The purpose of this study was to breed all-male large yellow croaker. To achieve this goal, we first successfully induced sex reversal of YY supermale fish to produce YY pseudo-female through adding 17β-estradiol (E<sub>2</sub>, 120 mg/kg). These YY pseudo-female were then crossed with YY supermale fish to establish a pure-bred YY supermale fish population. Subsequently, the YY supermale fish were hybridized with female fish to produce all-male fish. During the breeding process, YY pseudo-female, YY supermale and XY all-male fish were identified by sex-specific molecular markers and gonad histological observation. The results showed that in the experiment of inducing sex reversal of YY supermale fish. The survival rate of YY pseudo-female fish was 63.4 %, and the hatching rate of its offspring pure-bred YY supermale fish was 68.06 %. There was no significant difference in growth, testes development stage, sex steroid hormone levels and the expression of testis development-related genes levels between pure-bred YY supermale and normal male fish. In the temperature tolerance assay, the all-male group exhibited significantly higher survival rates than the control under both high- and low-temperature stress. In the hypoxia tolerance experiment, the respiratory oxygen consumption rate of all-male fish decreased more with the decrease of dissolved oxygen in water, and the asphyxiation point was lower. It shows that the all-male fish is more adaptable to the adverse environment. In summary, it is feasible to obtain a pure-bred YY supermale fish population by inducing YY supermale fish sex reversal. Recent research has shown the potential of <em>Lasia spinosa</em> Thw. (LST) to improve semen quality in boars, so we aimed to investigate the effect of LST supplementation on semen quality in subfertile bulls, focusing on semen characteristics in both fresh and frozen–thawed ejaculates as well as the transcriptomic changes in testicular tissues. Six imported beef bull sires presenting poor semen quality for &gt; 5 months at the Bull Semen Production Center were included in the experiment. They received daily supplementation with LST (5 g/100 kg body weight). Semen and serum testosterone evaluations were performed weekly for 24 weeks. For histologic and transcriptomic evaluations, testicular biopsies were performed on three LST-supplemented bulls, pre- and post-supplementation, and three control bulls. We observed significant improvements in movement kinetics, viability, acrosome integrity, membrane integrity, and sperm head morphology in LST-supplemented bulls. Frozen–thawed semen presented significant enhancements in parameters such as motility and mitochondrial activity. Serum testosterone significantly increased after supplementation, correlating significantly with sperm motility and mitochondrial activity. Transcriptomic analysis identified 16 downregulated genes in subfertile bulls; LST supplementation induced upregulation of 11 genes related to germ cell structure, spermatogenesis, and sperm quality, including <em>CNFN</em>, <em>KRT17</em>, <em>KRT1</em>, <em>KRT19</em>, <em>DSC3</em>, <em>KLK8</em>, <em>SPRR4</em>, <em>SPRR2E-like</em>, Serpin B4-like, and <em>WFDC18-like</em>. Transcriptional studies comparing both types of embryos mainly utilized oocytes retrieved from slaughterhouse material for IVP, thereby introducing the possibility, that differences between IVP and <em>in vivo</em> embryos are in part attributable to differing sources of oocytes. The aim of the present study was therefore to compare the proteome of oocytes retrieved from slaughterhouse material, with and without a period of <em>in vitro</em> maturation, and <em>in vivo</em> matured oocytes obtained from superovulated donor cattle. For each group the protein pattern of four technical replicates containing ten oocytes each were analyzed via SWATH™-MS. In total, 1208 proteins were detected, 160 of which were differentially abundant in at least one group comparisons. Clustering analysis revealed that oocyte maturation environment is the primary driver of molecular difference, with the <em>in vitro</em> matured oocytes exhibiting a highly distinct proteome compared to both the <em>in vivo</em> matured and immature oocytes. <em>In vitro</em> matured oocytes were characterized by an overall enhanced translational activity affecting mainly proteins involved in energy metabolism, transcription and translation, and oocyte activation. An organized regulation of key pathways was observed during oocyte maturation <em>in