Neurokinin B (NKB) plays a crucial role in gonadotropin secretion, which is essential for reproduction. The impact of menopause and obesity on MT2 and NKB concentrations remains an enigma. Therefore, the present study sought to investigate how obesity and menopause affect serum concentrations of MT2 and NKB.</p><p><strong>Methods: </strong>In this cross-sectional study, 120 female volunteers were divided into four groups: menopausal overweight, menopausal obese, menopausal normal weight (control), and reproductive normal weight (control). The participants' serum levels of MT2, NKB, iron, ferritin, and relevant biochemical parameters were analysed using the ELISA method.</p><p><strong>Results: </strong>The mean serum concentrations of NKB, MT2, and ferritin were higher in the menopausal overweight and obese groups (p<0.05). The mean concentrations of NKB, MT2, and estradiol were determined to be higher, and the levels of iron, hemoglobin, glucose, FSH, LH, LDL, and total cholesterol were found to be lower in the reproductive control group when compared with the menopausal control group (p<0.05). A positive correlation was identified between BMI and NKB, MT2, and ferritin concentrations in menopausal overweight and obese groups (p<0.05).</p><p><strong>Conclusions: </strong>Obesity elevated NKB and MT2 concentrations in menopausal women. The increase in MT2 levels in menopausal overweight or obese women may be one of the factors responsible for the increase in ferritin and body fat ratio. As BMI raised, NKB, MT2, and ferritin concentrations raised in the menopausal overweight or obese groups; therefore, depression of MT2 in obese patients could be beneficial for therapeutic purposes. Specific gene variations, such as Factor V (FV) G1691A (Leiden), FV H1299R, and Prothrombin (FII) G20210A, have been implicated in thrombotic events and CAD. However, their precise role in CAD development remains controversial. This study investigated the prevalence and association of these gene variations with thrombosis and CAD in the Turkish population.</p><p><strong>Methods: </strong>A case-control study included 406 healthy individuals and 64 CAD patients. Genotyping for FV G1691A, FV H1299R, and FII G20210A was performed using a strip assay. Fisher's exact test compared allele and genotype frequencies between the CAD and control groups.</p><p><strong>Results: </strong>No significant differences were observed in genotype frequencies of FV G1691A, FV H1299R, and FII G20210A between the CAD and control groups (p>0.05). Similarly, allele frequencies did not differ significantly between the two groups (p>0.05).</p><p><strong>Conclusions: </strong>The findings suggest that FV G1691A, FV H1299R, and FII G20210A variations may not play a significant role in the development of CAD in the Turkish population studied. These results are consistent with the existing conflicting literature on the association between these gene variations and CAD. How to quickly and accurately evaluate the progression of AP has always been a hotspot and difficulty in clinical research.</p><p><strong>Methods: </strong>Sixty-four AP patients were admitted to our hospital between August 2022 and June 2023, and 60 healthy people during the same period were selected for retrospective analysis, with AP patients as the observation group and healthy people as the control group. HMGB-1 and AChE levels were detected in both groups, and the diagnostic value of HMGB-1 and AChE for AP was analysed using the receiver operating characteristic (ROC) curve. Subsequently, the differences in the expression of HMGB-1 and AChE in AP patients with different severities were further observed. In addition, we detected albumin (ALB), transferrin (TRF), and total protein (TP) in the observation group and analysed their correlation with HMGB-1 and AChE. Finally, a 6-month prognostic follow-up was performed to analyse the predictive value of HMGB-1 and AChE for poor prognosis in AP using ROC curves.</p><p><strong>Results: </strong>Compared with the control group, HMGB-1 was higher in the observation group, which was positively correlated with the severity of AP (P<0.05), while AChE was lower, which was negatively correlated with the severity of AP (P<0.05). HMGB-1+AChE had a sensitivity of 48.44% and a specificity of 88.33% for diagnosing AP (P<0.05, cut-off>0.639). In addition, HMGB-1 and nutrient proteins were positively correlated, and AChE and nutrient proteins were negatively correlated in the observation group (P<0.05). Magnesium is a bioessential element with multiple physiological effects relevant for vital body functions. It stabilizes excitable membranes contributing to proper neuromuscular, cardiovascular and respiratory functions. Ionized Mg2+ (iMg2+) is free bioactive form of Mg2+ in body fluids. Magnesium disorders in critically ill patients are often overlooked. Chronic Mg2+ deficiency is a condition of growing incidence in the general population and a significant factor in overall morbidity and mortality in critical illness. Main goal of this study was to investigate the existing literature related to determination of iMg2+ in the critically ill and to raise awareness of the problem of chronic Mg2+ deficiency in these patients.