They possess impressive antibacterial activity against Gram- positive pathogens. Oxazolidinones act through a unique mechanism of inhibitory bacterial protein biosynthesis. Linezolid (Zyvox TM ) has been the only drug of oxazolidinone class to reach the market after USFDA approval. Here we review our efforts directed to the discovery of novel oxazolidinone antibacterials evolved through structure-activity relationship (SAR) and an overview of contemporary on going efforts.","PeriodicalId":89320,"journal":{"name":"Anti-infective agents in medicinal chemistry","volume":"121 1","pages":"258-280"},"PeriodicalIF":0.0,"publicationDate":"2008-09-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.2174/187152108785908820","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68022098","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Unsaturated nucleoside analogues are recognized as an important class of biologically active compounds and appear to be prominent drugs in the management of several viral infections, including HSV, HIV, HBV, HCV and HCMV infections. Currently, unsaturated nucleoside mimetics, such as stavudine, abacavir and entecavir have been approved for the treatment of viral infections, while elvucitabine and  -L-2 - F-d4C are in clinical trials. The purpose of this review is to give an update of the recent developments on unsaturated nu- cleoside and nucleoside analogues, in both cyclic and acyclic forms, which possess promising therapeutic potential, mainly antiviral. It covers analogues with ring sizes from three to six and provides useful data, in the aim to enhance chemical reactivity or to study the fixation of the sugar conformation.","PeriodicalId":89320,"journal":{"name":"Anti-infective agents in medicinal chemistry","volume":"7 1","pages":"219-244"},"PeriodicalIF":0.0,"publicationDate":"2008-09-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.2174/187152108785908848","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68022163","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Glycodendrimers containing either the GM1os ligand or its greatly simplified relative galactose, were found to be very strong cholera toxin inhibitors, especially at higher valencies. Related glycodendrimers containing the galabiose epitope were strong inhibitors of the adhesion of the bacterial pathogen Streptococcus suis. Finally, a synthetic vaccine against Bordetella pertussis was prepared by conjugating three different relevant peptidic epitopes to a single scaffold.","PeriodicalId":89320,"journal":{"name":"Anti-infective agents in medicinal chemistry","volume":"7 1","pages":"193-200"},"PeriodicalIF":0.0,"publicationDate":"2008-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://sci-hub-pdf.com/10.2174/187152108784911269","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"68021708","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
