Methods: Experimental model of knee OA was replicated in the right hind limbs of 54 male rabbits by using modified Hulth modeling method. The rabbits were randomly divided into Model group, Lugua Polypeptide group and Radiofrequency Hyperthermia group. After modeling, Lugua Polypeptide group was given intramuscular injection of Lugua polypeptide; Radiofrequency Hyperthermia group was treated with radiofrequency hyperthermia; Model group was not given any special treatment. On the 7, 13 and 19 day after radiofrequency hyperthermia, six experimental rabbits were chosen from each group and sacrificed to take out medial femoral condyle cartilages in the right hind limbs. Modified Mankins rating was applied to the morphological evaluation. Meanwhile, quantitative real-time PCR was used to detect the content of type II collagen in cartilage tissues of medial femoral condyle. Results: At each time point after treatment, Mankins scores were decreased in all the 3 groups, with that of Model group significantly higher than those of the other two groups (Model group > Lugua Polypeptide group > Radiofrequency Hyperthermia group). The contents of type II collagen in cartilage tissues were increased in all the 3 groups, with that of Radiofrequency Hyperthermia group significantly higher than those of the other two group (Model group < Lugua Polypeptide group < Radiofrequency Hyperthermia group). The difference between groups was of statistical significance (p < .05). For Radiofrequency Hyperthermia group, Mankins scores were decreased gradually as the treatment time went by, with the content of type II collagen in cartilage tissues increased. The difference between time points was of statistical significance (p < .05). Conclusions: Radiofrequency hyperthermia is superior to Lugua polypeptide in the treatment of knee OA, at least in rabbits. Its therapeutic mechanism may be related to the significant increase in type II collagen in cartilages.","PeriodicalId":63789,"journal":{"name":"临床病例讨论杂志(英文版)","volume":"31 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2018-09-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"80747889","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, the effect of mifepristone on sperms is poorly studied. This research is intended to investigate the biological effect of mifepristone on human sperm function in vitro. Methods: Spermatozoa were incubated in the culture medium containing 0.04 μM, 0.4 μM, 4 μM and 40 μM of mifepristone for one hour after capacitation. It was required to measure sperm motility, viability, DNA integrity, acrosomal reaction, sperm hyperactivation, and the concentration of free calcium ions in spermatozoa. Results: After treating sperms with different concentrations of mifepristone in vitro, sperm viability, motility, morphology and DNA integrity were measured. The differences between the control group and the experimental group were of no statistical significance (p > .05). When the concentration of mifepristone was higher than 0.4 μM, the proportion of sperm hyperactivation was significantly reduced (p < .01). According to further examination on the rate of acrosomal reaction (AR rate) and the concentration of free calcium ions in sperms, it was found that acrosomal reaction was significantly inhibited with the concentration of free calcium ions in sperms decreased when the concentration of mifepristone was higher than 0.4 μM. Conclusions: The in-vitro spermicidal mechanism of mifepristone is realized by inhibiting acrosomal reaction, hyperactivation and decreasing the concentration of free calcium ions.","PeriodicalId":63789,"journal":{"name":"临床病例讨论杂志(英文版)","volume":"30 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2018-09-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"83096818","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) techniques were used to determine the GR gene bcl-I single nucleotide polymorphism in 76 children patients with asthma and 50 healthy controls. The plasma cortisol levels were detected by radio immunoassay in two groups respectively. Results: There were three genotypes found in bcl-I gene locus of these two groups, which were genotype CC, GG and CG. The frequencies of CC, CG, GG genotypes were 48.7%, 27.6%, 23.7% in the asthma group and 76%, 20%, 4% in the control group. The difference was of statistical significance ( p < .05). The frequency of G allele in the asthma group was 37.5%, which was significantly higher than 14% in the control group ( p < .05), indicating that G allele was associated with asthma. Conclusions: GR gene bcl-I polymorphism is significantly associated with the occurrence of bronchial asthma. G allele may be a susceptibility gene for steroid-resistance in asthma. Steroid-resistance has no correlation to the plasma cortisol level.","PeriodicalId":63789,"journal":{"name":"临床病例讨论杂志(英文版)","volume":"11 