Considering the importance of knowing the microbiota of wild and exotic birds and the scarcity of epidemiological data in the Amazon region, the aim of this study was to evaluate the fungal diversity of the gastrointestinal tract of psittacines kept as pets in the municipality of Rio Branco, Acre, Western Amazonia, Brazil. Between August 2020 and December 2021, 20 psittacines underwent clinical evaluation and samples were taken from the oral and cloacal cavities by swabbing. The biological material was sent for fungal cultivation and the isolates were analyzed according to their morphological and biochemical characteristics. The gastrointestinal tract samples from 13 birds resulted in 64 isolates distributed among 15 fungal genera. According to the data obtained, there is a high diversity of fungal microorganisms of zoonotic interest in the gastrointestinal tract of psittaciformes. Eight treatments were tested to evaluate LAB viability under varying concentrations of nitrite (0.5-2.0 mg/L), ammonia (1.0-3.5 mg/L), pH levels (6.0-8.5), and temperatures (20 °C and 26 °C), as well as their effects on nitrite and ammonia concentrations in the system. Except for the bioremediator treatment at 26 °C, where no LAB viability was observed after 5 h at ammonia concentrations above 1.5 mg/L, all other treatments supported the growth of the strains under the tested conditions. The viability of LAB in recirculating aquaculture systems (RAS) was confirmed, with concentrations ranging from 13.0 to 14.0 log CFU/mL detected up to three days after inoculation in the presence of fish. After this period, reapplication was necessary. Additionally, ammonia and nitrite levels fluctuated over time, demonstrating the influence of LAB on water quality. The virulence of its etiological agent, Corynebacterium pseudotuberculosis, depends on the activity of enzymes, such as phospholipase D, and its waxy lipid mycolic acid layer, which induces cytotoxicity on the tissue cells of the host. Phospholipase D and mycolic acids contribute to inflammation, edema, and bacterium spread during abscess development. Here, we assessed the adhesion and internalization capacity of six isolates of C. pseudotuberculosis biovar ovis in a non-phagocytic cell lines (Madin-Darb Bovine Kidney (MDBK)) and the cytopathic effects produced in these cells. The interaction of the bacteria strains with MDBK as assessed through scanning electron microscopy and, their cytopathic effects was analyzed by Giemsa staining under light microscopy. Both microscopy assays were monitored after 6, 24, and 72 h of infection. The cytopathic effects evaluated were the formation of cytoplasmic vacuoles, alteration in the nucleus/cytoplasm ratio, and cell lysis. Nine strains were isolated from different clinical departments at a hospital in Brazil, were identified by VITEK®2, Matrix-Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry, 16S rRNA sequencing, and 23S rRNA PCR. Enterobacterial Repetitive Intergenic Consensus-Polymerase Chain Reaction (ERIC-PCR), Fourier-transform infrared (FTIR) spectroscopy, antimicrobial susceptibility testing, and biofilm formation and disinfectant tolerance tests were applied. ERIC-PCR, FTIR, and VITEK®2 results exhibited no correlation between the strains, indicating different origins in the Hospital. Most strains expressed resistance to several antibiotics but minocycline and cefiderocol, which were therefore regarded as optimistic therapy options. All strains produced biofilms on polystyrene and most of them (n = 7) on stainless-steel surfaces. The sodium hypochlorite 0.5% was shown to be the most efficient disinfectant for biofilm eradication. While minocycline, a broad-spectrum tetracycline antibiotic, is a conventional therapeutic agent for C. acnes infections, its efficacy against biofilms remains unclear.</p><p><strong>Objective: </strong>This study employed an in vitro biofilm model to examine the effects of varying minocycline concentrations at different stages of biofilm formation.</p><p><strong>Methods: </strong>Using our established in vitro model of C. acnes early- and later-stage biofilms, we exposed the biofilms to different minocycline concentrations. Morphological changes were assessed visually, biofilm viability was measured via XTT assay, biofilm biomass was quantified by crystal violet staining, and three-dimensional structural alterations were analyzed using confocal laser scanning microscopy (CLSM).</p><p><strong>Results: </strong>A low minocycline concentration (0.25 mmol/L) inhibited biofilm formation by reducing bacterial adhesion, whereas a higher concentration (> 4.0 mmol/L) eradicated mature biofilms and eliminated embedded bacteria.</p><p><strong>Conclusion: </strong>The study results demonstrate that minocycline effectively inhibits C. acnes biofilm formation, supporting our hypothesis regarding its inhibitory effects. Additionally, this study addresses a gap in existing research on minocycline's impact on bacterial biofilms. Our findings also provide a reference for investigating minocycline's effects on biofilms formed by other bacterial species. The pathogenic potential of the strains studied was verified by the ability of the strains to invade human gut epithelial cells (Caco-2), as well as to survive in human macrophages (U-937). The virulence of eight S. flexneri strains, representative of the decades analyzed, was evaluated using the Galleria mellonella infection model. The phylogeny of the 50 strains was verified by Multilocus Sequence Typing (MLST). The percentage of invasion in Caco-2 cells was ≥ 40% and survival in U-937 was ≥ 60% for