Udosen","doi":"10.4236/ajmb.2022.122007","DOIUrl":"https://doi.org/10.4236/ajmb.2022.122007","url":null,"abstract":"The goal of this study was to determine whether mutation of the Mn-binding site of wild-type recombinant Phlebia radiata manganese peroxidase 3 affected the pH-dependence kinetic parameters. pH range investigated was 2.5 – 12.0. The catalytic efficiency of the mutant enzymes at high and low pH in comparison to the wild-type was investigated using standard rPr-MnP3 protocol. Wild-type recombinant Phlebia radiata MnP3 enzyme showed optimal activity with Mn (II) as substrate at pH 5.0 and remained moderately active (approximately 40%) in the pH range of 6.0 - 9.0. The rPr-MnP3 mutants’ maximum activity ranged between 5.5 and 8.0. Wild-type and mutants rPr-MnP3 enzymes exhibited a similar pH profile with optimum pH of 3.0 for ABTS oxidation. Mutation has severely decreased the catalytic efficiency for Mn (II) oxidation at pH 5.0. The rPr-MnP3 enzymes showed enhanced affinity for Mn (II) at alkaline pH and a more alkaline range for catalysis than ever reported for any Manganese Peroxidase. This study reveals that at higher pH, rPr-MnP3 can function with alternative ligands in the Mn (II) site and does not have an absolutely obligate requirement for an all carboxylate ligand set. RTK plays a vital role in cellular communica-tion and transmission of signals to the adjacent cells and regulates different functions of the cell, such as cellular growth, differentiation, metabolism and motility. RTK s triggers growth factor receptors such as epidermal growth factor, insulin growth factor-1 receptor, platelet derived growth factor receptor, and fibro blast growth factor receptor and vascular endothelial growth factor receptor, thereby initiating and regulating cell growth and proliferation. MAPK/RAS and PI3/AKT pathways are the major pathways of RTK’s function. Dysregulation of these RTK’s and pathways often leads to many diseases such as Noonan Syndrome, Logius Syndrome, CFC syndrome and different types of cancer. Point mutation and over expression of receptors and mutations in Ras leads to 30% of human cancers. In this study the Expression of Nanobodies in E. coli WK6 cell periplasm was performed. The protein expression and purity was and analyzed by Affinity Chromatography, SDS PAGE and Western Blot. Upon elution with Imidazole, the concentrations observed using the OD280 nm of the eluted fractions EI, E2 and E3 were observed to be 0.42 μg/ml, 0.13 μg/ml and −0.46 μg/ml respectively. This gives an Antilog of 7.88 kDa which showed the calculated molecular size of our band. The SDS-PAGE gel reading was confirmed using Western blot analysis and illustrated as the specific binding of the mouse Anti-His antibody to the Histidine tag of the Nanobody. The Nanobody protein expression was then analyzed further with western blotting showed a strong signal at the region corresponding to the 15 kDa marker indicating presence of the Nanobody gene. Isolation was made on selective media (Tetrazolium chloride media) and R. solanacearum was identified based on morphological, pathological and biochemical properties and polymerase chain reaction (PCR) by using the species-specific primers. Studies showed that 81.54% (106) samples were positive on tetrazolium chloride solid medium. Among them 90 isolates were virulent and rest of them were avirulent. Fifty isolates were selected for chemical characterization based on hypersensitivity test. R. solanacearum is gram negative, aerobic facultative bacteria on the basis of chemical characterization. Fifty tested isolates expressed as race 3 while in biovar test forty-eight showed as biovar III and the rest two showed as biovar I. Much information can be gleaned from the study of cell organelles. The differences in the genomes of organelles, such as the mitochondrion and the chloroplast are amenable to phylogenetic and cladistic studies. These differences include the genome sequence, GC%, genome length and gene order. The conserved nature of the organelle genomes and the gene inventory of both mitochondrial and chloroplast genomes also make this easier to accomplish. This paper includes a review of existing organelle genome software. These include gene annotation and genome visualization tools as well as organelle gene databases for both mitochondrion and plastid. A new R tool, available on github, called “Organelle DNA Lineages”, or ODL, was written to compare and classify organelle genomes based on their genome sequence and gene order. The software was run on the mitochondrial genomes of a set of 51 cephalopod species, delineating ten separate monophyletic groups, including argonauts, nautiluses, octopuses, cuttlefish, and six squid groups. The occurrence of Aspergillus species in garri on sale in markets in Benue State, Nigeria, was studied by molecular techniques. Aspergillus species were isolated and purified on Potato Dextrose Agar. DNA from the purified isolates was extracted using the ZR fungal DNA miniprep and amplified by PCR mix made up of 12.5 μL of Taq 2X Master Mix. Primer sequences for the fungi characterization were internal transcribed spacers ITS 4 and ITS 5. The phylogenetic tree was plotted between the isolated organisms and reference sequences and evolutionary analysis was conducted in MEGA X. Result revealed that one thousand, six hundred and forty-six Aspergilli were isolated comprising of 980 and 666 isolates from the white and yellow garri respectively. Aspergillus flavus, A. fumigatus, A. niger, A. aculeatinus, and A. aculeatus were identified. Twenty percent (20%) of the strains had aflatoxin D structural gene, 50% amplified AFLP and 70% of the strains expressed AFLQ genes needed for the biosynthesis of aflatoxin B1. Majority of the strains that showed the expression of these structural genes were consistent with Aspergillus flavus. Methodology: Using bacteria cultural method, microbial species were isolated from feacal materials, identified and quantitated through application of genomic spectrophotometric systems with a quantitation of some marker biochemical parameters for Diabetes. Result: We observed a concentration of firmicutes, bacteriodetes, protecbacteria and bifidobacterium with Escherichia coli population predominating. Biochemical parameters reveal a 3-fold raised value for some bromakers in type 2 diabetes. At a confidence interval of 95% paired sample test results gave significant differences for all tested pairs.
