Pueraria Lobata , Hovenia dulcis Thunb , and turmeric ) in an AAI mouse model. Methods The AAI mouse model was established through alcohol gavage. Male Kunming mice were first given three doses of Tengcha–Gegen Plant Solid Beverage for 30 min and then gavaged with 52% (V/V) liquor (0.15 mL liquor/10 g body weight). The impact of Tengcha–Gegen Plant Solid Beverage on AAI was evaluated by the sober-up time, the plasma alcohol content, and the plasma and liver superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) contents or activities. Results The Tengcha–Gegen Plant Solid Beverage could shorten the sober-up time and decrease plasma alcohol levels at 0.2 and 0.4 g/kg body weight. The mechanistic studies showed that Tengcha–Gegen Plant Solid Beverage increased the plasma SOD and liver GSH levels but decreased the plasma MDA levels at the indicated doses. Conclusions This study confirmed that Tengcha–Gegen Plant Solid Beverage might have anti-alcoholism therapeutic potential and protect the liver against high oxidative stress induced by acute alcohol exposure in AAI mice.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"20 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147884181","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, our previous studies showed that 6-HKA was difficult to penetrate the blood brain barrier (BBB) to exert its pharmacological effects in rats. Therefore, this study aims to investigate the pharmacokinetics and cerebral cortex distribution of 6-HKA in MCAO mice, to determine whether the ability of 6-HKA to cross the BBB is contingent upon barrier integrity. Methods This study established and validated the ultra-performance liquid chromatography tandem mass spectrometric (UPLC-MS/MS) methods for the quantification of 6-HKA in mouse plasma and brain, and investigated the pharmacokinetics and cerebral cortex distribution of 6-HKA in MCAO mice and sham-operated mice. Results This study demonstrates that the mean residence time (MRT 0−t ) of 6-HKA in the MCAO group was significantly prolonged compared to the sham-operated group, and there was a higher distribution of 6-HKA in the ischemic cerebral cortex than in the contralateral normal cerebral cortex. Conclusion This study illustrated that in MCAO mice, the prolonged MRT 0−t of 6-HKA and the increased permeability of BBB made the accumulation of 6-HKA in the ischemic cerebral cortex, thereby enabling 6-HKA to exert neurorestorative effects.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"20 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147891143","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods Rats were orally administered with “Xinshuixiao granules” containing medicinal compounds. The CCK-8 assay was employed to determine the appropriate drug serum concentration and its effect on IMCD3 cell viability. Additionally, we assessed the influence of “Xinshuixiao” drug-containing serum on IMCD3 cell activity following dDAVP intervention. Cells were divided into blank, model, low, medium, and high-dose “Xinshuixiao” drug serum groups, as well as a Tolvaptan group. After 24 h of drug treatment, ELISA was used to measure the levels of arginine vasopressin (AVP), cyclic adenosine monophosphate (cAMP), and protein kinase A (PKA) in the cell supernatant. Western blotting was employed to evaluate AQP2 protein expression, and real-time polymerase chain reaction (Real-time PCR) was used to assess AQP2 mRNA expression levels. Results In comparison to the blank group, the model group exhibited elevated expression of AVP, cAMP, PKA ( P<0.05), as well as increased AQP2 mRNA and protein expression ( P<0.05). When compared to the model group, all treatment groups displayed varying degrees of reductions in AVP, cAMP, and PKA expression ( P<0.05), along with decreased AQP2 mRNA and protein expression, with statistically significant differences ( P<0.05). Conclusion “Xinshuixiao” drug-containing serum exerts an inhibitory effect on the overexpression of AQP2 induced by dDAVP in IMCD3 cells, which may be associated with its suppression of the cAMP/PKA signaling pathway.