Sphingosine 1-phosphate (S1P) is one of the most studied lysophospholipid having wide role in various pathophysiological and cellular events, via signalling through five distinct GPCR subtypes, S1P receptors 1–5 (S1PR1-S1PR5).The importance of S1P pathway in drug targets have gained attention significantly after the approval of three S1PR modulators, fngolimod, siponimod and ozanimod, as medicines for patients with multiple sclerosis (MS). S1P, a signal transmitter, is converted to S1P by phosphorylation of Sphingosine kinase (SK1 or SK2) enzymes. S1P acts as an extracellular and intracellular ligand for receptors, S1PR1–5. The SK1/S1P axis and the connected S1PR1–5 plays important roles in many cellular events involving cell signalling, the cell survival/apoptosis, the production of pro-inflammatory cytokine responses, maintaining vascular integrity etc. Further sphingolipids, glycosphingolipids and ceramides acts as receptor/co-receptor for viral entry & downstream signalling, formation of lipid raft-mediated microdomain, viral induced apoptosis. S1P receptor modulator fingolimod shows antiviral activity against human immunodeficiency virus. Currently, the clinical trials of ozanimod (a sphingosine receptor modulator), and opaganib (a SK2inhibitor) are being conducted for treatment of COVID-19. In this review we explored the different roles of sphingolipids, S1P, S1PR, SK in viral infections including a special focus on SARS-CoV2. The World Health Organization estimates that there were roughly 228 million cases of malaria in 2018, which resulted in 405,000 tragic deaths. There was an approximate 20 % mortality rate among children diagnosed with CM who were admitted to the hospital. 67 % of the victims were children less than five years old. Patients who survive CM may experience lifetime post-CM complications and a higher risk of childhood neurodisability. The present review discuss about cellular mechanisms and immunological responses causing cerebral malaria. The breakdown of tight junctions and adherence junctions between endothelial cells (ECs) is the cause of an increase in the permeability of the blood brain barrier (BBB). It is obvious that this barrier can be breached by two different mechanisms: a) the apoptosis of ECs, and b) a loosening of the tight junctions between cells. T cells have been shown to have an important role in the development of cerebral malaria in an experimental mouse model (ECM). EphA2 receptors has been identified as contributing to many neurological illnesses and plays an important role in CM, associated with an impairment of the BBB. EphA2 is a key target protein that promotes EC apoptosis by targeting ephrin A ligand-expressing CD8+ T cell adhesion. Prominent role of the EphA2 receptors in CM has never been highlighted so far. Despite an intensive research, there is still a lack of successful malaria vaccines. The most advanced vaccines created so far, RTS,S/AS01 and R21/Matrix-M, focus on combating the early stages of the P. falciparum parasite. This review also discuss about the progress made for developing an effective vaccine for cerebral malaria.

","PeriodicalId":73214,"journal":{"name":"Health sciences review (Oxford, England)","volume":"11 ","pages":"Article 100175"},"PeriodicalIF":0.0,"publicationDate":"2024-04-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.sciencedirect.com/science/article/pii/S277263202400028X/pdfft?md5=e829e31fb49c0e933eac97008172fe6b&pid=1-s2.0-S277263202400028X-main.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140551074","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Recent years have established the significance of the gastrointestinal microbiome in the study of biological microorganisms. Diverse techniques for cultural extraction and analysis make it difficult to cultivate intestinal gut bacteria species. To determine the causative microorganism, it is necessary to investigate microbes and bacteria. Novel and efficacious remedies for illnesses are required. Bioinformatics and next-generation sequencing can assist in the analysis of enormous quantities of sequenced data for bacterial investigations, thereby expanding sequencing. However, advancements in sequencing technology have broadened the scope of bioinformatics identification and analysis. With next-generation sequencing techniques such as metagenomic, 16S rRNA, and meta-transcriptomic sequencing, experimental data regarding the immunological response of the intestinal gut microbiome to genetic manipulation seems to be helpful in identifying the diseases. Thus yet, the primary etiology of these ailments has not been determined. Regretfully, no medication can prevent the onset of these illnesses. The primary risk factor for these illnesses developing is age. Recently, there has been an increase in interest among researchers to investigate the potential therapeutic benefits of herbal remedies for neurodegenerative diseases. Flavonoids, tannins, terpenoids, alkaloids, and phenols are examples of secondary metabolites that have been used to prevent adverse effects and delay disease progression. Baicalein (5,6,7-trihydroxyflavone), a flavonoid found in the traditional Chinese herb Huangqin (Scutellaria Baicalensis George), helps block the age-associated damage pathways linked to neurological diseases. This review offers a plausible argument in favour of baicalein's beneficial effect in models of AD, PD, HD, and epilepsy. Baicalein reduces amyloid-beta and Tau oligomers, oxidative stress, and acetylcholinesterase activity in AD by binding to the enzyme's active site. Additionally, Baicalein reduces mitochondrial dysfunction against N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced neuro-inflammation in PD via inhibiting the NLRP3/caspase-1/gasdermin-D pathway. In HD, Baicalein was used to test its possible neuroprotective ability via N-methyl-D-aspartate receptor antagonism against quinolinic acid-induced neurotoxicity. Moreover, Baicalein reduces inflammation in epilepsy by controlling the insulin-like growth factor 1 receptor in microglia to prevent pilocarpine-induced epilepsy. The protective effect of Baicalein provides the potential future therapeutic approach against various neurological disorders.

","PeriodicalId":73214,"journal":{"name":"Health sciences review (Oxford, England)","volume":"11 ","pages":"Article 100172"},"PeriodicalIF":0.0,"publicationDate":"2024-04-03","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.sciencedirect.com/science/article/pii/S2772632024000254/pdfft?md5=d261778a472fcc729aaf20654136ebde&pid=1-s2.0-S2772632024000254-main.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140618250","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Eleven studies incorporating perspectives of 305 MTP were included and analysed using thematic synthesis. Physical pain, psychological difficulties, vulnerability and powerlessness influence MTP healthcare experiences. MTP described the positive and negative experiences of staff interactions and the integral role of communication on their healthcare experiences. A need for follow-up care, rehabilitation and to acknowledge the barriers to regaining independence and normality, was also reported. Varying needs related to psychological support, information provision, rehabilitation and follow-up care, impacted on MTP healthcare experiences and their expectations from services.
