In the study, the ovarian cancer cell lines (BT 549) were rolled into a control group (normal culture-defined as BT549/NC) and a response group (transfected with non-coding RNA LINC00857 cultured cells-defined as BT 549YAP cells). The expression and proliferation ability of the transcription factor YAP-TEAD in the two groups of cancer cells were analyzed and compared. The results showed that the YAP-TEAD expression rate was the highest in Bt549 cells; the YAP content grade (0.18) in BT 549-YAP cells was lower than BT 549/NC (0.2) after transfection (P< 0.05); and the apoptotic rate of the response group (80%) was higher than that of the control group (25%) after the intervention. With the extension of culture time, the expression of CCN1 mRNA decreased (P< 0.05), and CCN2 mRNA increased (P< 0.05). After 12, 24, 36, and 48 hours, the apoptosis rate of the reaction group at different time points was higher than that of the control group (P< 0.01). When YAP-TEAD was down-regulated, the in vitro proliferation ability of BT 549-YAP cells was weakened compared with BT 549/NC and parental cells. It was concluded that the non-coding RNA LINC00857 can target the transcription factor YAP-TEAD in the Hippo signaling pathway to decrease its expression, thus inhibiting the proliferation, migration, and invasion of cancer cells, and promoting cell apoptosis.</p>","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"162-170"},"PeriodicalIF":1.6,"publicationDate":"2022-02-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40616926","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

TB patients and HIV-infected/AIDS patients registered in Fuzhou Center for Disease Control and Prevention were selected as research subjects. The population characteristics of TB/HIV dual infection and mutation of drug-resistant genes were discussed. It was found that TB patients aged 20-40 years had the highest HIV infection rate, followed by those aged over 40 years. The rate of HIV infection in smear-negative TB patients was higher than that in smear-positive TB patients. HIV/AIDS patients aged 20-40 had the highest TB infection rate. In addition, men had higher rates of HIV than women, and married people had lower rates of HIV than single people. Mycobacterium tuberculosis (MTB) had the highest resistance to isoniazid (42.86%), followed by ofloxacin (34.82%), streptomycin (33.81%), and rifampicin (32.15%). Among the 113 cases of multi-drug resistant strains, 82 cases had mutations in the rpoB gene, with a gene mutation rate of 55.75%. The mutations ranged from codon 511 to codon 569. A total of 31 cases had mutations in the katG/inhA gene. Of which, there were 17 cases of katG single gene mutation, 9 cases of inhA single gene mutation, and 5 cases of combined katG and inhA gene mutation. It was suggested that it was necessary to carry out key TB/HIV two-way screening for TB patients older than 40 years old/smear-negative and male, single, and HIV-infected/AIDS patients aged 20-40. The resistance of MTB to antiTB drugs in this area was generally high, and the drug resistance of retreated patients was significantly higher than that of newly treated patients. Among the resistance genes, the rpoB gene had the highest mutation frequency, followed by the katG gene and inhA gene.</p>","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"109-118"},"PeriodicalIF":1.6,"publicationDate":"2022-02-28","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40627987","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Normotensive rats were used to study antihypertensive activity. The plant extract and its fractions relaxed the carbachol-induced contraction in the tracheal fragments and shifted the concentration-response curve of carbachol towards the right confirming the muscarinic receptor antagonist activity. The relaxation of phenylephrine-induced contraction in an aortic fragment by the extract showed α- adrenergic blocking activity. Furthermore, the extract produced a cardio-selective response in the paired atria and decreased the blood pressure in anesthetized normotensive rats. This study aimed to evaluate the expression changes of Cytokeratin 19 (CK19), vascular endothelial growth factor (VEGF), P53, ki67, and c-ert-B2 in OSCC patients. For this purpose, 30 patients were selected as the case group and 30 healthy individuals as the control group. The expression of CK19 and VEGF genes in their blood serum was measured. Also, the expression of ki67, P53, and c-ert-B2 markers in squamous cell carcinoma was evaluated using immunohistochemistry. T-test was used to analyze the data. The results showed that the presence of CK19 marker in people with OSCC was positive in 17 out of 30 patients and VEGF marker in 23 out of 30 patients. The mean of ki67 positive, P53 positive, and Cerb-B2 positive cells were 399.4, 221.4, and 26.8, respectively. The correlation test between the indices showed a statistical correlation between the incidence of ki67 and P53 (r = 91.5% and p = 0.02). While statistical correlation was not seen between the incidence of ki67 and Cerb-B2 index (r = -1.7% and p = 0.97) and P53 and C-erb-B2 index (r = -13% and p = 0.8) (p <0.05). In general, the expression of VEGF and CK19 genes is higher in patients with OSCC than in healthy individuals. Therefore, examining the expression level of these two biomarkers in the blood of OSCC patients can be considered as a diagnostic screening method in the early stages of the disease. The immunohistochemical study of squamous cell carcinoma can also be used as a diagnostic screening test in the early stages of the disease.</p>","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"233-239"},"PeriodicalIF":1.6,"publicationDate":"2022-02-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40494317","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
