The patients were divided into two groups according to positive and negative ATAbs, and the clinical characteristics, experimental results and prognoses of the two groups were compared. \u0000 \u0000 \u0000Results \u0000Among the 53 patients, 16 were combined with positive ATAbs, of which 11 were female (68.8%); 37 were combined with negative ATAbs, of which 11 were female (29.7%); number of female patients in the positive ATAbs group was significantly larger than that in the negative ATAbs group (P 0.05). There were no significant differences in prodromal manifestations, first symptoms (abnormal mental behavior or cognitive impairment, and epilepsy), main clinical manifestations, and incidence of complications between the two groups (P>0.05). Its possible pathogeneses include overlapping mechanism of vestibular pathways and anxiety-related neural networks, classical and operant conditioning, failure of re-adaptation, changes in brain activity areas, and molecular pathological mechanisms. Current treatments include psychotherapy, medication, vestibular and balance rehabilitation therapy, cognitive behavioral therapy, and non-invasive vagus nerve stimulation for refractory persistent posture-perceived dizziness. Five d after modeling, the 48 successfully modeled nude mice were randomly divided into solvent control group, Res group, TEM group, combination drug group, Wnt signaling pathway agonist group, and Wnt signaling pathway inhibitor group(n=8); and dimethyl sulfoxide (10 mg/kg), Res (10 mg/kg), TEM (25 mg/kg), Res (10 mg/kg+TEM (25 mg/kg), Res (10 mg/kg)+TEM (25 mg/kg)+lithium chloride (2 mg/kg), and Res (10 mg/kg)+TEM (25 mg/kg)+IWR-1 (5 mg/kg) were given, respectively, once/d for 30 d. During the administration, the survival status of nude mice in each group was continuously observed, tumor volume was measured by MR imaging every 5 d. Thirty d after administration, TUNEL was used to detect the apoptosis of tumor cells, and immunofluorescence was used to detect the immunofluorescent intensity of O6-methylguanine-DNA methyltransferase (MGMT) and β-catenin in the tumor tissues. Western blotting was used to detect the protein expression levels of Wnt signaling pathway-related proteins (Wnt2, and β-catenin), MGMT, and glycogen synthase kinase 3β (GSK3β). \u0000 \u0000 \u0000Results \u0000As compared with the TEM group, the combination drug group and Wnt signaling pathway inhibitor group had significantly decreased tumor volumes 20, 25, 30, and 35 d after modeling (P<0.05); as compared with the combination drug group, the Wnt signaling pathway inhibitor group had significantly decreased tumor volumes while Wnt signaling pathway agonist group had significantly increased tumor volumes 20, 25, 30, and 35 d after modeling (P<0.05). TUNEL showed that the apoptosis rate of tumor cells in the combination drug group and Wnt signaling pathway inhibitor group was significantly increased as compared with that in the temozolomide group (P<0.05); as compared with that in the TEM group, the apoptosis rate of tumor cells in the Wnt signaling pathway inhibitor group was significantly increased while that in the Wnt signaling pathway agonist group was statistically decreased (P<0.05). Among them, 63 patients accepted direct mechanical thrombectomy and 53 accepted bridging therapy. According to modified Rankin scale (mRS) scores 90 d after surgery, patients were divided into good prognosis group (mRS scores≤2) and poor prognosis group (mRS scores ≥3). Univariate analysis was used to compare the general data (age, gender, body mass index, and underlying diseases) and perioperative conditions (immediate heart rate, systolic and diastolic blood pressures immediately after admission, operative time, and anesthesia time) between the two groups of patients. Multivariate Logistic regression analysis was used to identify the perioperative risk factors influencing the prognoses of patients with acute ischemic stroke accepted mechanical thrombectomy. \u0000 \u0000 \u0000Results \u0000Among the 108 patients, 65 had good prognosis and 43 had poor prognosis. Univariate analysis showed that there was no significant difference in general data between the two groups (P>0.05), but there were significant differences in heart rate immediately after admission, National Institutes of Health Stroke Scale (NIHSS) scores immediately after admission and 3 d after operation, maximum hemoglobin and blood glucose values from immediately after admission to 3rd d of operation, and thrombolysis in myocardial infarction (TIMI) blood flow classification (P<0.05). Intraoperative ultrasound could clearly show the locations and sizes of the lesions, and relations of lesions with surrounding structures to choose appropriate surgical routes. The degrees of resection were judged and complications were recorded. The patients were followed up for 3-6 months, and modified Rankin scale (mRS) was used to evaluate the prognoses. \u0000 \u0000 \u0000Results \u0000In these 31 patients, 35 cavernous hemangiomas were diagnosed by MR imaging before surgery; 33 lesions were removed surgically, and the other two lesions located in the contralateral side of the operative field were not removed. The accuracy of intraoperative ultrasound guiding the lesions was up to 100%. There were no postoperative infections or deaths. (2) Experiment two: SH-SY5Y cells were treated with 0, 1, 3, 5, 7 mmol/L Rop, respectively; the cell activity was measured 48 h after Rop treatment; the semi inhibitory concentration (IC50) of Rop was calculated by MTT assay; the SH-SY5Y cells were divided into control group (PBS for 48 h), Rop group (Rop at IC50 for 48 h), BDNF+Rop group (20 μg/L BDNF for 2 h, and Rop at IC50 for 48 h), Akt pathway activator SC79+Rop group (5 mg/L SC79 for 2 h, and Rop at IC50 for 48 h), and BDNF+Akt pathway inhibitor API-2+Rop group (20 μg/L BDNF+10 μmol/L API-2 for 2 h, Rop at IC50 for 48 h); the morphological changes of the cells were observed under microscope; MTT assay was used to detect the cell activity; flow cytometry was used to detect the cell apoptosis, and immunohistochemistry was used to detect the expressions of Akt and PCNA; the expressions of B lymphoma-2 (Bcl-2), Bcl-2-associated X (Bax) and cysteine protease-3 (Caspase-3) were detected by reverse transcription (RT)-PCR and Western blotting. Western blotting was used to detect the expressions of Akt and phosphatidylinositol 3-kinase (PI3K). \u0000 \u0000 \u0000Results \u0000(1) As compared with the 0 mmol/L Rop group, the 1, 2, 3, 4, and 5 mmol/L Rop group had significantly decreased cell activity, significantly increased apoptosis rate, and statistically smaller number of Akt and PCNA positive cells (P<0.05).
