The aim of the current study was to investigate oxidative stress and changes in CP in cattle with traumatic reticuloperitonitis (TRP). The study was divided into two groups, experimental (TRP) and healthy control group, with 10 animals in each group. Total leucocyte count, neutrophil and lymphocyte counts were higher in the TRP group compared to the control group and this increase was statistically significant (P < 0.001). The level of malondialdehyde (MDA) in TRP group was statistically significantly higher than the control group (P < 0.001). The level of glutathione (GSH) in the TRP group was statistically significantly lower than in the control group (P < 0.001). Serum Amyloid A (SAA) and CP values were higher in the TRP group and this difference was statistically significant (P < 0.001). It was concluded as a result of ROC analysis that CP, which has similar values with SAA, can be used diagnostically to confirm the inflammatory status in cattle with TRP.</p>","PeriodicalId":7247,"journal":{"name":"Acta veterinaria Hungarica","volume":" ","pages":"193-201"},"PeriodicalIF":0.7,"publicationDate":"2023-12-11","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"138794866","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Pairs of testes from German Mutton Merino (n = 7) and Hungarian Black Racka (n = 7) rams were collected at a slaughterhouse, transported to the laboratory and stored overnight (4-5 °C) before processing. Post mortem ram epididymal spermatozoa (REPS) were obtained from the cauda epididymidis by slice or incision methods. Fresh samples were extended to 200 × 106/mL cell concentration, filled into mini straws and equilibrated at 5 °C for 2 h. Freezing was performed manually in a Styrofoam box. The fresh and post-thaw total motility, progressive motility and kinematic parameters of REPS were assessed using the CASA technique. The collection method did not affect significantly the fresh and post-thaw motility and kinematic parameters. Merino had higher (P < 0.05) testicular weight. Racka had significantly better fresh and post-thaw linear movement but had statistically the same (P > 0.05) cryotolerance as Merino. In conclusion, both collection methods were found suitable for REPS retrieval. The REPS from Racka exhibited better linear movement values than those from the Merino breed. The cryotolerance of REPS of both breeds was comparable.</p>","PeriodicalId":7247,"journal":{"name":"Acta veterinaria Hungarica","volume":" ","pages":"210-218"},"PeriodicalIF":0.7,"publicationDate":"2023-11-24","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"138298107","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

One hundred and sixty-one cows were fitted with a Ruminact© HR-Tag approximately 5 days before calving. Drenching and control groups were established based on calving dates. Animals in the drenched group were treated three times (Day 1/day of calving/, Day 2, and Day 3 postpartum) using a feed additive containing calcium propionate, magnesium sulphate, yeast, potassium chloride and sodium chloride mixed in approximately 25 L of lukewarm tap water. Blood samples were collected on Days 1, 2, 3, 7 and 12. Cows with below the average RT were categorised as \"low rumination\" and those above it as \"high rumination\" animals. Drenching decreased the plasma concentrations of total protein, urea and creatinine and increased the levels of alanine aminotransferase (ALT), gamma-glutamyl transferase (GGT) and chloride. Low rumination time prepartum resulted in higher concentrations of beta-hydroxybutyrate, total protein and activities of alkaline phosphatase and GGT, while it decreased the activity of ALT and the concentrations of calcium, magnesium, sodium and potassium. The day of lactation had an effect on all parameters except for potassium.</p>","PeriodicalId":7247,"journal":{"name":"Acta veterinaria Hungarica","volume":" ","pages":"147-158"},"PeriodicalIF":0.7,"publicationDate":"2023-11-24","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"138298105","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Medical records of 24 female Holstein cows that underwent dehorning surgery were used in this study. In four groups, the sedation of animals was carried out by one of the four intravenous treatments: 0.1 mg kg-1 xylazine (Xyl group, n = 6), 5.0 μg kg-1 medetomidine (5.0 Med group, n = 6), 10.0 μg kg-1 medetomidine (10.0 Med group, n = 6) or 20.0 μg kg-1 medetomidine (20.0 Med group, n = 6). The clinical sedation score (CSS) and heart rate (HR) were recorded. The CSSs after intravenous administration of each α2-adrenergic receptor agonist increased rapidly and peaked at 12.5 (10.0-16.0) at t = 20 min in the Xyl group, 11.5 (10.0-15.0) at t = 10 min in the 5.0 Med group, 16.0 (14.0-16.0) at t = 20 min in the 10.0 Med group and 16.0 (14.0 - 16.0) at t = 20 min in the 20.0 Med group. A similar degree of bradycardia was observed after every sedative treatment. This study determined reference ranges for bovine serum chemistry and complete blood cell count (CBC) according to Holstein milking-cow age. Seventy-two healthy Holstein calves and cows (<1 week to milking age) were grouped: 1 (n = 7, <1 week), 2 (n = 10, 1 month), 3 (n = 13, 3 months), 4 (n = 13, 6 months), 5 (n = 10, 1 year, nulliparous), and 6 (n = 19, milking cows, parous). Fresh blood samples were obtained from the jugular vein between 10:00 and 12:00 AM in the winter; serum chemistry and haematologic profiles were assessed. Serum chemistry and CBC differed significantly by age. Age-related differences were observed for albumin, alkaline phosphatase, creatinine phosphokinase, creatinine, gamma-glutamyl transpeptidase, glucose, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, magnesium, phosphorus, calcium, total bilirubin, total cholesterol, total protein, triglyceride, blood-urea nitrogen, non-esterified fatty acid, and beta-hydroxybutyric acid levels. Age differences in creatinine and C-reactive protein were not noticeable. Among CBC parameters, age-related differences were observed for white-blood-cell, lymphocyte, red-blood-cell, and platelet counts; hemoglobin level; haematocrit; mean corpuscular volume, mean corpuscular hemoglobin, and mean corpuscular-hemoglobin concentration. A total of 1,840 serum samples (595 goats, 1,245 sheep) were collected from 92 villages in the region and examined by indirect ELISA. Thirty-nine goat and 304 sheep sera were found positive for small ruminant lentivirus antibodies. The prevalence was 24.42% in sheep and 6.55% in goats. An average of 18.64% was found in small ruminants in the Aegean region. The prevalence of small ruminant lentiviruses was found to be higher in Pirlak and Pirlak hybrid sheep and in Saanen and Saanen hybrid goats ( P < 0.05). The prevalence was found to increase with age, as it was higher in animals older than 48 months than in the age groups of 0–24 or 25–48 months ( P < 0.05). Moreover, the small ruminant lentivirus prevalence was higher in sheep than in goats ( P < 0.0001). With the epizootiological approach, this study is expected to contribute to the control and prevention of small ruminant lentivirus infections.","PeriodicalId":7247,"journal":{"name":"Acta veterinaria Hungarica","volume":"11 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135943986","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Due to the varying responses to different treatments, novel therapeutic approaches are required. Sodium butyrate (NaBu) is the first histone deacetylase inhibitor and exhibits anti-cancer activity in different types of human cancers. However, no study is available in the literature evaluating the effects of NaBu on CMT cells. Therefore, for the first time, we aimed to investigate the in vitro efficacy of NaBu on three subtypes of CMT cells. Primary cells were isolated from neoplastic tissues of three dogs with CMT. Cell viability, apoptotic percentage and changes in the cell morphology, apoptosis-associated gene and protein expression levels were evaluated. Our findings showed that NaBu decreased the growth of the cells and increased the percentage of apoptosis in three subtypes of CMT cells. Additionally, NaBu treatment affected the expression levels of genes and proteins associated with apoptosis, especially in S cells. Our preliminary results, for the first time, revealed the efficacy of NaBu in CMT cells and the importance of the histological subtype in NaBu response. However, the molecular classification of CMT and the underlying mechanisms of the different NaBu responses should be further investigated.","PeriodicalId":7247,"journal":{"name":"Acta veterinaria Hungarica","volume":"18 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135943985","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