all 50 S. flexneri strains. Seven of the eight S. flexneri strains caused the death of more than 50% of the Galleria mellonella larvae. All 50 strains were typed as ST245 by MLST. In conclusion, the percentage of cell invasion, macrophage survival and G. mellonella mortality by the strains studied highlighted their pathogenic potential and virulence. This study evaluated the prevalence of AMR in E. coli isolates from dairy farms in Santa Fe, Argentina, focusing on two animal categories: pre-weaned calves and lactating cows. A cross-sectional study was conducted from November 2019 to March 2020, yielding 545 E. coli isolates from fecal samples across 54 dairy farms and three collective calf-rearing farms. Resistance was assessed using the disc diffusion test for multiple antimicrobial classes. The study revealed a higher risk of AMR in calf isolates compared to cow isolates, particularly for gentamicin and tetracycline. A significant proportion of isolates were multi-drug resistant (MDR, 25.13%), with MDR isolates predominantly found in calves and collective calf-rearing farms. The MDR isolates were 9 times more probable to be found in calves than in cows (OR = 9.09; P < 0.001). The most frequent antimicrobial (AM) class combination in MDR E. coli isolates were beta-lactams, tetracyclines, and quinolones (75.18%). Thus, the present study aimed to evaluate the in vitro antimicrobial activity of polyhexamethylene guanidine hydrochloride (PHMGH) and polyhexamethylene biguanide hydrochloride (PHMB) against standard and clinical isolated bacteria strains that causes clinical mastitis (Staphylococcus aureus ATCC 6538 and clinical isolate, Staphylococcus epidermidis ATCC 12228, Streptococcus agalactiae ATCC 12386, Streptococcus dysgalactiae ATCC 12238, and Escherichia coli ATCC 14948 and clinical isolate). The polymers were tested at concentrations ranging from 0.3 ppm to 5000 ppm and compared to the commercial product benzalkonium chloride (BKC). The minimum inhibitory concentration (MIC) technique determined the lowest concentration of polymers capable of inhibiting microbial growth, while the minimum bactericidal concentration (MBC) characterized the lowest concentration with bactericidal or bacteriostatic action. Fractional inhibitory concentration index (FICI) assessed the synergy of polymers with BKC. To determine the time required for microbial death in the presence of polymers and BKC, the bactericidal kinetics technique was employed, and the minimum inhibitory concentration for biofilm inhibition (CIMB75) assessed the lowest concentration of polymers inhibiting 75% or more of biofilm formation. The data indicated that both PHMGH and PHMB were effective against all tested microorganisms at concentrations ranging from 0.3 to 9.6 ppm, as determined by MIC and MBC. FICI showed indifferent effects of polymer combinations with BKC against most microorganisms, except for PHMGH against Staphylococcus aureus, which exhibited additive effects. Bactericidal kinetics demonstrated that polymers immediately reduced microbial load by more than 90% against tested microorganisms, unlike BKC. CIMB75 showed that polymers inhibited 75% of biofilm formation, with PHMGH proving more effective. The antimicrobial activity of the ethanol extract of S. tragus was assessed against a range of bacterial and yeast strains using the microdilution method. The results revealed significant inhibitory effects, with Minimum Inhibitory Concentration (MIC) values ranging from < 6.25 to > 50 mg/mL, indicating that the extract possesses considerable antimicrobial efficacy. Additionally, the extract demonstrated substantial biofilm inhibition against biofilm-forming strains of Pseudomonas aeruginosa, reducing biofilm formation concentration-dependently. Molecular docking studies were performed to predict the interaction between the bioactive compounds of S. tragus and the LasR-OC12 HSL complex of Pseudomonas aeruginosa (PDB ID: 3IX3). Key compounds such as 1,8-Diazacyclotetradecane-2,7-dione, phytane, ethyl oleate, and linoleic acid ethyl ester displayed notable binding affinities, with 1,8-Diazacyclotetradecane-2,7-dione exhibiting the highest binding affinity (-5.7 kcal/mol) and lowest inhibition constant (0.066 mM), suggesting strong interaction with the target protein. The study also assessed the toxicity profiles of these compounds, indicating varying levels of acute toxicity and bioavailability, with most compounds showing low toxicity and favorable pharmacological profiles. Overall, this study demonstrates the potential of S. tragus extract as a source of antimicrobial agents capable of interfering with microbial growth and biofilm formation, along with promising molecular interactions, as indicated by docking studies. Here, non-cholera Vibrio spp. were isolated from water samples in Guanabara Bay (GB, Brazil), a recreational estuary. Thirty-five strains non-susceptible to antimicrobials were investigated regarding their pathogenic and resistance profiles. Genes encoding for hemolysins and secretion systems were among the detected virulence traits. Beta-lactam, aminoglycoside, and quinolone resistance genes were investigated in strains phenotypically resistant to these classes, revealing the carriage of the bla<sub>SHV</sub> and aac(6')-Ie-aph(2')-Ia genes in 50% and 11.1% of the tested strains, respectively. Integrons (classes 1 and 3) and heavy metal resistance genes were detected, likely reflecting local pollution levels. Genome sequencing of five strains revealed further insights regarding their associated resistome and virulome, confirming phenotypic-derived results.