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"20 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147915100","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Epigallocatechin 3-gallate (EGCG), the most abundant catechin derived from green tea, has gained widespread attention due to its excellent anti-inflammatory and anti-oxidative properties. This research aims to investigate the effect and mechanism of EGCG in alleviating AD by integrative approach of an in vivo AD mouse model, network pharmacology and molecular docking. Materials and Methods: EGCG was administered by intragastric administration at the dose of 100 mg/kg, once daily for 14 consecutive days to AD mice, a model established by repeated application of 2, 4-dinitrofluorobenzene (DNFB) to the mouse dorsal skin. Results: Macroscopical and histopathological examination showed that oral administration with EGCG could markedly relieve AD-like skin phenotypes, as evidenced by a substantial reduction in severity score, serum immunoglobulin E (IgE) level, epidermal thickness, and the infiltration of inflammatory cells and mast cells into the dermal layers. Mechanistically, the network pharmacology analysis indicated that the protective effects of EGCG against AD were mainly related to the regulation of AGE-RAGE signaling pathway in diabetic complications, Th17 cell differentiation, IL-17 signaling pathway, NF-κB signaling pathway, Toll-like receptor signaling pathway, TNF signaling pathway, etc. In silico molecular docking demonstrated the high binding affinity of EGCG to the core targets such as ABCB1, MMP2, MMP9, MAPK14 and STAT1. Furthermore, western blotting (WB) results showed that EGCG obviously downregulated the expression of RAGE and STAT1, thereby regulating the AGE-RAGE signaling pathway to improve AD. Conclusions: These findings indicated that EGCG can be a promising therapeutic candidate for AD.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"19 12","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-12-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147918406","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods Male C57BL/6J mice were fed either a normal diet or a high-fat diet (HFD). DR1 (60 mg/kg) was administered to both normal and HFD-induced obese mice for 12 weeks. The obese phenotype and pharmacodynamics effects were assessed through hematoxylin-eosin staining of epididymal fat, liver, small intestine, and colon tissues, as well as serum biochemical detection. Moreover, 16S ribosomal ribonucleic acid (16S rRNA) sequencing was employed to profile the taxonomy and function of gut microbiota, while SCFAs were quantified using ultra-performance liquid chromatography coupled with mass spectrometry methods. Results Compared to HFD group, the tissue lesions were significantly alleviated by DR1 treatment in obese mice, accompanied by the significant reduction of average body, epididymal fat and liver weights, as well as the normalization of insulin sensitivity and glucose homeostasis ( P < .05, P < .01 or P < .001). 16S rRNA sequencing analysis showed that DR1 treatment potently ameliorated HFD-induced dysbiosis of gut microbial composition and function. Notably, DR1 reversed the ratio of Firmicutes/ Bacteroidetes to normality. Moreover, DR1 improved intestinal barrier structures and enhanced SCFAs concentrations in the caecal contents (especially propionate and butyrate). Conclusion These results suggested that DR1 could reverse the gut microbial dysbiosis and increased SCFAs levels in HFD-induced obese mice, thereby ultimately supporting the anti-obesity value of DR1.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"19 12","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-12-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147879702","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This study aims to investigate the therapeutic effect and potential mechanisms of YGP in rats with cortisol-induced DOR. Methods The cortisol-induced DOR model was used to investigate the protective effects and mechanism of YGP on DOR. The DOR model was established by intramuscular injection of 25 mg/kg cortisol for 28 consecutive days. Meanwhile, the rats were weighed and administered intragastrically YGP or progynova. To evaluate the mitigating effect of YGP on DOR, pharmacodynamic indicators were assessed including body weight, estrous cycle, H&E staining, and serum levels of AMH, FSH, LH, and cortisol. Oxidative stress (OS) and apoptosis pathways were further validated using Western blotting analysis. Result YGP significantly ameliorated the symptoms of DOR. Administration of YGP improved the ovarian index, estrous cycle, and body weight. After treatment with YGP, cortisol, FSH, and LH levels declined and AMH levels elevated. YGP increased the number of primary, secondary, and antral follicles, but decreased the number of atretic follicles. Mechanically, YGPtreated cortisol-induced DOR via the Keap1/Nrf2 signaling pathway which regulates OS. YGP also reversed the aberrant expression of apoptosis-related proteins Caspase-3, Bcl-2, and Bax. Conclusion YGP could reduce ovarian OS and apoptosis in cortisol-induced rats via regulating the Keap1/Nrf2 signaling pathway, thereby enhancing ovarian reserve function.","PeriodicalId":19019,"journal":{"name":"Natural Product Communications","volume":"19 11","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-11-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147879007","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
