{"title":"Protein aggregation in Huntington’s and Parkinson’s disease: implications for therapy","authors":"Erich E Wanker PhD (Group Leader)","doi":"10.1016/S1357-4310(00)01761-5","DOIUrl":"10.1016/S1357-4310(00)01761-5","url":null,"abstract":"<div><p>The accumulation of highly insoluble intracellular protein aggregates in neuronal inclusions is a hallmark of Huntington’s disease (HD) and Parkinson’s disease (PD) as well as several other late-onset neurodegenerative disorders. The aggregates formed <em>in vitro</em> and <em>in vivo</em> generally have a fibrillar morphology, consist of individual β-strands and are resistant to proteolytic degradation. Although the causal relationship between aggregate formation and disease remains to be proven, the gradual deposition of mutant protein in neurons is consistent with the late-onset and progressive nature of symptoms. Recently, circumstantial evidence from mouse and <em>Drosophila</em> model systems suggests that abnormal protein folding and aggregation play a key role in the pathogenesis of both HD and PD. The discovery of the genetic defect in this disease, namely mutations in the gene encoding the common cytokine receptor γ chain, γ<sub>c</sub>, was reported just over seven years ago. In the subsequent period, a tremendous amount of knowledge about the biology and function of this protein has been generated. Moreover, γ<sub>c</sub>-knockout mice have been generated and their immune systems successfully reconstituted by gene therapy. Treatment to modify this disease is currently unavailable, but needed urgently. The amyloid-cascade hypothesis proposes that amyloid β-peptide (Aβ), found in the plaques characteristic of AD, plays an early, critical role in the disease process. It follows that preventing the generation of Aβ could be therapeutically useful in all cases of AD.