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2018-09-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"91380576","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: hUCMSCs were isolated from umbilical cord tissues of full-term healthy fetuses after cesarean section and then cultured. Cells in the third passage were chosen for the use of experiment after identification. SA strains isolated from wounds of burn patients in our burn wards were used in the following experiment. Cells were divided into 0, 10, 100 and 1,000 ng/ml lipopolysaccharide (LPS) groups by use of the random number table (similarly hereinafter). Cells were cultured with culture medium containing mesenchymal stem cells (MSCs) after being treated with medium containing corresponding mass concentrations of LPS for 12 h. At post culture hour (PCH) 6, 12 and 24, 6 wells of culture supernatant of cells in each group were obtained to measure the content of LL-37 with enzyme-linked immunosorbent assay (ELISA). Ninety blood agar culture plates were divided into ciprofloxacin control group (CC), ciprofloxacin + supernatant group (CS), and ciprofloxacin + supernatant + LL-37 antibody group (CSL), with 30 blood agar culture plates in each group. Blood agar culture plates in group CC were coated with 1.5 × 10 8 colony forming unit (CFU)/ml bacteria solution prepared with normal saline. Blood agar culture plates in group CS were coated with 1.5 × 10 8 CFU/ml bacteria solution prepared with normal saline and hUCMSC culture supernatant (cultured by MSC culture medium, the same below) in double volume of normal saline. Blood agar culture plates in group CSL were coated with 1.5 × 10 8 CFU/ml bacteria solution prepared with normal saline, hUCMSC culture supernatant in double volume of normal saline, and 2.6 μL of LL-37 antibodies at the concentration of 2 μg/ml. At PCH 12, 24 and 48, 10 blood agar culture plates were taken out from each group to observe the distribution of SA colonies on blood agar culture plates and to measure diameters of zones of inhibition of ciprofloxacin. The minimum inhibitory concentration (MIC) of ciprofloxacin against SA in each group was recorded. Fractional inhibitory concentration (FIC) indexes of ciprofloxacin in group CS and group CSL at PCH 12, 24 and 48 were calculated, with the synergistic effect evaluated. Data were processed with factorial design ANOVA, one way ANOVA, LSD- t test, Kruskal-Wallis test and Mann-Whitney U test. Results: (1) At each PCH, the content of LL-37 in cell culture supernatant in 10 ng/ml LPS group, 100 ng/ml LPS group or 1,000 ng/ml LPS group was higher than that in 0 ng/ml LPS group (with t values ranging from 11.22 to 33.36, p values all below .01); the content of LL-37 in cell culture supernatant in either 100 ng/ml LPS group or 1,000 ng/ml LPS group was higher than that in 10 ng/ml LPS group (with t values ranging from 2.24 to 18.73, p < .05 or p < .01); the content of LL-37 in cell culture sup","PeriodicalId":63789,"journal":{"name":"临床病例讨论杂志(英文版)","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2018-06-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"88611647","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Methods: From May 2013 to October 2015, 20 cases of patients received femoral anterolateral free flap transplantation, with the flap transported to the recipient site. After reascularization, the phase I of thin flap repairing was performed in each “vein type” area. Among those, there existed 8 cases of direct suture in the donor site, and 12 cases of skin transplantation by intermediate split thickness skin graft. Results: All the postoperative flaps survived, with 2 cases of vascular crisis and 2 cases of poor venous return at the edge of flap. No complications such as flap infection and necrosis occurred. Wounds and incisions in donor and recipient sites were healed in phase I. 20 cases of patients had been followed up for 5-24 months, and the average follow-up time was 15 months. The skin texture of flap was similar to the surrounding skin, with good abrasion resistance, no ulceration and no phase II flap repairing; the sensation of flap recovered to S3-S4 level; the two-point discrimination was 3.5-6.0 mm, with the average value of 5.0 mm. The function of patients’ fingers recovered well without any complications such as contracture and deformity etc. In the last follow-up, Testative Evaluation Standard of Upper Limb Function, which was established by Chinese Medical Society for Surgery of Hand, was used to assess efficacy, and the evaluation results were as follows: excellent in 9 cases, good in 5 cases, fair in 6 cases; in accordance with the evaluation criteria listed in Michigan Hand Outcome Questionnaire (used to assess patients’ satisfaction with the appearance of flaps after operation, and the results were as follows: very satisfactory in 9 cases, satisfactory in 9 cases, and moderate in 2 cases.
