{"title":"Studies on medicamentous inhibitory therapy for esophageal precancerous lesions--3- and 5-year inhibitory effects of antitumor-B, retinamide and riboflavin.","authors":"P Lin, J Zhang, Z Rong, R Han, S Xu, R Gao, Z Ding, J Wang, H Feng, S Cao","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Since 1983, we have been conducting inhibitory therapy for precancerous lesions of the esophagus in two regions of Henan Province considered high-risk areas of esophageal carcinoma. Our goal was to effect a 50% reduction in the canceration rate of marked esophageal dysplasia. By means of a cytological survey, 2531 cases of marked esophageal dysplasia and 3393 cases of mild esophageal dysplasia were selected. The former were randomly divided into 3 groups for antitumor-B (ATB, a mixture of Chinese herbs), retinamide (4-ethoxycarbophenylretinamide) and placebo treatment respectively, and the latter into 2 groups treated with riboflavin and placebo respectively. Treatment was continued for 3 or 5 years (administration rate greater than 90% in all groups) and esophageal cytology reexamined (reexamination rates were 94.1% and 92.5% respectively). Our results were as follows: 1) ATB 3- and 5-year subjects saw the canceration rate of marked esophageal dysplasia drop by 52.2% and 47.3% respectively as compared to control (P less than 0.01). 2) Retinamide lowered the canceration rate by 37.3% after a 3-year treatment period, with this reduction reaching 43.2% after an additional 2 years of treatment with increased dosages (P less than 0.05, P less than 0.01). 3) In the riboflavin group, the canceration rate of mild esophageal dysplasia was reduced by 22.2% and 34.8% after 3 and 5 years of treatment respectively, but these differences were not statistically significant. In the first group, only cyclosporine solvent containing no cyclosporine was given to rats. In the second group, cyclosporine A was given intraperitoneally from the 1st to the 5th days after transplantation at a daily dosage of 2 mg/100 g BW. In the third group, besides the first courses of cyclosporine A treatment as in group II, second and third courses were given on the 30th to 34th and 60th to 64th days after transplantation. {"title":"Study on enhancement of tumor-killing effect and formation of new fluorophore in tumor cells.","authors":"X J Fan, E H Cao, S J Chi, Y X Wang","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Hyperthermia (42-44 degrees C) and photosensitizing therapy can destroy S180 tumor cells, reduce malignant ascites and prolong the survival times of mice with carcinomas. The highest curative effect was observed when using a combination of the two treatments. Heating to 44 degrees C has a greater destructive effect on tumor cells than has heating to 42 degrees C. The results show that this is due to a synergistic interaction between these two treatments. The fluorescence spectrum of S180 cells was determined before and after treatment, and the indication was that the synergistic effect is probably related to a new fluorescence product; the greater the intensity of the new fluorescence, the more marked the synergy of hyperthermia and photosensitizing therapy. The clone expressing rSMP-B was isolated by epitope selection from a rat testis lambda gt11 vector cDNA library. An Eco R1-cut 2.0 kb cDNA fragment encoding rSMP-B was identified. The cDNA fragment (RSD-1) was isolated, subcloned and restriction mapped. {"title":"Inhibition of human immunodeficiency virus reverse transcriptase by Chinese medicines in vitro.","authors":"X Tang, H Chen, X Zhang","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Thirty-five Chinese medicines, including extracts of traditional medicines or clinically useful recipes, herbs and isolated compounds, were tested for inhibitory activity against human immunodeficiency virus reverse transcriptase in vitro. Fourteen of the 35 medicines were found to be active. Baicalin, in particular, was studied in great depth. We found it to be a noncompetitive inhibitor of HIV RT, with an effective concentration (IC50) of 22 mumol/L. {"title":"ADPRT-mediated decrease of cellular NAD content and the detection of chemically induced DNA damage--development of a new short-term screening test for mutagens.","authors":"Y Yu, Y Dai, M Fang, X Chen","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>It was found that the DNA-damaging agents N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), methyl-methanesulphonate (MMS) and 4-nitroquinoline-N-oxide (4NQO) could stimulate ADP-ribosyl transferase (ADPRT) activity and reduce the cellular NAD content in a dose-dependent way. The reduction of NAD after DNA damage could be partially or completely prevented by ADPRT inhibitors, 3-aminobenzamide or nicotinamide, which showed no influence on reduction of NAD induced by metabolic blocking agents. Therefore, a simple and specific method to detect DNA-damaging mutagens by measuring ADPRT-mediated decrease of cellular NAD content was explored. Using beta-naphthoflavone, a mixed function oxygenase inducer, together with induced or uninduced human amnion FL cells, it was found that aflatoxin B1, benzo(a)pyrene, 2-acetylaminofluorene, 9,10-dimethylanthracene and ethylcarbamate could induce the ADPRT-mediated decrease of cellular NAD content, while 4-acetylaminofluorene, anthracene, isopropyl-N-(3-chlorophenyl)-carbamate, beta-propiolactone, gamma-butyrolactone, cyclophosphamide and safrol could not. The data were analyzed by computer, and the frequencies, averages of skin and organic dose per examination, and population doses per capita (He, LSD, GSD, SSD) were found. {"title":"The immunohistochemistry and in situ cDNA-mRNA hybridization of carbamyl phosphate synthetase I in enzyme-altered liver cells during carcinogenesis.","authors":"Y Yang, L Ding, S Li","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The changes of carbamyl phosphate synthetase I (CPS 1) in diethylnitrosamine-(DEN)-induced enzyme-altered liver cells were studied by means of immunohistochemical (PAP) and in situ cDNA-mRNA hybridization methods. The experimental rats were treated with DEN, 2-acetylaminofluorene (2-AAF) and 2/3 hepatectomy according to Solt-Farber's protocol and were further promoted by oral daily administration of 0.05% phenobarbital in drinking water. The results showed that the average number of lesions showing abnormal expression of CPS 1 was relatively constant over the course of the experiment (8 months), while the number of normally expressing lesions gradually decreased. The former lesions were also larger in volume than the latter ones. We conclude that in DEN-initiated lesions the abnormally expressed CPS 1 lesions may grow continuously, thus leading to the formation of larger nodules.
