Deoxynivalenol (DON) produced by various Fusarium mould species can induce cytotoxicity and oxidative stress in the gastrointestinal tracts of humans and farm animals. The aim of this study was to investigate the effects of Que on DON-induced oxidative stress in a non-tumourigenic porcine IPEC-J2 cell line. Two experimental designs were used in our experiments as follows: (a) pretreatment with 20 µmol/L Que for 24 h followed by 1-h 1 µmol/L DON treatment and (b) simultaneous application of 20 µmol/L Que and 1 µmol/L DON for 1 h. Cell cytotoxicity, transepithelial electrical resistance (TER) of cell monolayers and extracellular/intracellular redox status were studied. It was found that DON significantly decreased TER and triggered oxidative stress, while Que pretreatments were beneficial in maintaining the integrity of the monolayers and alleviated oxidative stress. However, co-treatment with Que was unable to preserve the integrity and redox balance of the cells exposed to DON. We aimed to elucidate the underlying mechanism and investigated whether gintonin affects the concentration of the extracellular matrix remodelling factor matrix metalloproteinase-9 (MMP-9) in tears during rabbit corneal wound healing in vivo. Twelve eyes (six rabbits) were divided equally into three groups. All eyes underwent corneal de-epithelialisation. The control group received Tearin Free sodium hyaluronate 0.1%, the solcoseryl group received solcoseryl-120 concentrate, and the gintonin group received 2.5 mg gintonin in sodium hyaluronate 0.1%. All preparations were administered for 5 days and the concentration of MMP-9 was measured in tears via ELISA on days 0, 1, and 5. MMP-9 concentrations in all groups were increased at day 1 and reduced at day 5. Of note, we found a significant change over the time frame for the gintonin group (P < 0.05) but not for the control or solcoseryl groups (P > 0.05) Moreover, increased MMP-9 levels between days 0 and 1, and their reduction between days 1 and 5, were significant in the gintonin group compared to those in the other groups (P < 0.05); however, and once more, these changes were not significant between the control and solcoseryl groups (P > 0.05). However, there are no data on the prevalence of infection and the clinical forms or geographic distribution of the disease. The aim of this study was to determine the seroprevalence of Streptococcus equi subspecies equi in horses resident in Croatia, in order to estimate the geographic distribution of infection. The study included 291 horse sera from the eight counties where the majority of Croatian horses are kept. Sera were tested by indirect ELISA (iELISA) for the presence of serum antibodies against S. equi protein A (SEQ_2190) and protein C (SeM). Positive horses were detected in all counties. Overall seroprevalence was 16.5 per cent (48/291), ranging from 7.1 to 29.6 per cent. A positive association was observed between the population size of the horses in the counties and the seropositivity rates: the larger the population, the higher the seropositivity. The results of this study suggest that S. equi infection is widespread in Croatia. For that purpose, the turtles were anaesthetised with ketamine-medetomidine or propofol, and the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione S-transferase (GST) and the level of malondialdehyde (MDA) were determined by spectrophotometry. The possible genotoxic effects of the anaesthetic agents were estimated by comet assay. A total of 24 turtles were included in this study. The animals were divided into four groups according to the anaesthetic protocol and according to whether endoscopy would be performed. Significantly decreased activities of CAT were found only in the propofol group and in turtles undergoing coelioscopy. Both anaesthetic protocols induced significantly increased MDA levels, while no differences were observed after the intervention. A significant increase in GST activity was detected in turtles after both anaesthetic protocols, but after coelioscopy significant changes in GST activity were found only in the propofol group. However, no differences in SOD activity and no DNA damages were detected in either group. In our study we tested inhibitory activity against Paenibacillus larvae and the biofilm formation activity by various lactic acid bacteria isolated from honey bee guts or fresh pollen samples in the presence of different sugars added to the cultivation media. In addition, we