The effect of the most common endogenous and exogenous antioxidants in the biochemical processes in muscle fatigue was analyzed. It has been found that when investigating ad/bcm and dg/cgm alleles of the blood group system D in the stallions, there was the physiological tendency of the decrease in the semen activity on average less than 5 points; at the presence of ad/cgm, ad/d, ad/ de, ad/dk, bcm/d, bcm/de, bcm/dg bcm/dk cegm/cgm, cegm/d, cegm/dg, cegm/dk cgm/ceg, cgm/cgm, cgm/dg, cgm/dk de/ cgm, de/dk dg/di, dk/d, dk/de, dk/dk alleles the spermogram of the average quality with the activity of the semen on average from 5 to 7 points was obtained; at the presence of bcm/cgm, dg/dk, de/d, egm/d, cgm/de alleles high parameters of semen activity, more than 7 points, were received. The tendency of the increase in the semen concentration was revealed in the examined stallions having a/- allele of the blood group C. The possible decrease in the semen concentration in the stallions having a/-allele of the blood group K was shown. The alleles of the blood group system D had the highest statistical impact on the volume of the ejaculate, the activity, concentration and the number of pathological forms of semen, the correlation coefficient was 0,36; 0,31; 0, 43 and 0,27, respectively. It was established that neonatal administration of MSG to rats leads to increasing levels of the interleukin (IL)-1β and IL-12, and to decreasing ofthe IL-4, IL-10 and tumor growthfactor (TGF)-β levels in the bloodserum. After administration of the B. animalis VKL - B. animalis VKB - L. casei IMV B-7280 composition to obese rats the level of the IL-lP in blood serum wasn't differ from that in the obese rats, that didn't receive of the probiotic bacteria. But there was no statistically signifcant difference comparing with intact rats. The level of the IL-12B p4O in blood serum was decreased under influence of the B. animalis VKL - B. animalis VKB - L. casei IMV B-7280 composition (18.9%, p < 0.05) and B. animalis VKL (10.5%, p < 0.05) compared with obese rats, not receiving probiotic bacteria, but remained higher than in intact animals. After administration to obese rats ofthe B. animalis VKL - B. animalis VKB - L. casei IMV B-7280 composition the levels ofthe IL-4, IL- 10 and TGF-β increased in blood serum comparing with obese rats, not receiving probiotic bacteria. The level of the IL-10 also increased under influence of the B. animalis VKB, and IL-4 - under influence of the L. casei IMVB-7280. Our results suggest that these probiotic bacteria and probiotic composition are able to down-regulation the inflammation in rats with MSG-induced obesity but the strongest anti-inflammatory effects have probiotic composition. {"title":"STABILOMETRIC VALUES OF HUMANS UNDER THE CONDITIONS OF FORWARD AND BACKWARD BENT POSITIONS.","authors":"V V Garkavenko,&nbsp;E V Kolosova,&nbsp;V D Maksimov","doi":"10.15407/fz62.01.062","DOIUrl":"https://doi.org/10.15407/fz62.01.062","url":null,"abstract":"<p><p>Using stabilometry study the center of pressure (COP) displacement over the horizontal surface and power spectrum characteristics of postural sway in the sagittal and frontal planes (Y and X- oscillations, respectively) during an upright stance (US) and under the conditions of body bents forward and backward (BF and BB, respectively) were investigated in a group of 15 volunteers. The tests were performed under the conditions of opened and closed eyes. It was demonstrated that the power spectrum of sway depended on posture, it was greater under the conditions of body bents than during the upright stance. In all three positions blocking of optical input resulted in an increase in power spectrum as well of Y- and X-oscillations in a number of frequency bands of body sway. A significant dependence on the visual control for the mean value of COP under the conditions of bents backward was also found. Data on changes in the lateral oscillations under the conditions of body bents in the sagittal plane showed the essential role of ankle proprioceptors in the genesis of the lateral oscillations at keeping balance during standing. The study is performed on 65 white outbred male rats. TBI is applied with one blow on animal’s cranial vaults with blow energy of 0.52 J. The rate of mortality within the first 5 days after the injury is 87%. Experimental animals have got severe closed TBI. Blood contents of circulating immune complexes, C-reactive protein, ceruloplasmin, proinflammatory - interleukins(IL-1b, IL-6, IL-8 and tumor necrosis factor a -TNF-a) are investigated. The circulating immune complexes levels are increased 3.1 times in 24 hours and 4.4 times on the 5th days of trauma reflecting the progressive accumulation of metabolites and toxins in brain tissue and in the blood of injured animals. Blood levels of C-reactive protein are markedly increased in all periods of observation exceeding the control levels 3.5 times in 3 hours and 21.3 times after 5th day of trauma. Thus the study results suggest that the acute phase of systemic inflammation sets at the end of the 1st day after the trauma and it progresses in the course of traumatic brain disease. Blood contents of IL-1b increases continuously: 4.7 times in 3 hours; 7.6 times in 24 hours; and 17.4 times on the 5th day after trauma. The other interleukins levels are also increased but to a lesser extent. TRPV4 selective agonist, GSK1016790A, in the presence of the agonist of α-adrenoceptors phenylephrine (PhE) evoked biphasic contractile reaction with initial relaxation (63,5% ± 7,1) followed by significant vasoconstriction (142% ± 17,9). GSK1016790A evoked similar effects in PASM rings with and without endothelium, indicating that its main site of action was TRPV4 expressed in smooth muscle cells. TRPV4 selective blocker, HC-067047, completely inhibited the effects of