The experiments of the authors indicate that the anaphylactogenic effect of ATS is substantially stronger than that or normal horse serum. {"title":"Determination of the molecular weight of IgM by thin-layer chromatography combined with immunodiffusion in normal and pathological conditions.","authors":"J Lajos, J Veres, E Auber, A Szammet","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>An immunodiffusion method combined with thin-layer chromathography was elaborated for the assessment of the molecular weight of IgM. Sera from 100 normal individuals as well as from 162 patients with different types of paraproteinaemia, 100 patients with uveitis and 55 venous umbilical cord sera were analysed. Presence of 75 IgM, besides the usual 19S IgM was detected in 21% of the normal sera, in 27% of the paraproteinaemic sera, in 30% of the uveitic sera and in 5.4% of venous umbilical cord sera. IgM level in the sera was determined by radial immunodiffusion. The IgE concentration of the different fractions was determined by a solid-phase radioimmunoassay. The results of these studies indicate, that polyclonal IgE behaves similarly to IgE of myeloma origin. {"title":"The sensitizing properties of anti-mouse anti-thymocyte horse serum.","authors":"E Osz, L Réthy","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The authors have compared the anaphylaxis due to active and passive sensitization of mice. In the case of active sensitizing, anti-mouse anti-thymocyte horse serum (ATS), and/or normal horse serum (NHS), whereas in the case of passive sensitizing, plasma, peripheral leukocytes, spleen cells and thymocytes of sensitized animals were used. Provocation of shock was carried out by intravenous administration of ATS or NHS. Irreversible anaphylaxis occurred in a significantly higher rate in the case of ATS than NHS sensitivity, produced either actively, or passively. The humoral immune response to soluble and insoluble human IgG was evaluated by the measurement of the amount of specific antibodies. The immobilizations of antigen was made by antigen-antibody precipitation and attachment of the protein to polyacrylamide and agarose matrices. The optimum circumstances of fractionation were determined and it was found that the majority of IgM is precipitated between 2 and 5% polyethylene glycol concentration and 1.1 and 1.8 M ammonium sulphate concentration. They evaluate their results in relation of age, sex and occupation and presented literary data about the subtypes of the carriers in Budapest. They compared them with data of Hungarian and several other authors respectively.