vivo</em>, e.g. activation of autophagy and apoptosis pathways, while changes in protein abundance <em>in vitro</em> seem to be undirected. These attributes position TC as a valuable candidate for conservation and selective breeding. Nevertheless, their suboptimal growth performance and low reproductive efficiency have restricted the broader application of this genetic resource. To date, few studies have explored the molecular mechanisms underlying reproductive function in TC ovarian tissues. In this study, we conducted a 30-day monitoring of egg production rates in TC and Romaine chickens (RC). During the peak laying period, we measured the expression levels of follicle-stimulating hormone (FSH) and progesterone (P4). Additionally, we performed whole transcriptome sequencing (RNA-seq) on ovarian tissues collected from three TC and RC specimens to construct a competing endogenous RNA (ceRNA) regulatory network associated with the reproductive performance of TC. RC hens showed a higher egg-laying rate (<em>P</em> &lt; 0.001) and higher expression levels of follicle-stimulating hormone and progesterone (<em>P</em> &lt; 0.01) than TC hens. Whole-transcriptome sequencing analysis revealed substantial differences in gene expression profiles, identifying 2314 differentially expressed mRNAs (DEmRNAs), 532 differentially expressed long non-coding RNAs (DElncRNAs), and 148 differentially expressed microRNAs (DEmiRNAs) between the two groups. Based on these findings, ceRNA network was constructed. In summary, the mRNAs, lncRNAs, and miRNAs identified in the ceRNA network constructed in this study may play critical roles in regulating follicular development and hormone secretion in TC. During the critical sex differentiation window (15–60 days post hatching), administration of 100 mg/kg LNG through prey-mediated delivery induced 100 % sex reversal of genetic females into functional neo-males, as confirmed by histological and molecular analyses. The neo-males exhibited normal testicular development, complete spermatogenesis, and reproductive competence comparable to XY males. <em>In vivo</em> gene expression analysis revealed that the masculinized state induced by LNG was characterized by the significant upregulation of male-biased genes (<em>dmrt1</em>, <em>gsdf</em>, <em>amh</em>, <em>sox9</em>, and <em>arβ</em>) and the suppression of female-biased genes (<em>foxl2</em> and <em>cyp19a1a</em>). Furthermore, our <em>in vitro</em> results suggested that LNG could act directly on ovarian cells to induce a similar molecular shift, also modulating reproductive regulation-related genes (<em>fshr</em> and <em>lhr</em>). Crossbreeding experiments demonstrated that the all-female progeny derived from LNG-induced neo-males showed 21 % greater growth performance by 240 dph compared to mixed-sex populations. For that, triplicate groups of golden mahseer adults (female: 0.8–1.3 kg, <em>n</em> = 6; male: 0.5–1.1 kg, <em>n</em> = 3) were fed with either of two diets - control and test diet. Broodstocks fed the test diet showed improved reproductive outcomes, such as a marginal increase in fecundity and a significantly greater fertilization rate, in comparison to control diet. The incubation period and hatching were unaffected by the broodstock diet; however, eggs from test diet-fed broodstocks had a shorter hatching window. Survival of larvae from these broodstocks was significantly higher. Egg quality assessment showed that broodstocks fed test diet produced significantly larger eggs with higher carotenoid content. Male broodfish fed the test diet showed significant improvement in milt quality parameters, <em>viz.</em> milt volume, sperm count, spermatocrit, and sperm viability, with decreased sperm abnormalities. Although there were no significant changes in seminal plasma protein, glucose, or triglyceride levels, the total antioxidant capacity was significantly higher, and thiobarbituric acid reactive substances (TBARS) were lower in the test diet group. In females that were fed the test diet, plasma 17β-estradiol and vitellogenin were significantly elevated. At 75 days after hatching, larvae from broodstocks fed with the test diet showed improved growth and survival and elevated critical thermal maximum. Nicotinamide mononucleotide (NMN), a precursor of NAD<sup>+</sup> and a cofactor of Sirtuin deacetylases, exhibits significant antioxidant activity. This study aims to evaluate whether NMN supplementation combined with prolonging maturation time enhances cytoplasmic maturation and improves porcine