Helgeland","doi":"10.1016/0926-6534(65)90035-7","DOIUrl":"10.1016/0926-6534(65)90035-7","url":null,"abstract":"<div><p>The incorporation of [1-<sup>14</sup>C[glucosamine into various submicrosomal fractions and cell sap from rat liver, as well as into plasmaproteins, has been examined after intraperitoneal injection. It has been shown that after 1 h there is a significant incorporation of protein-bound radioactivity into the submicrosomal fractions. 69% of the total radioactivity was found in the ultrasonic extract of the microsomal pellet, 28% in the subsequent deoxycholate extract and 3% associated with the ribosomes. Of the total radioactivity in the ultrasonic extract and the deoxycholate extract, more than 80% was found in the perchloric acid precipitate and only about 5–10% in the phosphotungstic acid precipitate. 15% of the protein-bound radioactivity in the perchloric acid precipitate of the ultrasonic extract was associated with a sialic acid fraction and about 1% found in the hexose fraction. Most of the remaining radioactivity was associated with glucosamine. Horatinsulphuric acid, a mixture of analogous polymers, is obtained by the method of trypaflavin precipitation of sulphate esters. Fractionation made by column chromatography using DEAE-Sephadex separates the mixture into several fractions in which the sulphate content varies from 10 to 30%. Cell wall mucopeptide of <em>Bacillus anthracis</em>, strain V<sub>1</sub>B-189 was prepared from cells grown in the presence and absence of penicillin. Both types contain an unusual inner membrane.</p></span></li><li><span>2.</span><span><p>2. These were analysed for reducing sugars, amino sugars and amino acids. Both types contained glutamic acid, alanine and diaminopimelic acid as the principle amino acids in the mole ratios of 1:1.5:1.</p></span></li><li><span>3.</span><span><p>3. The structure of the polysaccharide backbone of heparin was investigated by means of graded hydrolysis procedures.</p></span></li><li><span>2.</span><span><p>2. Desulfated acetamido heparin was partially hydrolyzed with acid and the products were fractionated on a cation exchange resin. This yielded glucosamine, disaccharides and a mixture of oligosaccharides.</p></span></li><li><span>3.</span><span><p>3. Two disaccharides were isolated as their acetamido derivatives. One of these was identified as <span><math><mtext>4-O-(2-</mtext><mtext>acetamido-2-deoxy-α-</mtext><mtext>d</mtext><mtext>-glucopyranoside)-</mtext><mtext>d</mtext><mtext>-glucopyranosyl-uronic acid</mtext></math></span> and the other as <span><math><mtext>4-O-(</mtext><mtext>d</mtext><mtext>-glucopyranosyluronic acid</mtext><mtext>)-2-</mtext><mtext>acetamido</mtext><mtext>-2-</mtext><mtext>deoxy-</mtext><mtext>d</mtext><mtext>-glucopyranose</mtext></math></span>.</p></span></li><li><span>4.</span><span><p>4. The <em>K</em><sub><em>m</em></sub> for cytidine 5′-diphosphateglycerol is 1.6·10<sup>−3</sup><em>M</em>. It is suggested that the function of this enzyme is to prevent the accumulation of cytidine 5′-diphosphateglycerol under conditions where the rate of teichoic acid synthesis is decreased.