GSK1016790A confirming the specific role of TRPV4 in these vascular responses. Application of Ca2+-free external solution reduced the relaxation phase and completely abolished the sustained contractile response to GSK1016790A (from 43,9 % to 0,3 %). The biphasic reaction could be explained as an initial calcium store depletion by PhE and further calciuminduced calcium release activated by TRPV4 that causes BKCa activation, membrane hyperpolarisation and vasorelaxation, followed by Ca2+ entry via TRPV4 and contraction. Rats were either kept on the ad libitum diet or on a calorie restricted diet (70-80% reduction in total calories) without reduction in essential nutrients. It has been determined that calorie restricted diet decreased the ceramide, sphingomyelin, cardiolipin and phosphatidic acid levels in the all investigated tissues of the rats. At the same time, calorie restriction diet prevented the age-induced ceramide and phosphatidic acid accumulation, ceramide/sphingomyelin ratio elevation, and sphingomyelin and cardiolipin content decrease in the tissues of 24-month-old rats. In addition, tissue specificity of calorierestricted diet effects has been determined. The Elevated levels of cardiolipin and phosphatidic acid were determined in the heart and liver of 24 months-old rats under calorie-restricted diet, in comparison to control animals, whereas in the brain the caloric restriction diet had the opposite effects. Intraportal administration of\u0000the test compound at doses of 2,5; 5 and 10 mg/kg resulted in\u0000a significant increase in the volume of secreted bile by 20,9,\u000031,2 and 20,4%, respectively, as compared with the control. Using the method of thin layer chromatography it was established\u0000the mild stimulating effect of Corvitin on the processes\u0000of bile acids conjugation with taurine and glycine, especially\u0000when administered at a dose of 5 mg/kg. This flavonoid did not\u0000affect the concentration of glycocholic acid, however increased\u0000the content of glycochenodeoxycholic and glycodeoxycholic\u0000acids in the mixture between 15 to 35,1%. Regarding free bile\u0000acids, the concentration of cholic acid, chenodeoxycholic and\u0000deoxycholic acids in the mixture was increased significantly\u0000relative to control only after Corvitin application at dose 10 mg/\u0000kg. In the first case – from 17,9 to 29,8%, in the second – from\u000025 to 65,4%. At the dose of 5 mg/kg, Corvitin significantly\u0000increased the ratio of bile cholates conjugation (maximum by\u000023,2%), whereas 10 mg/kg of the drug decreased this index\u0000by 27,0%. After administration of Corvitin, the hydroxylation\u0000ratio in all experimental groups differed little from the control:\u0000at the dose of 5 and 10 mg/kg this parameter decreased by\u000014%. It is shown that the influence of a sustained training accompanied by the development of exercise-induced hypoxia on the structural readjustments of tissues of muscles, lungs, and myocardium can be conditionally divided into 2 groups: with destructive and compensatoryadaptive features. The changes with destructive character include, firstly, those of the ultrastructure of biological bar- riers such as, in particular, hyperhydration of barriers on the whole and their separate layers, which deteriorates the conditions of oxygen diffusion; second, the destructive changes in mitochondria (it increased the number of damaged lung organelles by 4.1 times, in the heart - at 4.5-5.5 times depending on subpopulations and in muscle - by 3.5-12.2 times also depending on the subpopulation of mitochondria), which are accompanied by a decrease in the energy potential of the mitochondrial apparatus, are observed. To the changes with compensatory-adaptive character, we refer an increase in the number of functioning capillaries (by 80% in the gastrocnemius muscle and by 60% in the myocardium), which prevents the development of secondary tissue hypoxia; intensification of pinocytosis in endotheliocytes; activation of mitochondrial morphogenesis, which was accompanied by an increase of the number of organelles at gastrocnemius muscle by 65%, in the lungs - in 4 times and in heart by 60-80% depending on the mitochondrial subpopulations; and appearance of young mitochondria and mitochondria with moderate degree of swelling, which favors the growth of the energy power of the mitochondrial apparatus of cells.</p>","PeriodicalId":73031,"journal":{"name":"Fiziolohichnyi zhurnal (Kiev, Ukraine : 1994)","volume":"62 6","pages":"72-80"},"PeriodicalIF":0.0,"publicationDate":"2016-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40437377","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

To evaluate diastolic function of the heart, we used pressure-volume (PV) conductance catheter system (Millar Instruments, USA). It was shown that H2S levels in. the isolated mitochondria and whole heart homogenates obtained from old age rats were significantly lower comparing with adult animals. The markers of combined oxidative and nitrosative stress (the rate of 0 ₂⁰ H generation, pools of H₂0₂, diene conjugates, malondialdehyde, uric acid, the activity of iNOS, nitrate reductase, and NO pools) were increased in the old hearts in line with cNOS uncoupling. Such changes in NOS coupling resulted in the loss of diastolic relaxation (decrease of the rate of relaxation of the left ventricle (dp/dtmin) by 33%, 3-times increase of the end-diastolic pressure, 1.5-time increase of the time constant of left ventricular relaxation (Tau g) and 2-time increase of the end-diastolic stiffness). It has been found that NaHS inhibits oxidative and nitrosative stress, restores cNOS coupling and constitutive de novo synthesis of nitric oxide (NO), which promotes an improvement of the diastolic function (increase of the dp/dtmin by 20% and decrease of Tau g by 13%) .
