The film of extracellular fluid comprised between the cell and the electrode surfaces defines an artificial synaptic cleft of a few hundred femtoliters volume, in which the release of minute molecular amounts of chemicals produces a sudden and important concentration rise. This guarantees the detection of the released species with an extremely high signal-to-noise ratio, as well as a determination of its instant released flux since the collection efficiency is quantitative. In other words, the assembly cell/liquid cleft/ultramicroelectrode behaves as an artificial neuronal synapse. In this review we wish to elaborate on how this artificial synaptic arrangement may be used to monitor an oxidative stress response stimulated in a human fibroblast, and demonstrate that such long-time conjectured oxidative cellular bursts involve a subtle cocktail of femtomoles of hydrogen peroxide, nitrogen monoxide, peroxynitrite and nitrite ions. NO and O 2 - . Peroxynitrite reactions, including those with thiols, transition metal centers and CO 2 , that account for more than 95% peroxynitrite reactivity in biology, and b) the hydroxyl radical/nitrogen dioxide pathway, which arises from homolysis of peroxynitrous acid. Different methods have been utilized to expose biochemical or cellular systems to peroxynitrite including a) addition of authentic peroxynitrite, either as a bolus or a continuous infusion, and b) generation of fluxes of the radical precursors . NO and O 2 - . Reaction yields and product distributions are sometimes difficult to interpret, mainly due to the multiple possible and competing radical interactions. Peroxynitrite detection relies on a) the detection of footprints, and b) the modification of probes. This method involved a Soxtec extraction step and a solid phase extraction step for the nonionic surfactant extraction from sludge and wastewater samples, respectively. Wit h both methodologies, all recovery values were higher than 70 %. Quantitation and identification of individual compound were car- ried out by liquid chromatography-mass spectrometry (LC/MS) using electrospray (ESP) as ionization mode. Reverse phase LC chromatography allowed the elution of all oligomer constituents of each homolog component into a single peak. Gathering all the oligomers into a single peak increased the peak intensity and provided a way to determine low concentration of APEOs and AEOs in different environmental matrices. Information on the oligomers distribution of APEOs and AEOs could be obtained by the extraction of selected m/z ions from the TIC chromatograms. Positive ESP LC/MS analyses of AEOs and APEOs yielded pri- mary pseudo-molecular ions of the (M + Na)+ type for each oligomer. The negative ionization mode allowed the determination of the two metabolites, nonylphenol and octylphenol, characterized by the pseudo molecular (M-H) - ion. Limits of detection achieved were 0.1 μg/L in wastewater sample and 10 ng/g in sludge samples. For this purpose we humidified the headspace of dry and lowmoisture products to simulate the artifacts produced during the analysis of naturally moist products. This method implemented with and without dry purge of the trapping adsorbent also enabled us to determine the origin of the observed interference. Materials and methods Samples Two food products of animal origin, namely dehydrated beef stock and powdered Parmesan cheese, were selected for their low water content (respectively 2.3 and 25.0 %) and for the diversity of the chemical families present in their volatile fractions. Description of experiments carried out To target the origin of the water-related artifacts, different analyses were carried out with and without humidification of the headspace of the products, and with and without a dry purge step. A preliminary discussion is offered to explain the chemical basis for this assay and emphasize the importance of measuring NO. After reviewing some practical aspects and caveats of the chemiluminescence assay, we review its application in a variety of research and clinical settings, such as measurement of breath NO and serum nitrates. The importance of avoiding confounding effects of dietary nitrates when assessing the NO system in humans is discussed and new data are provided on the confounding effects of atmospheric NO on measurement of breath NO. The utility and ease of the chemiluminescence assay for these applications is contrasted with the challenge of using chemiluminescence for correlation of vascular tone and NO production, studies in which real-time measurements are required. Although the chemiluminescence assay is one of the most reliable, rapid and reproducible assays available, it is optimal for gas phase measurements and may be suboptimal for measurement under certain circumstances. All detected O 2 - release but some lack of sensitivity and specificity in probes and enzymes occured. Cytochrome c prepared with trichloroacetic acid (TCA), TCA-free or acetylated (C2506, C7752, C4186) provided similar superoxide-dismutase (SOD)-inhibitable absorbances at 20 μM. At a higher concentration (100 μM) TCA-prepared cytochrome c could no longer detect O 2 - production whereas TCA-free cytochrome c detected less O 2 - at 100 μM than at 20 μM. Catalase C-40 was not contaminated with SOD-like impurities. Lucigenin provided high artefactual amounts of O 2 - production from stimulated BAEC in the presence of NADH or NADPH. Moreover, even at a low concentration (5μM) lucigenin and NADH (or NADPH) decreased endothelium-dependent relaxation induced by acetylcholine to 50 %. The ratios of volatile, low-molecular weight PAHs versus high-molecular weight PAHs increase with side slope height. For instance, naphthalene/pyrene values increase from 0.3 to 3.4 in plants. Acenaphthene/fluoranthene values increases from 0.017 to 0.123 in soils. Moreover, soil PAHs can be classified into two categories according to variations of absolute concentrations with height : low-molecular weight PAHs showing an increase and high-molecular weight PAHs showing a decrease. These results demonstrate the occurrence of an atmospheric distillation effect which favour the concentration of high-molecular weight PAHs near the PAH source.