</p><p><strong>Methods: </strong>Search was conducted across electronic PubMed library database from 1975 to November 2024 using keywords concerning iMg2+ and critical care patients, to identify studies investigating the measuring of blood concentration of iMg2+ fraction in patients with critical health conditions. Search was limited to English language. Selection criteria included only studies on human, and excluded studies on animal population.</p><p><strong>Results: </strong>We identified 95 relevant studies. Reviewed papers show that abnormalities of Mg2+ levels are prevalent in critical illnesses. Their severity can impose vital threat. {"title":"A comparison of the optical method with the mechanical method in routine coagulation tests.","authors":"Gözde Ülfer","doi":"10.5937/jomb0-56100","DOIUrl":"10.5937/jomb0-56100","url":null,"abstract":"<p><strong>Background: </strong>This study aimed to compare the prothrombin time (PT), international normalised ratio (INR) and activated partial thromboplastin time (aPTT) values obtained using the photo-optical method and to assess these values according to the reference method, which was the mechanical method.</p><p><strong>Methods: </strong>Plasma samples from 340 patients, submitted to our hospital's biochemistry laboratory for PT, INR, and aPTT analyses, were assayed using the mechanical coagulometric measurement method in a Stago Compact Max3 automated coagulation analyser, which served as the reference device. The same samples were also analysed using the Sunbio UP5500 automated analyser with a simultaneous optical method. There were 30 turbid samples analysed in both devices without exclusion from the study. Correlation coefficient analysis was carried out using SPSS to assess intervariable correlations. Passing-Bablok regression analysis was performed in R software version 3.6.0 to compare PT, INR, and aPTT values between the two devices. Bland-Altman plots were used to analyse the agreement.</p><p><strong>Results: </strong>A good level of statistically significant agreement was found between the PT and INR values measured by the Stago Compact Max3 and Sunbio UP 5500 devices (Interclass Coefficient Correlation (ICC): 0.627, p=0.001; p<0.01 and ICC: 0.653, p=0.001; p<0.01, respectively). Additionally, there was an excellent level of statistically significant agreement for the aPTT values (ICC: 0.902, p=0.001, p<0.01). The Bland-Altman analysis revealed the mean 95% limits of agreement values as 2.46 (lower limit: -2.44, upper limit: 7.37) for PT, 0.07 (lower limit: -0.32, upper limit: 0.46) for INR, and 2.45 (lower limit: -1.67, upper limit: 6.58) for aPTT. The Passing-Bablok regression results indicated a systematic difference for PT measurement but no proportional difference. No systematic or proportional differences were found for the measured INR and aPTT values between the Stago Compact Max3 and Sunbio UP 5500 devices. The intra-assay and interassay coefficient of variation (CV) values from level 1 and 2 controls of the optical method were below 5%.</p><p><strong>Conclusions: </strong>The results from the optical method were consistent and reliable compared to the mechanical method. PT and INR results showed statistically good agreement, while aPTT results demonstrated excellent agreement. Among these cancers, colon cancer is the second leading cause of cancer-related deaths globally. This study aimed to detect malignant cells early by analyzing biochemical indicators associated with the transformation of digestive cancer cells. Tumour markers, primarily proteins associated with malignant cells, may play a role in preventing the spread and progression of the disease.</p><p><strong>Methods: </strong>In this study, the tumour markers analyzed included carcinoembryonic antigen (CEA), a-enolase (ENO1), a-fetoprotein (AFP), and calprotectin (CP) to detect digestive system cancer. These markers were measured in serum samples from sixty patients with digestive system cancer (the patient group) and sixty healthy individuals (the control group).