Crude sunflower oil was stored at room temperature and was regularly refined by applying physical refining process in laboratory scale studying lipid degradation. Effect of long-term storage and different steps of refining process on overall oxidative state and volatile compounds' formation was investigated by means of solid-phase microextraction (SPME). Results were achieved by a 100-μm polydimethylsiloxane fiber in the sample's headspace. Hexanal, tr-2-hexenal, tr-2-heptenal, tr-2-octenal, tr-2-nonenal, tr,tr-2,4-decadienal, α-pinene, pentylfuran, 1-octen-3-ol were used for external standard calibration. It was proved that hexanal and tr,tr-2,4-decadienal are aldehydes, which appear first in sunflower oil during long-term storage. Detection limit using splitless injection and flame ionization detector was in 1–5 mg/kg depending on individual components. SPME was successfully applied for both controlling decomposition of hydroperoxides during bleaching and aldehydes removal during deodorization. SPME and traditional static headspace techniques were compared. Temperature programming could successfully be used as a tool for retention control, providing gradient action that did not influence the detector performance. A 7-μm single carbon fiber was utilized as working electrode and detection was performed in a 50-μm i.d. fused silica detection capillary which also functioned as restriction capillary. Separation of tyrosine, 3-(3,4-dihydroxyphenyl)alanine, 3-chloro-tyrosine and 3-NO<sub>2</sub>-tyrosine was achieved in 6 min using a temperature program from 30°C to 100°C with a ramp of 8°C/min on a 25 cm 0.32 mm i.d. column packed with 3 μm Hypersil ODS. In SFE the analytes were derivatized <i>in situ</i> with pentafluorobenzyl bromide in the presence of the modifiers triethylamine and acetone to make them less polar, more volatile, and soluble in supercritical CO<sub>2</sub>. Parameters that affect derivatization and extraction were studied. Excess reagent was efficiently removed by LC prior to GC analysis. Pentane containing 3% ethyl acetate was suitable for elution of both the SFE solid phase trap and LC-GC. A LC fraction of 1600 μL could be transferred using an on-column interface and partially concurrent solvent evaporation. The whole analysis took place for 2 h in a closed system. The method was linear in the range 5–200 ng, with detection limits around 3 ng. The relative standard deviations of the method were 5–20%. The on-line SFE-LC-GC-MS system was applied for the analysis of aerosol samples collected with Teflon filters in Helsinki, Finland. Benzoic acid and butyric acid were identified at the level of nanograms per cubic meter, but pinonic acid was not observed. This versatile instrument can be operated in three separation modes, namely, pCEC, micro high performance liquid chromatography (μHPLC) and capillary electrophoresis (CE). In pCEC mode, both an electric field and a pressure can be applied across a packed capillary column, allowing fine tuning of the selectivity without changing the composition of the mobile phase. This technique also facilitates the use of gradient elution, making it more powerful for separation of complex mixtures. The technique was evaluated and compared with μHPLC by separating a mixture of organic acids and neutral pyridazinone derivatives. Experiments in pCEC were carried out with a fused-silica capillary column packed with 3-μm octadecyl silica (ODS) particles. A separation with baseline resolution was achieved for ten pyridazinone derivatives at optimum conditions. Compared with the separation results by μHPLC method, pCEC is more powerful than μHPLC for the separation of both neutral and charged compounds. We examined airborne CS degradation products heat-dispersed together with CS from canisters of a type used by law enforcement personnel for crowd and riot control. Air contaminants derived from CS were trapped using a polytetraflouroethylene filter. Analysis was by open tubular gas chromatography coupled to mass spectrometry. A liquid chromatographic system with packed capillary columns (0.2 mm internal diameter) was used to enhance sensitivity when analyzing the small sample volumes obtained after the ultrafiltration and the microdialysis. The microdialysis was performed with probes of our own construction, and to save analysis time, the microdialysis sampling was coupled on-line to the liquid chromatographic system. The reduction of back pressure in the microdialysis outlet tube and in the injector was found to be essential.
