Ekosistem padang lamun diketahui memiliki kemampun dalam menyerap karbondioksida dan menyimpannya dalam waktu yang lama. Telah dilakukan penelitian untuk mengetahui keanekaragaman jenis lamun, presentasi tutupan dan kandungan karbon pada padang lamun di perairan Pulau Manado Tua. Untuk mengetahui keanekaragaman jenis dilakukan pengamatan secara langsung pada setiap stasiun penelitian dan untuk mengetahui presentase tutupan lamun menggunakan metode line transek kuadran dengan mengadopsi Seagrass Watch dimana sampel lamun juga diperoleh dari line transek kuadran pada keseluruhan stasiun kemudian dianalisis selanjutnya menggunakan metode Loss on ignition (LOI). The purpose of this study was to obtain symbiotic fungi isolates, ethyl acetate extracts from ascidian symbiotic isolates, and to determine the antibacterial and antifungal activities of ethyl acetate extracts of symbiotic isolates using the diffusion method (disc diffusion Kirby and Bauer) against Escherichia coli, Staphylococcus aureus bacteria and against the Candida albicans fungus. The results obtained 1 ascidian species which was cultured to obtain pure isolates, the pure isolates were tested molecularly and the results showed that the isolate was Aspergillus flavus, then the isolates were grown on rice medium and extracted. The results of the fungal extract showed the presence of antibacterial and antifungal activity against Escherichia coli by 10 mm, Staphylococcus aureus by 19 mm and against the Candida albicans by 13.5 mm. Keywords : Ascidian, Eudistoma sp., Aspergillus flavus, Antibacterial, Antifungal, Symbiotic.                                                  AbstrakAscidian merupakan avertebrata laut yang dapat menghasilkan senyawa bioaktif yang seperti antibakteri dan antijamur. Tujuan dari penelitian ini yaitu mendapatkan isolat jamur simbion dari ascidian, mendapatkan ekstrak etil asetat dari isolat jamur simbion ascidian, dan menguji aktivitas antibakteri dan antijamur ekstrak etil asetat isolat jamur simbion dengan metode difusi agar (disc diffusion Kirby and Bauer) terhadap bakteri Escherichia coli, Staphylococcus aureus dan terhadap jamur Candida albicsans. Hasil penelitian didapatkan 1 jenis ascidian yang kemudian dikultur untuk mendapatkan isolat murni, isolat murni diuji secara molekuler dan didapati hasil bahwa isolat tersebut merupakan jamur Aspergillus flavus. Isolat tersebut kemudian ditumbuhkan pada media nasi dan diekstrak. Hasil ekstrak jamur tersebut menunjukkan adanya aktivitas antibakteri dan antijamur terhadap bakteri Escherichia coli sebesar 10 mm, Staphylococcus aureus sebesar 19 mm dan terhadap jamur Candida albicsans 13,5 mm. This organism is able to synthesize secondary metabolites from food. The purpose of this study was to obtain P. varicosa extract and symbiotic bacterial extract from P. varicosa, then determine the antibacterial activity of P. varicosa extract and the symbionic bacteria extract against Escherichia coli DSM498, Bacillus megaterium DSM32T and anti-UV activity. P. varicose symbiotic bacterial were isolated and extracted. The results obtained 5 bacterial isolates. The results of antibacterial assay of isolates PhVa 1.1, PhVa 1.3, PhVa 2.1, PhVa 2.3 and PhVa 2.4 shown that these isolates have an antibacterial activity against E. coli DSM498 and B. megaterium DSM32T. Anti-UV assay results shown an absorption at UV-A with the highest value of 1.991 at λ 340 nm. Organisme ini mampu mensintesis metabolit sekunder dari bahan makanannya. Tujuan dari penelitian ini yaitu untuk mendapatkan ekstrak P. varicosa dan ekstrak bakteri simbion dari P. varicosa, kemudian mengamati aktivitas antibakteri dari ekstrak P. varicosa dan ekstrak bakteri simbionnya terhadap bakteri Escherichia coli DSM498 dan Bacillus megaterium DSM32T serta menguji aktivitas anti-UV. Bakteri yang bersimbion dengan P. varicosa diisolasi dan diekstraksi, lalu diuji bioaktivitas antibakteri dan diuji anti-UV terhadap ekstrak P. varicosa dan ekstrak bakteri simbionnya. Hasil akhir dari penelitian ini yaitu didapatkan 5 isolat bakteri. Hasil uji