The chiral separation method for methyl jasmonate (MeJA) in tea (<em>Camellia sinensis</em> L.) was developed by air-assisted liquid-liquid microextraction (AALLME) with enantioselective gas chromatography-mass spectrometry (Es-GC-MS/MS). The Plackett-Burman factorial design was introduced to optimize the multiple parameters of AALLME. Sensitive Es-GC-MS/MS enabled the quantification of the MeJA stereoisomers, which were separated from each other with resolutions of 3.5, 2.7, and 2.5. The linearities were <em>R</em><sup>2</sup> &gt; 0.9971, the limits of detection were 0.08–0.12 ng/mL, the limits of quantification (LOD) were 0.28–0.39 ng/mL, and the recovery range was 80.60–107.03%. The method was applied to analyze the MeJA stereoisomer profiles in tea plants after geometrid feeding and mechanical wounding. (+)-MeJA appeared to be the main bio-active monomer that responded to injury stress. (+)-MeJA similarly increased in tea shoots at only 30 s after geometrid attack or mechanical wounding. The proposed method is superior as air-assisted liquid-liquid microextraction could induce the loss of volatile MeJA in a much shorter time, compared to the conventional enrichment procedure. A novel assay protocol for relative quantification of MeJA stereoisomers in plants responding to different injuries was applied.</p></div>","PeriodicalId":74667,"journal":{"name":"Reproduction and breeding","volume":"3 1","pages":"Pages 17-25"},"PeriodicalIF":0.0,"publicationDate":"2023-03-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"45777310","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Gynogenesis is an important breeding method in aquaculture that has been widely applied to many fish species. In this study, we adopted an improved method of gynogenesis that proceeds as follows: first, inactivated koi carp sperm were used to stimulate eggs of HM, and then the UV-inactivated koi carp (KOC) sperm and HM eggs were mixed and stirred for 20–30 s to activate the eggs. The activated eggs were placed in yellow mud at 0 °C for 20–22 min for cold shock treatment, and then placed in water at 4–6 °C, 8–10 °C, and 15–18 °C in order, each for 1–2 min. After incubation at 22–25 °C, gynogenetic <em>Hemibarbus maculatus</em> Bleeker (GHM) were obtained. Morphological measurements and blood cell comparisons showed that there were no significant differences between GHM and HM. At the molecular level, comparative analysis of microsatellites DNA indicated that one fragment of GHM came from the KOC. Murine gamma-herpesvirus 68 (MHV68) is widely regarded as a model for the study of human gamma herpesviruses such as Kaposi's sarcoma-associated herpesvirus (KSHV). In this study, the infection of MHV68 on zebrafish was investigated through incubating fertilized zebrafish eggs with MHV68. The development of zebrafish delayed or stopped at early stage separately when the zebrafish eggs were incubated with MHV68 at different dose for 2 h. The lytic gene of MHV68, including ORF65, RTA and ORF57, were detected in the genome of larvae hatched from the infected eggs. The transcription of the latent gene LANA but not of these lytic genes was detected by RT-PCR. The mRNA level of growth factor TGF-β1, but not of FGF3, was obviously decreased in the larvae from the MHV68 treated eggs. The mRNA levels of cytokines, such as IFNφ3, ISG15 and TNFα, varied post MHV68 infection; however, the transcription of the cytosolic DNA sensors, including DDX41, DHX9 and cGAS, did not change significantly. Moreover, the data generated in zebrafish cell line demonstrated that MHV68 could infect ZF4 cells. All the data support the conclusion that MHV68 could infect zebrafish embryo in the egg envelope and go into latent infection, which will delay the development of the embryos through some unknown mechanism.</p></div>","PeriodicalId":74667,"journal":{"name":"Reproduction and breeding","volume":"3 1","pages":"Pages 38-44"},"PeriodicalIF":0.0,"publicationDate":"2023-03-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"48099040","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Members of the <em>OsNramp</em> (<em>Natural resistance-associated macrophage protein</em>) gene family play important roles in the transport of divalent or trivalent cations in rice (<em>Oryza sativa</em> L.). This study explored a possible involvement of OsNramp4 in Cd2+ transport in rice. We employed the CRISPR/cas9 technique to obtain rice <em>OsNramp4</em> knockout lines. Using NMT (Non-invasive Micro-test Technology) system for real-time measurement, it was found that the net Cd2+ flux was significantly lower than that of wild-type. The yeast strain expressing <em>OsNramp4</em> grew poorly under Cd2+ stress, and accumulated more Cd2+ than the control strain, which enhanced the sensitivity of yeast to Cd2+. OsNramp4 was membrane-localized and mainly expressed in roots, but after tillering, its expression shifted to the nodes and glumes. Furthermore, the loss-of-function <em>OsNramp4</em> mutation lowered the root cell sap Cd2+ content, resulting in a significant decrease in Cd2+ content in shoot and Cd2+ accumulation in grains. We characterized the OsNramp4, an Al3+ transporter with the ability to alter the cellular distribution of Cd2+ in rice and to reduce Cd2+ content in the grain. It is regarded as an ideal model cell system in plants to study polar cell growth and cell polarity. The rapid pollen tube growth is empowered by exocytosis of large amounts of secretion vesicles that contain proteins, lipids and cell wall materials to the apex. Meanwhile, active retrograde endocytosis recycles excessively secreted vesicles for multiple rounds of exocytosis. Meanwhile, it also mediates uptake of extracellular molecules for signal transduction or vacuolar degradation. The dome morphogenesis and fast expansion of the pollen tube tip demand dynamic coordination and balance of antagonistic exocytosis and endocytosis. Nevertheless, the origins, molecular identities and their underlying molecular machineries of the apical vesicles still remain largely uncovered. In this review, we briefly summarize recent progresses in our understanding of the origins and identities of apical vesicles in the pollen tube tip. Due to the abuse of antibiotics, drug resistance among pathogenic bacteria and the deterioration of the water environment continue to occur. Therefore, it is essential to find probiotics that can effectively resist the erosion of pathogenic microorganisms, enhance fish's immunity, and promote fish's growth as an alternative to antibiotics.</p><p>In this experiment, two strains of probiotic bacteria antagonistic to <em>Flavobacterium columnar</em> were isolated from pond water samples. They were identified as <em>Bacillus amyloliquefaciens</em> and <em>Acinetobacter johnsonii</em> by 16S rRNA gene sequence analysis. To further analyze the physiological function of the two probiotics, we added <em>Bacillus amyloliquefaciens</em>, <em>Acinetobacter johnsonii</em> and two strain mixtures to the base feed and fed <em>crucian carp</em> separately. The results found that after 40 days of feeding, the growth performance of wild <em>crucian carp</em> and total protein(TP), globulin(GLO), albumin(ALB) and total bile acid(TBA) had no significant effect. However, <em>Bacillus amyloliquefaciens</em> could significantly reduce total bilirubin(TBiL), direct bilirubin(DBiL), indirect bilirubin(IBiL) and hypersensitive C-reactive protein(hs-CRP) in the serum of <em>crucian carp</em>. The addition of <em>Acinetobacter johnsonii</em> significantly reduced the content of indirect bilirubin(IBiL), acetonitrile/glutamate(A/G) and hs-CRP in the serum of wild <em>crucian carp</em>, which could effectively increase the range of superoxide dismutase(SOD) and reduce the production of malondialdehyde(MDA). After adding the mixed bacteria, Immunoglobulin M(IgM) in the serum of wild <em>crucian carp</em> increased significantly. In addition, the detection of intraperitoneal injection at 24 h, 48 h and 96 h was found to have no significant effect on the serum biochemical indicators of wild <em>crucian carp</em>.</p><p>The above results show that adding <em>Acinetobacter johnsonii</em> can enhance <em>crucian carp</em>'s anti-inflammatory, immunity and antioxidant properties. Therefore, the two bacteria can be used as potential probiotics for wild <em>crucian carp</em>. While, there are few reports on the study of gynogenesis between trans order species in freshwater fish. Even the meat quality comparison between the gynogenetic offspring obtained above and their parents is rarely studied. In order to understand whether the meat quality of naturally gynogenetic blunt snout bream (GBSB) is better than that of its parents (blunt snout bream♀ and Chinese perch♂), we analyzed the texture, composition, antioxidant capacity and nutrients of natural gynogenesis blunt snout bream and its parent muscle. In the muscle of texture characteristics of hardness, springiness, gumminess and chewiness, natural GBSB were better than those of the normal BSB. Muscle fiber size of GBSB is between the maternal and the paternal fish. Compared with the maternal parent, the natural GBSB had higher antioxidant enzyme activities (CAT, SOD and GSH) and higher mineral element (Na, Mg, Ca, P and Zn) contents.
