One is the induction of the vegetally-located stem cell of the mesoderm (the 3D macromere) by the first-quartet micromeres (which are located in the animal hemisphere). The other is the induction of dorsoventral symmetry in the first-quartet micromeres by the 3D macromere. As a consequence of the latter induction process, a dorsoventrally-organised prototroch is formed. In this paper, the moment of 3D induction is determined by deleting the inducing first-quartet micromeres at successive stages of development. Furthermore, the role of gap junctions in the two above-mentioned induction processes is investigated. This was done by studying lucifer yellow dye transfer between the 3D macromere and the first-quartet micromeres, in relation to these two induction processes. Analysis of the pattern of dye-transfer in vivo, in sections and with confocal laser scanning microscopy demonstrated that the moment of dye transfer coincides with the moment of 3D induction and with the moment of induction of the dorsoventral pattern of the prototroch. This indicates that gap-junctional communication may be involved in these two induction processes.
","PeriodicalId":91963,"journal":{"name":"Roux's archives of developmental biology : the official organ of the EDBO","volume":"205 7-8","pages":"401-409"},"PeriodicalIF":0.0,"publicationDate":"1996-05-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1007/BF00377220","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34821892","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Centrosomes labelled with this antibody are found at both poles of the first meiotic spindle and at the inner pole of the second meiotic spindle. At the transition to the second meiosis, there is no change in morphology of the centrosomes which are retained in the egg proper. In contrast, as the second meiosis proceeds from anaphase to telophase, centrosomes labelled with the antibody gradually become smaller, but are still recognized as tiny dots; each egg exhibits no more than one tiny dot. The first cleavage spindles exhibit a centrosome at one pole but not at the other. The spindle pole with a centrosome forms an aster which is inherited by the larger cell, CD, of the two-cell embryo; the centrosome-free spindle pole then becomes anastral and is segregated to a smaller cell AB. Centrosomes are present in the C and D cell lineages but not in the A and B lineages, at least up to the eighth cleavage cycle. During cleavage stages, centrosomes duplicate early in telophase of each mitosis, and their size changes in a cell cycle-specific fashion. Centrosomes which otherwise duplicate asynchronously in separate cells do so synchronously in a common cytoplasm. Centrosome duplication is inhibited by nocodazole but not by cytochalasin D. An examination of embryos treated with cycloheximide or aphidicolin also suggests that centrosome duplication during cleavages requires protein synthesis but no DNA replication per se. These results suggest that the centrosome cycle in Tubifex blastomeres is linked to the mitotic cycle more closely than is that in other animals.
","PeriodicalId":91963,"journal":{"name":"Roux's archives of developmental biology : the official organ of the EDBO","volume":"205 5-6","pages":"290-299"},"PeriodicalIF":0.0,"publicationDate":"1996-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34822851","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Heart formation was scored by an original heart index based on morphology. These explant studies revealed that anterior ventral endoderm plays a critical role in heart morphogenesis. RNA blot analysis and nuclear run-on assay showed that collagenase genes of caudal cells of tadpoles of the bullfrog, Rana catesbeiana, are up-regulated by thyroid hormone after only 24 h. The gene for collagenase was cloned from genome DNA of the bullfrog and its structure, including the 5'-upstream region with-1 kbp, was determined. The gene contained 4 exons with sequences of the enzyme active centre in exon 2. Cis-acting regulatory sequences were recognized in the 5'-upstream region as AP-1 and Sp1, which had also been found in the mammalian collagenase gene. A special sequence was noticed near -870 bp, 5'-AG-GTAAGAACAGGATA-3', that satisfies a general criterion for the identification of the thyroid hormone responsive element (TRE) proposed by Umesono et al. (1991) and is homologous to the element reported in genes of thyroid-hormone responsive proteins such as thyroid hormone receptor (TR) β, growth hormone, malic enzyme and myosin heavy chain. Results of the gel mobility shift assay supported the idea that the sequence functions as a TRE. The supershift procedure demonstrated that the complex formed in the gel mobility shift assay contains TR. The present study revealed the uniqueness of bullfrog collagenase as a metamorphosis-associated enzyme, because, to our knowledge, known mammalian collagenase genes do not contain THE in their transcription regulatory regions.
