The cells are tagged with 99mTechnetium. The procedure was performed on two groups of volunteers. The test was found to be very reliable (98%). A second group was studied with the administration of a placebo or 400 mg Aminophylline during the acquisition of the data. Although sodium MR signal from the brain parenchyma could hardly be detected on routine spin echo (SE) pulse sequence, free induction decay (FID) image with echo time of 1.9 msec demonstrated MR signal in the normal brain tissue. On the other hand, extracellular sodium of cerebrospinal fluid, large venous sinus, and vitreous humor offered high sodium MR signal intensity due to the quantity of sodium and the relatively longer transverse relaxation time (50-60 msec) compared to that of intracellular sodium (less than a few msec). Strong signal was obtained in gliomas with superior contrast, though the delineation of the tumor tissue from edema was as difficult as when seen on proton T2 weighted SE image. Meningioma itself gave much less of a sodium MR signal compared to glioma. Peritumoral edema associated with meningioma showed a markedly high sodium MR signal which was more prominent adjacent to the tumor, although proton SE image revealed the peritumoral edema as homogeneous. Pathological tissues such as brain tumors and edema could be readily depicted as significantly high sodium-23 MR signal, apparently different from normal brain tissues. The current status of in vivo sodium MR imaging is discussed. Use was made of an i.v. injection of 99mTc-labelled baboon serum albumin. Count ratios of lung to cardiac, liver to cardiac, and abdominal to cardiac regions were measured every 2 hr for 6 hr in control and septic shock baboons, and compared. Increased ratios obtained during shock pointed to an increase in extravascular albumin. Linear regression lines fitted to these count ratios provided regional albumin leak indices. These indices demonstrated statistically significant increases (P less than 0.05) during septic shock for the abdominal region during the 6-hr study, and for all regions, but especially the abdomen, when data were calculated over 4 hr. Investigation by manometry or radiography is invasive and nonphysiological. Scintigraphy of the clearance of small radiolabelled liquid boluses in the supine position, while sensitive and noninvasive, may also be nonphysiological and does not allow the simultaneous determination of gastric emptying. We thus studied the esophageal clearance of a semisolid test meal ingested in the upright position. Forty-seven patients with scleroderma and 24 with Sjogren's syndrome were compared with ten normal controls and ten patients with gastric emptying abnormalities but no esophageal involvement. Results of scintigraphy were also correlated with manometry and contrast radiography. Quantitative evaluation of esophageal tracer retention at ten minutes postingestion was: (mean +/- SD), 2.8 +/- 1.0% in normals, 2.9 +/- 0.9% in gastric dysmotility, 4.8 +/- 2.9% in Sjogren's syndrome, and 22.3 +/- 25.0% in scleroderma; similar results were found at 20 and 60 minutes. The T 1/2 of gastric emptying was 47.1 +/- 5.7 minutes in normals, 95.9 +/- 25.3 minutes in gastric dysmotility, 62.9 +/- 19.5 minutes in Sjogren's syndrome, and 52.9 +/- 13.5 minutes in scleroderma. We conclude that esophageal clearance of a semisolid test meal is a sensitive index of esophageal dysmotility and correlates well with results from manometry and contrast radiography but is noninvasive and quantifiable. A double isotope technique was used and the radioactivity was recorded in different organs (lungs, liver, spleen, kidneys, intestine) by a computerized gamma camera. Tc-99m-labeled erythrocytes were used as a marker for intravascular volume and In-111m-labeled transferrin for tracing extravascular plasma leakage. An organ-transferrin index (organ-TI) was calculated for each organ which corrects for changes in blood distribution. Fourteen sheep were anesthetized and ventilated. After stabilization (t = 0) all animals received E. coli endotoxin 10 micrograms/kg by IV infusion during 30 min. At t = 30, seven animals (group T) received IV infusion of terbutaline, 20 micrograms/kg/hr, during 4 h, while the other seven received normal saline and served as controls (group E). The endotoxin infusion caused an immediate and significant increase in the transferrin index in the lungs and in the liver in both groups. The transferrin index continued to rise in the control group towards the end of the experiment (t = 240), while in group T it reached a maximum 60 min after endotoxin. Four hours after endotoxin the transferrin index was significantly higher in the controls than in the terbutaline treated group, both in the lungs and in the liver (P less than 0.01). The results were compared with coronary angiographic findings, obtained within 3 months of the exercise test and Thallium-201 images, obtained on the same day and to the same exercise level as Technetium-99m-SESTAMIBI exercise. Exact segmental concordance between static Technetium-99m-SESTAMIBI and Thallium-201 was 77% and between the diastolic images and Thallium-201 76%. Concordance between static and diastolic images was 92%. Nineteen percent of all 900 segments demonstrated ischemia with both static and diastolic Technetium-99m-SESTAMIBI and 20% with Thallium-201. Overall sensitivity to detect hemodynamically significant coronary artery disease with static Technetium-99m-SESTAMIBI was 86%, specificity 78%, with diastolic images 89% and 70% and with Thallium-201 89% and 78% (differences not significant). Vessel-specific comparative detection rates revealed no statistical significant differences between both radiopharmaceuticals as well. Intra-observer variability for the static images was 12%, for the diastolic images 21% and Thallium-201 14%. Inter-observer variability for the static images was 14%, diastolic images 24%, and Thallium-201 17%. Syngeneic lymphocytes were labeled with Tc-99m at 125 microCi per 10(7) cells. Mean labeling efficiency was 67 +/- 17%, n = 44. Two groups of rats allogeneic (ALLO) (ACI/Lewis) and syngeneic (SYN) (Lewis/Lewis) were injected with labeled cells at two, four, five, and seven days post cardiac graft. At 20 hr post-injection, transplant and native hearts were harvested for radioanalysis and histology. Ratio of percent dose per gram transplant heart to native heart (T/N) revealed a significant difference between ALLO (T/N = 2.7 +/- 0.23, n = 5) and SYN (T/N = 1.73 +/- 0.12, n = 5) at two days P[F] = 0.0045. Maximum difference occurred at five days with ALLO mean T/N of 6.40 +/- 0.54, n = 5 (P[F] = 0.0001). There was no significant difference at seven days post graft with ALLO 2.85 +/- 0.30, n = 5 and SYN 2.02 +/- 0.05, n = 5 (P[F] = 0.0166).