aktivitas antibakteri yaitu isolat PhVa 1.1, PhVa 1.3, PhVa 2.1, PhVa 2.3 dan PhVa 2.4 memiliki aktivitas antibakteri terhadap bakteri E. coli DSM498 dan B. megaterium DSM32T. Marine bacteria contain chemicals compunds that have potentials on marine drugs discovery through the secondary metabolism. The purpose of this study was to isolate the symbiotic bacteria from nudibranches Phylidiella pustulosa and Thuridilla lineolate and to screen the antibacterial activity of these bacterial isolates against Escherichia coli and Staphylococcus aureus. Isolation and culture of the symbiotic bacteria was made on B1 (solid) media. The results of the antibacterial assay showed that the symbiotic bacterial isolates from Phylidiella pustulosa and Thuridilla lineolate were able to inhibit the growth of the Staphylococcus aureus and Escherichia coli test. Tujuan dari penelitian ini yaitu untuk mengisolasi bakteri yang bersimbion dengan nudibranch Phylidiella pustulosa dan Thuridilla lineolate, kemudian mengamati aktivitas antibakteri dari isolat bakteri tersebut terhadap Escherichia coli danStaphylococcus aureus. Isolasi dan kultur bakteri yang bersimbion dibuat pada media B1 (padat). Wantasen","doi":"10.35800/jplt.8.2.2020.29951","DOIUrl":"https://doi.org/10.35800/jplt.8.2.2020.29951","url":null,"abstract":"Mangrove is one of plants that can grow well on the coast which are affected by tides.Bone Baru village is located in North Banggai District, Banggai Laut Regency, Central Sulawesi Province which has area that about 840 ha and has mangrove area reaching 16.56 ha. This research was conducted from February to April 2020. The purpose of this research are to identify the types of mangroves and analyzing the structure of the mangrove community which include density, relative density, frequency, relative frequency, coverarge, relative coverage, importance value index, and diversity index. The data were analyzed using Microsoft Excel. Based on the results this study obtained 4 types of mangroves consisting of Bruguiera gymnorrhiza, B. cylindrica, Rhizophora mucronata, and Lumnitzera littorea. The highest relative density was 0.13 ind / m2 and the relative density was 91.83% the highest frequency type and the frequency relative value were 5 ind / m2 and 52.63%respectively. The highest mangrove cover was 3.79 m2 with relative cover of 52.21%. The highest important value index was 227.72% . The highest diversity index was found at station 2 with an average value (H ') of 0.69.Keywords: mangrove, community structure, habitat, BanggaiABSTRAKMangrove merupakan kelompok tumbuhan yang dapat tumbuh dengan baik di pesisir pantai yang dipengaruhi oleh pasang surut air laut. Penelitian ini dilakukan di desa Bone Baru, Kecamatan Banggai Utara, Kabupaten Banggai Laut, Provinsi Sulawesi Tengah yang memiliki luas wilayah mencapai 840 ha dan memiliki luas area mangrove mencapai 16,56 ha. Penelitian ini dilakukan dari bulan Februari-April 2020 dengan menggunakan metode Line Transect. Tujuan dari penelitian ini adalah mengidentifikasi jenis-jenis mangrove dan menganalisa struktur komunitas mangrove Data yang diambil meliputi kerapatan jenis, kerapatan relatif jenis, frekuensi jenis, frekuensi relatif jenis, penutupan jenis, penutupan relatif jenis, indeks nilai penting, dan indeks keanekaragaman yang kemudian di analisa dengan bantuan program komputer Microsoft Excel. Berdasarkan hasil penelitian ditemukan 4 jenis mangrove yaitu Bruguiera gymnorrhiza, Rhizophora mucronata, Lumnitzera littorea dan Bruguiera cylindrica. Kerapatan jenis tertinggi 0,13 ind/m2 dan kerapatan relatifnya 91,83%. This study took samples of coral mucus in 2010 at 3 (three) different locations, namely Bunaken (May); Morotai (September) and Raja Ampat (November), which focused on the analysis of Research on hard coral (Scleractinian coral) contaminated with bacteria is still not much done, especially in Indonesian waters. This study took samples of coral mucus in 2010 at 3 (three) different locations, namely Bunaken (May); Morotai (September) and Raja Ampat (November), which focused on