From 260 faecal samples, 236 E. coli isolates were obtained, of which 30 (12.7%) proved to be non-O157:H7 STEC. The presence of STEC-serogroup virulence genes stx1, stx2 , eaeA and ehxA was detected by PCR. Further virulence genes and other genes used for phylogenetic grouping were also examined among the STEC isolates. The most dominant serogroup was O145, followed by O103 and O45. Shiga toxin gene ( stx1 ) was found in 70% of the STEC strains, stx2 and ehxA were detected in 17 isolates (56.7%). Other virulence genes, including fimH , traT, sfa/focDE , iutA , cdt3 , hly, fyuA and ibeA, were detected in 100%, 60%, 46.7%, 46.7%, 36.7%, 10%, 6.7% and 3.3%, of the strains, respectively. More than 70% of the isolates were susceptible to most of the tested antibiotics. We concluded that the presence of STEC O145, O103 and O45 (12.7%), together with the earlier described STEC O157:H7 (4.2%) in cattle, intended for slaughter, may represent a threat to public health. Infection prevalence, hatching rate, hatching dynamics of fish eggs were quantified, and the study was complemented with histopathology and phylogenetic analyses. We experimentally detected intraspecific differences in the pathogenicity of S. parasitica on rainbow trout eggs. The isolate from rainbow trout eggs was the most virulent to eggs of the same host, whereas isolates from carp skin and fry did not cause as much damage to the eggs. Comparing the outcome of two experimental settings, we confirmed that invasion of fish eggs is more effective by hyphae growth than by the actively moving zoospores. In addition, our findings highlighted that S. parasitica isolates with 100% identical ITS DNA sequences, could differ significantly in virulence. These isolates can be clearly distinguished based on a 650-bp DNA fragment of the DNA-directed RNA polymerase II subunit ( RPB2 ) gene.","PeriodicalId":7247,"journal":{"name":"Acta veterinaria Hungarica","volume":"166 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-10-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135945125","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"农林科学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

After quality control, the average length of sRNA of camel milk exosomes was 18-24 nucleotides. A total of 2,659 miRNAs were identified, including 2,458 known, and 201 new miRNAs. Among the known miRNAs, miR-148a and let-7i had the highest expression levels. The results of gene ontology enrichment analysis indicated that the target genes of camel milk exosome miRNAs were involved in multicellular organismal, catabolic and other biological processes. They play role in the extracellular region, in the cytoskeleton and other cell components, in protein binding, but also have structural molecule activity and other molecular functions. According to the results of the Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis, the target genes of camel milk exosome miRNAs are involved in Alzheimer's disease, non-alcoholic fatty liver disease, Staphylococcus aureus infection and other pathological pathways.