</p><p><strong>Results: </strong>The results revealed highly significant changes (p-value <0.00001) in the levels of CEA (56.698±44.558 ng/mL), ENO1 (51.784±10.395 ng/mL), AFP (116.275±38.956 ng/mL), and CP (287.425±33.181 ng/mL) in the cancerous group compared to the control group, suggesting their potential utility in early diagnosis. We assessed the impact of sarcoidosis symptoms, pulmonary function, fatigue, radiographic findings, comorbidities, treatment, and serum levels of 25-hydroxyvitamin D (25(OH)D) on depressive symptoms in sarcoidosis patients.</p><p><strong>Methods: </strong>In a cross-sectional study, we measured depressive symptoms using the Center for Epidemiologic Studies - Depression Scale (CES-D) and fatigue using the Fatigue Assessment Scale (FAS). Presence of depressive symptoms was defined with CES-D scores ≥16 and ≥20, respectively. Fatigue was defined as having an FAS score of ≥22.</p><p><strong>Results: </strong>A total of 400 patients were included in the study. CES-D score ≥16 had 128 patients, while CES-D score ≥20 had 86 patients. In a multivariate binomial logistic regression model, the following independent predictors of CES-D score ≥16 were identified: female gender (odds ratio, OR 1.983), chronic sarcoidosis (OR 2.311), serum levels of 25(OH)D ≤20 ng/mL (OR 2.326), persistent dry cough (OR 2.173), FAS Scores ≥22 (OR 9.243), and chest X-ray stage 3 (8.851). {"title":"The significance of serum sST2 and cfDNA in children with severe pneumonia complicated by myocardial damage.","authors":"Tingting Zhao, Ye Liu, Haoran Jia, Dexing Wang, Meng Du, Weiwei Wang","doi":"10.5937/jomb0-51197","DOIUrl":"10.5937/jomb0-51197","url":null,"abstract":"<p><strong>Background: </strong>The paper aimed to explore the significance of serum soluble ST2 (sST2) and circulating cell-free DNA (cfDNA) in predicting cardiac functions in children with severe pneumonia complicated by myocardial damage.</p><p><strong>Methods: </strong>This case series study evaluated the serum sST2 and cfDNA levels of 60 children with severe pneumonia complicated by myocardial damage, assessing clinical data, biomarker levels, and cardiac function.</p><p><strong>Results: </strong>We analyzed data from a cohort of 60 patients with a mean age of 4.47±1.88 years and a male: female ratio of 28:32. At baseline, patients had elevated levels of serum biomarkers, including sST2 and cfDNA, which were associated with cardiac function parameters and clinical outcomes. After 6 months, patients showed significant correlations between sST2, cfDNA, and cardiac function parameters, including left ventricular end-diastolic diameter (LVEDd), left ventricular end-systolic diameter (LVESd), and E/A ratio. {"title":"Effects of use of metformin or combination of metformin and pioglitazone on oxidative stress in type 2 diabetes mellitus.","authors":"Temiz Gençoğlu Ayşe Tuğçenur, Kalkan Emra Asfuroğlu, Oğuzhan Zengin, Özcan Erel, İhsan Ateş","doi":"10.5937/jomb0-50062","DOIUrl":"10.5937/jomb0-50062","url":null,"abstract":"<p><strong>Background: </strong>We aimed to examine how oxidative stress changes after treatment and its relationship with metabolic parameters in patients diagnosed with Type 2 Diabetes Mellitus treated with metformin alone or with a combination of metformin and pioglitazone.</p><p><strong>Methods: </strong>The study population consisted of a total of 60 patients, including the group diagnosed with Type 2 Diabetes Mellitus (T2DM) who received metformin 2x1000 mg (n=30) and the other group who received metformin with pioglitazone 2x15/1000 mg (n=30). Serum IMA (ischemia-modified albumin), TAS (total antioxidant status), TOS (total oxidant status) and thiol-disulphide homeostasis were measured before and after 12 weeks of treatment.</p><p><strong>Results: </strong>No significant change was detected in Native sulfhydryl (SH) and Total SH levels in both Metformin and combination groups. IMA levels increased significantly in both drug groups (p=0.03 and p=0.01, respectively). Although the TAS level increased in both groups, no significant change was detected. While the TOS and oxidative stress index (OSI) index decreased significantly in the combination group (p<0.001), the decrease in the metformin group was not significant. Noradrenaline is a widely used vasopressor to manage septic shock, but its optimal dosage remains unclear. This study aimed to investigate the effects of noradrenaline doses on the prognosis of patients with sepsis/septic shock and identify the influencing factors for patient survival.</p><p><strong>Methods: </strong>A retrospective study was conducted on 126 patients with sepsis/septic shock who received noradrenaline treatment in the intensive care unit (ICU). Patients were divided into survival (n=91) and death (n=35) groups.