the analysis of gram-positive and gram-negative bacteria. The method used for field sampling is time swim, which is by diving at a depth of 5-10 meters for ± 30 minutes and randomly taking samples of coral mucus using siring or by taking directly on corals (reef branching). Mucus samples were analyzed by bacterial isolation in the laboratory. The result shows that there were differences between gram-positive and gram-negative bacteria in the three research sites and that gram-positive bacteria were higher or dominant. Further research that can identify the bacteria species and explain its relationship to the ecosystem is highly recommended.Keywords: Bacteria, Scleractinian coral, gram-positive and -negative, Bunaken, Morotai, Raja Ampat                                               AbstrakPenelitian tentang karang keras (Scleractinian coral) yang terkontaminasi bakteri masih belum banyak dilakukan, terutama di perairan Indonesia. Penelitian ini mengambil sampel mucus karang pada tahun 2010 di 3 (tiga) lokasi berbeda, yakni Bunaken (Mei); Morotai (September) dan Raja Ampat (November), yang difokuskan pada analisis bakteri gram postif dan gram negatif. Metode yang digunakan untuk pengambilan sampel di lapangan adalah time swim, yaitu dengan penyelaman pada kedalaman 5-10 meter selama ±30 menit dan mengambil sampel mucus karang secara acak menggunakan siring atau dengan mengambil langsung pada karang (fraksi cabang). Sampel mucus dianalisis dengan cara isolasi bakteri di laboratorium. Hasil analisis menunjukkan bahwa ada perbedaan antara bakteri gram positif dan gram negative di tiga lokasi survei dan bakteri gram positif lebih tinggi atau dominan. The main problems recorded, namely: 1) lack of knowledge of marine fish aquaculture technology in floating net cage; 2) fish harvest time that requires a long time, and 3) poor financial management.  To overcome these problems, an approach is carried out through direct education and training, mentoring, and management of fisheries business management in the field to partners.  After that, each trainee in the partner group is given the opportunity until they can do it themselves.  The target that will be achieved at the end of this education and training is that all group members and their families have knowledge of: 1) superior marine fish polyculture technology in floating net cage and 2) good financial management. Keywords: Fish cultivator group, floating net cage, South Motandoi village, financial management, marine fish polyculture technologyAbstrakTujuan program kemitraan masyarakat (PKM) adalah meningkatkan kapasitas kelompok pembudidaya ikan laut yang mandiri dan berkelanjutan. Target khusus adalah 1) peningkatan kuantitas dan kualitas produk ikan hasil budidaya laut dalam karamba jaring apung, dan 2) peningkatan pemahaman dan ketrampilan mitra dalam hal teknologi budidaya ikan laut dalam karamba jaring apung yang efektif dan efisien. Permasalahan utama yang terekam, yaitu: 1) kurang pengetahuan tentang teknologi budidaya ikan laut dalam karamba jaring apung (KJA); 2) waktu panen ikan yang membutuhkan waktu yang lama, serta 3) pengelolaan keuangan yang kurang baik. Untuk mengatasi masalah tersebut dilakukan pendekatan melalui penyuluhan dan pelatihan, pendampingan, dan pendidikan manajemen usaha perikanan secara langsung di lapangan kepada mitra. Mangrove forests with their uniqueness are natural resources that have the potential to be used as tourist attractions. The purpose of this study was to determine community structure, mangrove canopy cover and suitability of mangrove ecotourism land. This study used the line transect method for mangrove community structure, and hemispherical photography for the percentage of mangrove cover. The results of the study of the mangrove community structure were the highest density value, namely at station 1 on the transect 2 types of S.alba 0.16 ind / m2 relative 88.89% and the lowest density value in the type R. mucronata and R.apiculata 0.01 ind / m2 relatively 5.56% on transect 2 at station 1. The highest frequency type value at