oocyte quality. The blastocyst rate increased significantly after parthenogenetic activation when maturation time was extended from 44 h to 47 h or 50 h, peaking at 47 h. Prolonged maturation improved mitochondrial function and cortical granule distribution but induced ROS accumulation and apoptosis. Notably, supplementation with 100 μM NMN reversed these negative effects, significantly increasing the total number of blastocyst cells, reducing ROS levels, and upregulating cytoplasmic quality-related gene expression. In conclusion, extending maturation to 47 h with 100 μM NMN supplementation enhances oocyte maturation and embryonic developmental potential <em>in vitro</em>. Davies","doi":"10.1016/j.anireprosci.2025.108025","DOIUrl":"10.1016/j.anireprosci.2025.108025","url":null,"abstract":"<div><div>This study aimed to determine whether trophoblast-derived extracellular vesicles (EVs) alter the gene expression and secretion profiles of inflammatory mediators in circulating leukocyte populations. Circulating leukocyte populations (CD14<sup>+</sup>, CD4<sup>+</sup>CD25<sup>-</sup>, CD4<sup>+</sup>CD25<sup>+</sup>, CD8<sup>+</sup>, and TCR-γδ<sup>+</sup> cells) were exposed to either isolated bovine trophoblast-derived EVs (tEV), EV-free trophoblast cell culture supernatant (nEV), or whole trophoblast culture supernatant (wSP) and their transcription level and secretion of immune mediators was assessed. Extracellular vesicle concentrations measured by nanoparticle tracking analysis in the nEV, tEV, and wSP treatments were 1.78 × 10⁸, 9.81 × 10⁸, and 3.22 × 10⁸ particles/mL, respectively. Scanning electron microscopy confirmed the presence and morphology of EVs, which ranged in diameter from 25 to 308 nm, with most particles falling between 72 and 134 nm. Trophoblast-derived EVs significantly altered gene expression in multiple immune cell populations, including CD8⁺ cytotoxic T cells, CD4⁺CD25⁻ resting T helper cells, and CD4⁺CD25⁺ activated T helper cells. In CD8⁺ cells, tEVs upregulated <em>IL13</em> and showed trends toward increased <em>FOXP3</em> and <em>IL23</em> expression. CD4⁺CD25⁻ cells treated with tEVs exhibited increased <em>IL6</em> and trends toward reduced <em>IL1B</em> and <em>IL13</em>, while CD4⁺CD25⁺ cells showed elevated <em>CXCL8</em> and tendencies for higher <em>IL10, IFNG</em>, and <em>TNFA</em> expression. No significant changes in gene expression were observed in TCR-γ/δ⁺ cells across treatments. Trophoblast-derived EVs decreased the secretion of CCL4 and NCAM1. Trophoblast-derived EVs contain miRNAs and proteins involved in many biological processes including cell proliferation, stem cell biology, cell migration, and immune response. Yet, the mechanisms of TCS-induced oocyte degeneration and effective protective strategies remain unclear. This study explored the protective role of spermidine (SPD) against TCS-induced meiotic disruption and fertilization defects in porcine oocytes, focusing on its antioxidant and anti-apoptotic mechanisms. We found that TCS exposure disrupted meiotic progression by inducing spindle defects, chromosome misalignment, mitochondrial dysfunction, oxidative stress, DNA damage, and apoptosis, ultimately impairing maturation and fertilization. SPD supplementation significantly alleviated these effects by stabilizing the cytoskeleton, reducing oxidative stress, and suppressing apoptosis, thereby restoring oocyte quality and developmental competence. A total of forty Brown Swiss cows were evaluated for endometritis using rectal, vaginal and ultrasonographic examinations on pp day 28 and were divided into the following groups: Mild (<em>n</em> = 10), moderate (<em>n</em> = 10), severe (<em>n</em> = 10) endometritis and, control (<em>n</em> = 10) groups. The serum concentrations of Ca, P, Fe, Zn, and Cu were assessed at various pp time points (days 0–7, 14, 21, and 28), and notable alterations were detected in the endometritis groups. Compared to the control, Ca were significantly lower in the endometritis groups, particularly between days 1–5 and on day 21. Likewise, the concentrations of Fe, Zn, and Cu were lower in the endometritis groups, particularly on days 14 and 21. Furthermore, Receiver Operating Characteristic (ROC) analysis demonstrated that Ca, P, Fe, Zn, and Cu had significant diagnostic value for endometritis at specific pp time points. Specifically, Ca achieved the highest diagnostic accuracy on day 14 (AUC=0.920), P on day 21 (AUC=0.910), Fe on day 14 (AUC=0.950), Zn on day 28 (AUC=1.000) and Cu on day 21 (AUC=1.000).
