{"title":"THE STUDY OF THE RELATIONSHIP BETWEEN THE ACTIVITY OF EPIPHYSIS AND GONADS IN MALE RATS IN DIFFERENT SEASONS.","authors":"V Hnatiuk, N Kononenko","doi":"10.15407/fz62.06.095","DOIUrl":"https://doi.org/10.15407/fz62.06.095","url":null,"abstract":"<p><p>In male rats of different ages, we studied the functional activity of the pineal gland and the gonads during different seasons by determining the levels of melatonin and testosterone in the blood serum. It was found that the highest levels of melatonin observed in male rats in summer and winter, and the lowest - in autumn. The lowest level of melatonin has been established in the group of rats aged 9 months - 127.28+5.11 pmol/l that was 22% lower than the level of melatonin in 3 months old rats (P!0,05), and 20% below the level of melatonin in rats of 15 months old (Ps0,05). The highest level determined in rats aged 3 months (corresponds to the age of man -14 years old), the lowest - in rats 20 months-(corresponds to the age 55-56 years old). At the same time, the low level of melatonin was defined in rats aged 9 months in the autumn (corresponds to the age of man 29-30 years). In the study of testosterone levels in different seasons it was found that the highest level in all age groups presents in autumn, the lowest - in winter. When comparing the levels of testosterone in different age groups, it was determined that the fall of the highest level of testosterone present in the blood of males age 9 months and 15 months, which corresponds to the age of 29-30 and 43-44 of human years. Significant difference between the levels of testosterone in the groups that correspond to the reproductive age - rats of age 3, 9 and 15 months, are present only in the autumn; spring and summer - significant differences between the levels of testosterone absent. The degree of correlation differed in rats of different ages in different seasons: the highest correlation between the levels of melatonin and testosterone was determined in rats aged 9 months in autumn. The got results testify that maintenance of MPO in plasma of blood of patients with acute infarct of myocardium depends on the functional state of neutrophils, which, in its turn, is related to the clinical features of patients. Low maintenance is associated exceptionally with low activity of MPO of neutrophils, and also other oxygen radicals. For the study of hypofunction of the pineal gland, animals were maintained under constant light stimulation during 7 days. It has been established that the hypofunction epiphysis point out disturbances of the phasic structure of the ion-regulating function in relation to chronograms of the intact group of rats. In terms of physiological pinealectomy observed in- crease in the excretion of sodium ions (6,11±0,81 mmol/h) and the concentration of this cation in the blood plasma remained elevated around the clock. {"title":"Experimental therapy of graft-versus-host disease by mesenchymal stromal cells grown on oxide nanocoatings.","authors":"A N Goltsev, I V Rassokha, T G Dubrava, L V Ostankova, M V Ostankov, V I Safonov, A V Zykova","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>Immune aggression to transplanted allogeneic bone marrow, i.e. the graft-versus-host disease (GVHD), could be decreased by the suppression of effector and/or activation of T- regulatory cells (Treg). This task could be solved by co-transplantaiton of allogeneic bone marrow and mesenchymal stromal cells (MSCs). This study demonstrated the elevated immune modulating activity of MSCs by their culturing in vitro on Al₂0₃ oxide nanocoatings. Introduction of the cells to the animals with GVHD resulted in an increased content of Treg in the spleen of bone marrow recipients, reduced severity of the pathology, and higher survival of animals. According to the data, used deep diaphragmatic breathing in mode of biofeedback harmonizes breathing and blood circulation which positively affects the body's adaptive potential. However, developers present the portable instruments of biofeedback primarily as a means of psychological relaxation. The aim of this study is to determine whether such devices are able to significantly affect heart rate variability and how long this effect lasts. There were 30 males aged 18 to 20 years that participated in this study. They were randomly divided into three equal groups with up to 10 people in each of them. Members of the first group were doing breathing exercises using a portable biofeedback StressEraser device, the second group used Em-Wave2, and the third one used hardware and software system MyCalmBeat. Each session lasted from 10 to 15 minutes. There were 10 sessions held daily in the morning. HRV was recorded by remote heart rate monitor Polar RS800CX and software Polar ProTrainer5. 5-minutes' stationary segments of the curve chest electrocardiogram removal were used in order to calculate the HRV indexes. It was found that 15 minutes deep breathing in mode of biofeedback significantly affects the heart rate variability of healthy young people. In particular, it increases the overall heart rate variability according to the statistical and spectral analysis. It was established that in all 3 groups of experiment participants the spectral curve low-range \"Low frequency\" (LF) suffered the major shifts. These shifts reflect the activity level of the sympathetic autonomic nervous system (ANS) as well as vascular-motor center according to the conventional interpretation. It was found that all three groups suffered a specific weight decrease of Very Low Frequency (VLF)% respectively 11.8±2.2%;(p <0,01); 6.6 ± 2.8%; (p <0,02) and 6.8 ± 2.4% (p <0,05). This means that activity redistribution between different regulatory components of heart rate occurred in favour of the peripheral ANS parts. Heart rate variability changes of this kind remained valid for at least one month after the experiment. However in the absence of blood circulation in vitro, the tissue oxygenation strongly depends on the experimental conditions. It is well established that both hypoxia as well as hyperoxia can modulate the neuronal network activity. Thereby changes in tissue oxygen level during experiment may affect the final result. In the present study we investigated the effect of oxygenation on seizure susceptibility in the hippocampal slice preparation using 4-aminopyridine (4-AP) model of ictogenesis in inmature rats. We found that changing the medium perfusion rate in the range of 1-5 ml/min greatly affects the tissue oxygenation, amplitude and frequency of 4-AP-induced synchronous neuronal activity. The decrease in the flow rate as well as substitution of the oxygen in the extracellular medium with nitrogen causes a strong reduction of 4-AP-induced synchronous neuronal discharges. Our results demonstrate a significant linear correlation between the power of 4-AP-induced neuronal activity and the oxygen level in slice tissue. Also we demonstrated that the presence of medium flow is a necessary condition to support the constant level of the slice oxygenation. The parameters of blood\u0000oxygen transport (р50, рСО2, рО2, рН, HCO3\u0000-, ABE and ect.),\u0000lipid peroxidation products (conjugated dienes, Schiff bases)\u0000and blood hepatic markers (ALT, AST) were detected. It’s\u0000found that HIR leads to decline in hemoglobin oxygen affinity,\u0000activation of lipid peroxidation processes and elevation of ALT\u0000and AST activities in the 1st group. Hypoxic preconditioning\u0000(2nd group) markedly increased hemoglobin oxygen affinity,\u0000reduced the lipid peroxidation processes and ALT and AST\u0000activities in the blood during HIR.