station 1 on the transect 1 type S. alba which is 1.00 relative 100% and the lowest frequency at station 1 transect 2 types R, piculate which is relative 0.50 33.33. The highest closing value of type on transect 2 standard 1 type S. alba 10.26 relative value 92.63%. Whereas the lowest type 3 transect closure is type R. piculate 0.49 the relative is 7.86%. The highest Important Value Index is at station 1 on transect 1, namely type S. alba 300 and lowest at station 1 on transect 2 types R. mucronata 30.55. The highest diversity value on transect 2, 0.43. Evenness index value (E) at station 1 on transect 1 is 0 and transect 2 is 0.39. While station 2 on transect 1 0 and transect 2 0.51. regarding the criteria for mangrove damage, at 2 stations included in the rare category with inya 50% canopy cover. Echinoderms consist of approximately 6000 species and all live in sea water. Starfish is one source bioactive compounds. Active compounds from starfish have been known to have activities antioxidant, antibacterial, anti-inflammatory, antifungal and immunostimulatory, there are also blue starfish that are potential as antitumor and antibacterial agents. This study aims to obtain crude extracts from several types of starfish, and testing the antibacterial activity of crude starfish extracts against bacteria Streptococcus agalactiae, Escherichia coli and  Staphylococcus aureus. Test the antibacterial activity of crude starfish extracs Protoreaster nodosus, Achaster tipycus, dan Linckia laevigata against bacteria E. coli, S. aureus, dan S. agalactiae use the agar diffusion method by means of a well.  L. l extract  can inhibit bacteria e. coli at an extract concentration of 1000 mg/ml with a diameter of inhibitory zone 11.0 mm, whereas at a concentration of 750 mg/ml the test bacteria can still grow on the bacteria S. agalactiae and S. aureus can still grow even though the extract has be given  L. l Keywords: Potential Starfish, Antibacterial Activity, Staphylococcus aureus, Escherichia coli, and Streptococcus agalactiae Bintang laut merupakan salah satu spesies dari kelas Asteroidea, dan dikelompokkan kedalam filum Echinodermata. Filum Echinodermata  terdiri atas kurang lebih 6000 spesies dan semuanya hidup di air laut. Bintang laut  merupakan salah satu sumber penghasil senyawa bioaktif. Senyawa aktif dari bintang laut telah diketahui memiliki aktivitas antioksidan, antibakteri, antiinflamasi, antifungi dan imunostimulator, ada juga bintang laut biru yang potensial sebagai antitumor dan agen antibakteri. Penelitian ini bertujuan untuk mendapatkan ekstrak kasar dari beberapa jenis bintang laut, dan menguji aktivitas antibakteri ekstrak kasar bintang laut terhadap bakteri Streptococcus agalactiae, Escherichia coli dan  Staphylococcus aureus. Uji aktivitas antibakteri dari ekstrak kasar bintang laut Protoreaster nodosus, Achaster tipycus, dan Linckia laevigata terhadap bakteri E. coli, S. aureus, dan S. agalactiae mengggunakan metode difusi agar dengan cara sumur. The purpose of this study is to determine the antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria and to test the anti-UV activity of the ascidian water fraction. Antibacterial activity test was carried out using the diffusion method (disc diffusion Kirby & Bauer) and the water fraction of ascidian that showing antibacterial activity tested in a UV spectrophotometer to see the anti-UV activity. Results of the study were obtained 4 types of ascidians extracted from partitioned into water fraction, n-hexane fraction, methanol fraction. All three fractions were tested for antibacterial activity and the result showed that there were antibacterial activities of ascidian extract Clavelina sp. against both test bacteria with inhibition of S. aureus and E. coli with strong categories. Ascidian Phlebobranch sp. showed the presence of antibacterial activity with inhibition of S. aureus and E. coli with very strong categories (16,6 mm). extract Eudistoma sp. showed the presence of antibacterial activity with inhibition S. aureus and E. coli in the medium category (9 mm).
