Methods Ultrasonic-assisted extraction was utilized to isolate polyphenolic compounds from Pu’er tea flowers, and the extraction process was systematically optimized using response surface methodology (RSM). Results The optimal conditions were determined as follows: ethanol concentration of 55%, solid-liquid ratio of 1:45 (g/mL), extraction temperature of 55 °C, extraction time of 90 min, and ultrasonic power of 480 W. Under these optimal conditions, the total polyphenol yield reached 4.95±0.08 mg/g. The extracted polyphenols exhibited notable in vitro bioactivities. They demonstrated strong antioxidant capacity, with IC 50 values of 0.068 mg/mL for DPPH• scavenging and 0.093 mg/mL for ABTS• + scavenging. In addition, the polyphenols showed significant hypoglycemic potential via inhibition of α-glucosidase, with an IC 50 value of 0.73 mg/mL. Moreover, their hypolipidemic effect was evidenced by bile salt binding capacity, with IC 50 values of 2.49 mg/mL for sodium taurocholate and 4.40 mg/mL for sodium glycocholate. Conclusion In summary, polyphenols derived from Pu’er tea flowers possess remarkable antioxidant, hypoglycemic, and hypolipidemic properties. (Gouqi, GQ) is a prominent component of traditional Chinese medicine with well-established medicine-food homology properties. Owing to its high nutritional profile, GQ is widely recognized for its diverse pharmacological benefits, including protective effects on the kidney. The primary objective of this study was to evaluate the anti-urolithiatic activity of GQ aqueous extract and to elucidate its underlying therapeutic mechanisms in the context of calcium oxalate (CaOx) nephrolithiasis (Kidney stone). Methods The anti-kidney stone activity of the GQ extract was assessed in a Drosophila melanogaster (fruit fly) model of CaOx kidney stones induced by dietary sodium oxalate (NaOx). Oxidative stress levels were quantified via dihydroethidium (DHE) staining. The expression profiles of key enzymes involved in endogenous oxalate synthesis—namely, agxt (the ortholog of human alanine-glyoxylate transaminase, AGT) and daao1/daao2 (encoding two D-amino acid oxidases, DAOs)—were analyzed using RT-qPCR technology. Excretory function was evaluated by counting fly fecal deposits. In vitro crystallization assays were performed to examine evolution of CaOx crystal morphology and explore the regulatory activity of the GQ extract during crystal formation. Results Our results demonstrated that the GQ extract effectively attenuated CaOx kidney stone formation through multiple interconnected mechanisms. Specifically, GQ extract mitigated NaOx-induced oxidative stress, improved urinary excretion, reduced endogenous oxalate biosynthesis, and directly altered the morphological dynamics of CaOx crystallization. Conclusions These findings substantiate the significant stone-preventive potential of GQ, whose therapeutic effects are mediated via a pleiotropic mode of action. Methods Ultra-high-performance liquid chromatography coupled with quadrupole electrostatic field orbitrap tandem high-resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS) was used to identify the components of ND in the blood. Potential target sites of these chemical components were identified using the SwissADME database, while disease-related targets for depression were selected from the GeneCards database. Intersecting targets between the two databases were then identified. Cytoscape software was used to construct a “component-target-disease” network to screen the core components of ND relevant to the treatment of depression. The STRING platform was used to construct a protein-protein interaction (PPI) network of the intersecting targets to identify the core targets of ND for treating depression. Gene ontology (GO) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were performed using the DAVID platform. Schrödinger software was used to determine molecular docking of the core components and core target proteins. Results Seventy-six blood components associated with 747 drug targets were identified,. Of the 2,000 depression-related targets, 212 overlapped with those of ND. A total of 16 core targets and 16 core components were identified. The GO function and KEGG pathway enrichment analyses indicated that treatment of depression with ND ’ s may be related closely to the mitogen-activated protein kinase (MAPK) signaling and Prion disease signaling pathways. The molecular docking results showed that piperine had the highest binding energy with tumor necrosis factor (TNF). Conclusion The core components of ND such as piperine, liquiritigenin, and piperanine may act on targets including TNF, mitogen-activated protein kinase 3(MAPK3), estrogen receptor 1 (ESR1, and interleukin-6(IL-6), resulting in regulation of signaling pathways such as MAPK, TNF, and Prion disease involved in the treatment of depression. Method The physical properties of 10 batches of raw, blanched, and fried ASA, including moisture content, hardness, relative density, and extract yield, were evaluated. A validated HPLC method was used to determine the contents of amygdalin and prunasin in water decoctions prepared from whole seeds and from five crushed particle size fractions (&lt; 0.85, 0.85–2, 2–4, 4–5, and 5–6 