","PeriodicalId":91963,"journal":{"name":"Roux's archives of developmental biology : the official organ of the EDBO","volume":"205 5-6","pages":"243-251"},"PeriodicalIF":0.0,"publicationDate":"1996-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34822882","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

We tested this hypothesis in a quail embryo model by examining the expression of retinoic acid receptors (RARs) and cytosolic retinoic acid binding protein (CRABP) in normal and vitamin A-deficient embryos during early development. RARα and RARγ mRNA were expressed at the same level in normal and vitamin A-deficient embryos during all stages studied. Expression of CRABP I was low in normal and vitamin A-deficient quail embryos during early development, but increased rapidly at later stages. Two transcripts of RARβ, 3.2 and 3.5 kb, were detected in quail embryos during developmental stages 6-12. In normal emryos the level of the 3.2-kb isoform increased as embryonic development advanced. The expression of the 3.5-kb transcript was significantly decreased in vitamin A-deficient embryos, while the 3.2-kb transcript was undetectable by northern analysis. In situ hybridization of stage 7-8 normal quail embryos using a chicken RARβ2 riboprobe revealed that RARβ2 expression was predominantly associated with the cell populations in heart-forming regions, somites, neural folds, notochord and the presumptive thyroid. In stark contrast, in the vitamin A-deficient quail embryo RARβ2 was not expressed in any of the above cell populations. We conclude that the expressions of RARβ and CRABP I are developmentally regulated. Additionally, the expression of RARβ2 during early embryogenesis is regulated by vitamin A status. We propose that RARβ2 plays an important role in the mechanism of action of retinoids in early avian development. The lack of expression of RARβ2 may be linked to abnormalities of the cardiovascular system.
","PeriodicalId":91963,"journal":{"name":"Roux's archives of developmental biology : the official organ of the EDBO","volume":"205 5-6","pages":"260-271"},"PeriodicalIF":0.0,"publicationDate":"1996-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1007/BF00365804","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34822884","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Here we have cloned the Xenopus HGF gene, covering a 14 kb 5'-upstream region and a 0.2 kb 5'-coding region. Within about 0.5 kb of the 5'-flanking region, the Xenopus HGF gene contained a TATA-like element AATGAAA, one putative NF-1 binding site, two NF-IL-6 binding motif sequences, one putative TGF-β-dependent inhibitory element (TIE) and one AP-1 binding site. A recombinant circular plasmid consisting of a 1.7 kb HGF promoter region and the bacterial chloramphenicol acetyltransferase (CAT) gene was first expressed at the late gastrula stage in the ventral mesoderm, as was the endogenous HGF gene. The expression of the fusion gene was induced in animal cap cells by activin A and bFGF although induction by the latter was not so strong. Using a series of 5'-deletion constructs introduced into animal caps, silencer elements, which seem to be essential for the gene's regionally correct expression, and the element responsible for induction by activin were found. The results show that the HGF gene promoter isolated here contains elements which may endow the gene with the regulative function for its temporally and spatially regulated expression, although the element necessary for induction by bFGF seems to be missing.
","PeriodicalId":91963,"journal":{"name":"Roux's archives of developmental biology : the official organ of the EDBO","volume":"205 5-6","pages":"300-310"},"PeriodicalIF":0.0,"publicationDate":"1996-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34822852","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Reamputation increased both the percentage rate of regeneration and the morphological complexity of the regenerates in all experimental series. Cell counts in lateral motor columns and spinal ganglia innervating the hindlimb, together with histological observations and mitotic index and labelling index determinations in reamputated and control limbs showed that improved regeneration in the reamputated limb was related to an increase in undifferentiated and proliferating cells in the stump. This study sought to determine whether the same pattern of axon growth is shared by an apterygotic insect, the silverfish. We have found that homologues to at least nine early differentiating winged insect neurons are present in the silverfish. The axon trajectories and the sequence of axon outgrowth from these neurons are very similar in silverfish and winged insects, suggesting that the pterygotic and apterygotic insects share a common developmental Bauplan for the construction of the central nervous system. Some of these neurons do show differences in several aspects of axon growth, including the relative timing of axonogenesis, the polarity of axon growth and the pattern of axon fasciculation. In addition, a major, early-appearing fascicle in the posterior commissure of the silverfish is pioneered by a neuron which does not appear to have an equivalent in the winged insects. These differences are similar in character to, albeit more pronounced than, differences previously reported between two winged insects, the fruitfly Drosophila and the grasshopper. Some of the features of early central axon growth, that set the silverfish embryo apart from the winged insects, are shared by crustacean embryos, providing support for the claim that insects and crustaceans share a common developmental Bauplan for the construction of central axonal pathways.
","PeriodicalId":91963,"journal":{"name":"Roux's archives of developmental biology : the official organ of the EDBO","volume":"205 5-6","pages":"272-281"},"PeriodicalIF":0.0,"publicationDate":"1996-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.1007/BF00365805","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"34822885","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In order to identify the functional homologue in this species we have cloned cDNAs of the precursor protein from Hydractinia and, for comparison, precursor sequences from two further anthozoans. The deduced preproproteins contain multiple copies of propeptides to be processed into a great variety of novel neuropeptides most of which are N-terminally different from MMA. Original MMA is only contained in the anthozoan precursors. Most of the novel neuropeptides will have the carboxyl terminus LWamide. Therefore, we term this novel neuropeptide family the LWamides. Peptides synthesized according to the precursor sequence of H. echinata and added to planulae trigger metamorphosis. In contrast, none of 11 other known biologically active peptides including carboxamidated neuropeptides were effective. Expression analysis by in situ hybridization and by antibodies against the H. echinata peptide reveals the presence of the gene product in planulae at the proper time and at the due spatial location expected for a natural role in metamorphosis.