tested the probiotic effect of a newly selected Apilactobacillus kunkeei V18 in an in situ experiment in bee colonies. We found antibacterial activity against P. larvae in four isolates. Biofilm formation activity of varying intensity was noted in six of the seven isolates in the presence of different sugars. The strongest biofilm formation (OD570 ≥ 1) was noted in A. kunkeei V18 in the presence of fructose; moreover, this isolate strongly inhibited the growth of P. larvae under laboratory conditions. A study was planned to evaluate the feasibility of this procedure in the management of abdominal cysts in sheep and goat. Adult ewes (n = 5) and one doe (n = 1) found to have abdominal cysts (one cyst/animal) on repeated survey ultrasonography (USG) were included in the study. The animals were restrained in standing position. A hypodermic needle (G-18) securely attached to a 10-mL Dispovan syringe was carefully passed under ultrasound guidance into the abdominal cyst in all these animals. Depending on the size of the cyst, 1.0-5.5 mL fluid was aspirated, and 0.5-2.0 mL of 20% hypertonic saline solution infused. The needle was thereafter kept in situ for 10 min. The maximum possible volume of the cyst content was reaspirated and the needle withdrawn. On day 7, sclerotherapy was repeated in five animals showing no appreciable reduction in cyst size by USG. USG was repeated on days 30 and 90. All the cysts except one responded to PAIR during this period. From this study it can be concluded that sclerotherapy using hypertonic saline (20%) is a minimally invasive, inexpensive, effective and safe interventional ultrasonographic technique for the treatment of abdominal cysts in sheep and goats. In this study we compared the effectiveness of lithium chloride to that of oxalic acid, a widely used miticide. The results of the present study confirm that lithium has superior efficacy to oxalic acid sublimation both as a main or a supplementary pre-wintering treatment at moderate infestation levels, restricted to certain pre-wintering conditions. A study was performed over a period of 5 months in Ireland with the aim to get an updated and more realistic prevalence of these retroviruses. A total of 183 EDTA-anticoagulated whole-blood samples were collected from cats distributed between 10 clinics. The samples were tested using both point-of-care enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR). Basic clinical data and vaccination history were also recorded for the sampled cats. The results of ELISA tests showed a prevalence of 10.4 and 3.3% for FIV and FeLV, respectively, and an apparent prevalence of 9.3% for FIV and 11.6% for FeLV with PCR. Phylogenetic analysis of the partial polymerase (pol) gene sequences obtained from 8 FIV-positive strains showed that all but one of the Irish strains belonged to FIV subtype A, and one to subtype B. {"title":"Experimental evaluation of cortical screw placement in lag fashion into the distal phalanx in standing horses.","authors":"Simon Izing,&nbsp;Dániel Béni,&nbsp;Szabina Molnár,&nbsp;Zoltán Bakos,&nbsp;Gábor Bodó","doi":"10.1556/004.2020.00054","DOIUrl":"https://doi.org/10.1556/004.2020.00054","url":null,"abstract":"<p><p>The objectives of this in vivo experimental study were to evaluate the feasibility of cortical screw insertion into the intact distal phalanx in standing sedated horses and to document potential postoperative complications. One cortical screw was randomly inserted in lag fashion into each distal phalanx in 9 horses. The second surgery on the contralateral limbs was performed 2-3 weeks after the first operation, when a 4.5-mm cortical screw was inserted in lag fashion into the distal phalanx of sedated horses following perineural analgesia. Following surgery, the drill hole was filled with an antibiotic-soaked swab, which was changed every 48 h. The horses were euthanised 8 weeks after the second surgery. The hooves were disarticulated and evaluated macroscopically and by computed tomography. The surgery time was 13.9 ± 4.8 min (mean ± SD). Pain scores and lameness gradually decreased after 7 days. Solar canal penetration (SCP) was detected in 10 out of the 18 distal phalanges (55.5%). In 7 out of the 10 penetrations intraoperative bleeding was obvious. No postoperative infection was observed. Screw insertion into the distal phalanx was easily and quickly accomplished in standing horses, but its advantages in horses with sagittal fractures should be investigated further.