mm). Accelerated stability testing was conducted according to Chinese Pharmacopoeia guidelines at 40 °C ± 2 °C and 75% ± 5% relative humidity for 30 days. Paper bags, polyethylene bags, and aluminum foil bags were evaluated under both vacuum and non-vacuum conditions. Key stability indicators—moisture content, fatty oil content, acid value, and amygdalin content—were monitored throughout the storage period. Result Processing significantly altered the physical properties of ASA ( P ≤ 0.001). The contents of amygdalin and prunasin in water decoctions prepared from crushed samples of all three processed ASA products were significantly higher than those from whole seeds ( P &lt; 0.001). The recommended optimal particle size ranges were 4–5 mm for raw ASA, 2–4 mm for blanched ASA, and 0.85–2 mm for fried ASA. In the stability study, samples packaged in paper bags showed marked deterioration: after 30 days, moisture content increased to 9.03%, the acid value rose to 2.84, and amygdalin content decreased by 15.41%. In contrast, polyethylene and aluminum foil maintained all stability indicators within acceptable ranges, with no significant benefit from vacuum treatment. Conclusion Processing altered the physical properties of ASA, and crushing enhanced the dissolution of its active components. Although 28 Traditional Chinese Medicine (TCM) prescriptions are approved for BPH, their key quality components and mechanisms remain unclear. Methods: Network pharmacology and molecular docking analyzed 28 prescriptions to identify components. High-performance liquid chromatography (HPLC) quantified these components. Optimized component ratios were validated in BPH cell lines, assessing hormone (PSA, DHT, E2) and inflammatory biomarkers (NO, NOS). Results: Analysis identified 75 active compounds. Twenty-six prescriptions shared a mechanism targeting proteins (eg, TP53, AKT1). Quercetin, luteolin, kaempferol, and β-sitosterol showed stable binding to targets in 17 prescriptions; their contents were quantified across 26 prescriptions. An optimized combination (quercetin: 81 µM, luteolin: 46 µM, kaempferol: 129 µM, β-sitosterol: 82 µM; molar ratio 1:0.6:1.6:1 relative to β-sitosterol) significantly inhibited BPH cell growth by 89.6%. It reduced PSA, DHT, E2 and increased NO, NOS levels. Conclusion: The quercetin, luteolin, kaempferol, and β-sitosterol are key quality components for BPH treatment. Incorporating them at the experimentally determined ratio within safe limits significantly enhances efficacy. Consequently, these findings provide practical insights for refining TCM quality standards, identifying key quality markers, and facilitating the development of new BPH-specific TCM treatments. Methods Public data from The Human Protein Atlas (HPA) were analyzed for DAAM1 expression. Molecular docking (PyMOL) and microscale thermophoresis (MST) were used to confirm the direct binding of IFY to the FH2 domain of DAAM1 in vitro . Cell Counting Kit-8 (CCK8) assay, wound healing assay, Boyden chamber assay, Western blotting and microfilament assembly (immunofluorescence) were assessed in OS and BC cell lines. Antitumor efficacy was further evaluated in a 4T1 mouse breast cancer xenograft model. Results DAAM1 was highly expressed in OS and BC tissues and cells. IFY directly bound the FH2 domain of DAAM1. IFY (10-15 μmol/L) significantly reduced cell viability, migration, invasion, microfilament assembly, and DAAM1 protein expression levels in OS and BC cell lines. In vivo , IFY (10 mg/kg, i.p., q.2d) markedly suppressed tumor growth in BC-bearing mice. Chrysanthemum morifolium extract (CME), a traditional herb with vision-protective properties, was evaluated for its potential to mitigate this damage. Methods Human conjunctival epithelial cells (HCoECs) were exposed to UV to model solar injury. CME was characterized and its effects were assessed through cell viability (CCK-8), lipid peroxidation (MDA assay), ROS production, and mRNA sequencing. Bioinformatic tools (eg, Swiss Target Prediction, STRING) identified flavonoid targets and ferroptosis-related networks. Results UV exposure triggered ferroptosis in HCoECs, evidenced by iron overload (TFRC1 upregulation), elevated ROS/MDA, and suppressed cell viability. RNA-seq revealed activation of ferroptosis pathways (eg, TFRC1, ACSL4) and inflammation signaling (eg, IL-17/HIF-1). CME pretreatment (100 μg/mL) significantly attenuated these effects, lowering MDA levels and restoring cell viability. Mechanistically, CME inhibited TFRC1-mediated iron influx and disrupted ferroptosis effector networks. Conclusion Our findings demonstrate that CME could protect HCoECs from UV-induced ferroptosis by chelating iron and suppressing lipid peroxidation. This study aims to systematically identify the key bioactive compounds of BJOE and decipher their anti-BC mechanisms through an integrative network pharmacology approach combined with experimental validation. Methods Active components and potential targets of Yadanzi were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database (TCMSP). BC-related genes were collected from public databases. A compound-target network was constructed using Cytoscape. Furthermore, protein-protein interaction (PPI) network construction, along with Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses, were performed to identify hub targets and signaling pathways. Finally, molecular docking, in vitro and in vivo experiments were performed to verify the network pharmacology results. Results Fifteen active components of Yadanzi were screened from the TCMSP database. After excluding compounds without target information, two key bioactive compounds (beta-sitosterol and luteolin) and 73 potential targets were retained, among which 71 were associated with BC. Network analysis revealed AKT1, CASP3, and TP53 as hub targets. Enrichment analysis indicated these genes are primarily involved in apoptosis regulation, PI3K-Akt signaling, and other cancer-related pathways. Molecular docking demonstrated strong binding affinity between luteolin and AKT1/CASP3/TP53, as well as between beta-sitosterol and CASP3, with binding energies ranging from −6.4 to −7.8 kcal/mol. In vitro experiments verified that luteolin inhibits the proliferation and migration of breast cancer cells and induces apoptosis, reducing viability by up to 50% ( P &lt; .001) and increasing apoptotic rates by approximately 2-fold compared to controls ( P &lt; .001). Western blot analysis indicated that luteolin regulate the expression levels of proteins associated with apoptosis. Xenograft mouse model verified that luteolin could delay tumor growth. Conclusion This study provides a novel network pharmacology-based framework for elucidating the anti-BC mechanisms of BJOE. It was suggested that the key active component luteolin can inhibit BC cell proliferation, induce apoptosis and regulate the expression of related proteins associated with apoptosis. Methods Using response surface methodology (RSM), the effects of ethanol volume fraction, solid-to-solvent ratio, and ultrasonic power on polyphenol concentration were investigated. The impact of temperature, pH, and feed concentration on the adsorption rate was examined and optimized using an orthogonal experimental design. The phenolic compounds in the extract were qualitatively profiled using liquid chromatography–mass spectrometry (LC–MS). Finally, the antioxidant activity was evaluated by determining the DPPH radical scavenging rate. Results The optimal extraction conditions were as follows: an ethanol volume fraction of 70% (v/v), a solid-to-solvent ratio of 1:13 g/mL, and an ultrasonic power of 150 W. Under these conditions, the polyphenol concentration reached 0.769 milligram gallic acid equivalents per milliliter. The optimal adsorption conditions for the macroporous resin were 40 °C, pH 4.0, and a feed concentration of 1.54 mg/mL, which provided an adsorption rate of 12.06%. A total of 43 phenolic compounds were detected in the extract using LC–MS, among which rutin exhibited a relatively high abundance. At a concentration of 20 mg/mL, the DPPH radical scavenging rate of the extract was 42.47%. Consequently, it is essential to identify the key factors influencing the quality of Lonicera japonica to enhance its cultivation standards. This study compares the chlorogenic acid content of Lonicera japonica from various production areas, explores the climate and soil factors that influence chlorogenic acid accumulation, and provides a basis for optimizing artificial planting methods and quality control of Lonicera japonica . Methods We collected Lonicera japonica samples from three origins: Henan, Hebei, and Shandong, determined the chlorogenic acid content using high-performance liquid chromatography (HPLC), and analyzed the variations in samples from different origins. We tested the soil physicochemical properties according to national standards, detected the soil microbial community composition using 16S amplicon sequencing technology, and analyzed the climatic and soil factors affecting chlorogenic acid accumulation. Results Chlorogenic acid content was highest in the Hebei Lonicera japonica samples. The annual mean temperature, precipitation, relative humidity, and wind speed negatively correlate with chlorogenic acid content. In contrast, the annual sunshine duration positively correlates with chlorogenic acid content. Rhizosphere samples from Shandong have higher total nitrogen, sulfur, and carbon content than those from Henan and Hebei. Furthermore, Henan soils exhibit slightly higher nitrate nitrogen content and slightly lower nitrite content, while Hebei soils have higher ammonium nitrogen concentration. The weakly acidic pH value was positively correlated with chlorogenic acid content. The rhizosphere soil of Lonicera japonica in Shandong exhibited the highest bacterial abundance, and the rhizosphere soil of Lonicera japonica from Henan yielded the highest number of ASVs. Strains 27F-1492R, KF-JG30-B3, Pla4_lineage, Bacillus , and bacteriap25 were highly abundant in rhizosphere soil samples from Henan. Ohtaekwangia , Caulobacter , and Larkinella were abundant in the Shandong rhizosphere samples. The abundances of Subgroup_10, Bryobacter , and Uncultured were negatively correlated with the chlorogenic acid content, whereas Terrimonas showed positive correlations. Conclusion Lonicera japonica from the Hebei Province had the highest chlorogenic acid content. Environmental factors affect the quality of Lonicera japonica.
