Cerebral Ischemia-Reperfusion refers to the body's damage mainly caused by oxidative stress. This study aims to investigate the alleviating effect of TFPFC on brain neurological impairment and its influences on Nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase 1 (HO-1) expressions in rats with Ischemia-Reperfusion. The rat model of Ischemia-Reperfusion was established, and rats were treated with TFPFC or normal saline. At 24 h after reperfusion, the neurological score, volume of cerebral infarction and cerebral water content were analyzed in different groups. The influences of TFPFC treatment on the proliferative activity and apoptosis of oxygen and glucose deprivation/reoxygenation (OGD/R) neural stem cells were detected via methyl thiazolyl tetrazolium (MTT) assay and flow cytometry. Moreover, the malondialdehyde (MDA) level and superoxide dismutase (SOD) activity were measured to evaluate the oxidative stress effect. The influences of TFPFC treatment on the protein and messenger ribonucleic acid (mRNA) expressions of Nrf2 and HO-1 were analyzed using reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting. The TFPFC treatment alleviated the neurological impairment in rats after Ischemia-Reperfusion and reduced the volume of cerebral infarction and cerebral edema status in rats with Ischemia-Reperfusion. TFPFC increased the proliferative activity of OGD/R neural stem cells and decreased damage and apoptosis. In addition, the TFPFC treatment reduced the MDA level, improved the SOD activity, and up-regulated the protein and mRNA expressions of Nrf2 and HO-1. The TFPFC treatment may improve oxidative damage and protect the nervous system through the up-regulation of expressions of transcription factors Nrf2 and HO-1.
","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"155-160"},"PeriodicalIF":1.6,"publicationDate":"2022-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"33529232","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The study investigates the effect of growth conditions on bacteriocin production by Lactobacillus plantarum. The response surface methodology was applied to optimize and determine the interaction among the process variables in bacteriocin production by Lactobacillus plantarum and determine their optimum levels. Chloroform-Methanol (2:1 v/v) was used for crude bacteriocin extraction through the liquid-liquid extraction method, and its antimicrobial activity was assessed. The sample has shown inhibitory activity against all the organism,s i.e., E. coli, S. aureus, K. pneumoniae and P. aeruginosa good diffusion. Compared to control and ciprofloxacin, the sample at all concentrations (Lab 9, 1%, 2%, 3%) has shown better inhibitory activity in pH-7 and-8 at 37 °C against S.aureus in good diffusion. The selected factors were carbon and nitrogen source, temperature, and pH. The bacteriocin was produced at maximum activity in MRS broth supplemented with 1% dextrose and 1% ammonium nitrate. RSM analysis found that the optimal temperature for bacteriocin production was 36°C at a pH of 6.5 using 1% inoculum. At the same time, the increase in the percentage of inoculum (2% and 3%) did not affect bacteriocin production. The quadratic model found that temperature and pH profoundly affected bacteriocin production.
","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"105-110"},"PeriodicalIF":1.6,"publicationDate":"2022-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"33504067","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In recent decades, oxidative stress (OS) has been shown to be involved in the progression of this disease through DNA, lipid and protein damage, resulting in synovial inflammation. There are many causes of OS; metabolism is involved in the production of reactive oxygen species (ROS) but pollution, diet and microbiota imbalances could lead to the overproduction of these ROS. A decade of research focused on understanding how OS is promoted by known RA risk factors is described herein. The use of antioxidants represents an integrative treatment for patients with rheumatoid arthritis, given the evidence of the damage caused by oxidative stress in this disease. Understanding the different factors that contribute to the development and progression of RA, such as OS, will pave the way not only for better pharmacological treatments but also for recommendations for dietary and health behaviours that will benefit patients with this disease.
","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"174-184"},"PeriodicalIF":1.6,"publicationDate":"2022-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"33503692","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

During CO poisoning, the body increases anti-inflammatory proteins, including heme oxygenase-1 (HO-1), in response to oxidative stress. Considering the myocardial injury resulting from CO poisoning and the lack of sufficient information about the effect of HBOT on HO-1, the present study evaluated the effect of hyperbaric oxygen therapy on heme oxygenase-1 (HO-1) in patients with acute carbon monoxide poisoning and myocardial injury. In this regard, in a before-after Quasi-Experimental study, 20 patients with carbon monoxide poisoning and myocardial injury were studied. All patients underwent 40 daily hyperbaric oxygen therapy sessions for 90 minutes at a pressure of 2.4 ATA. Also, 20 healthy individuals, as a control group, were participated. To evaluate and compare the mRNA level of the HO-1 gene, the Real-time PCR technique was used. Paired t-test was used to compare the two indices of 6min walking distance and pulmonary arterial pressure (PAP) before and after the intervention. The results showed that the difference during 12 weeks was 8.65 ± 4.91 for PAP, and this reduction in pressure was statistically significant (P = 0.0092). The distance traveled increased by 28 ± 10.88 m in 6 minutes at the end of the study (P = 0.0084). Regarding the expression level of HO-1, the results showed that the expression level in the intervention group before the test had a significant increase compared to the control group (p = 0.0004). However, after hyperbaric oxygen therapy, the expression of this gene decreased significantly, and there was no statistically significant difference with the control group (p = 0.062). Overall, the results showed that HBOT significantly decreased HO-1 gene expression in CO poisoning and myocardial injury patients. It indicates the importance of HBOT in the treatment and compensation of cardiac tissue damage caused by CO poisoning.
","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"36-39"},"PeriodicalIF":1.6,"publicationDate":"2022-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"33504970","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

SPSS, t-test, Mann-Whitney U, Chi-square, and Fisher performed data analysis and comparison. The results showed that the changes in hemodynamic variables and PaO2, SaO2, and ETCO2 were not statistically significant between the two groups. Aldrete's score at entry and exit of recovery was similar between the two groups. During the recovery period, the percentage of pain or chills in the group under complete intravenous anesthesia was significantly higher. There was no significant difference between the two groups regarding the frequency of nausea and hypotension. Also, the results of IDO gene expression showed that general anesthesia with the thoracic epidural block (dezocine and ropivacaine), which is involved in inducing immunological tolerance and suppressing immune responses, has no significant effect. The stress of performing surgery before surgery can play a role in suppressing the patient's immunity, and anesthesia of the thoracic epidural block (dezocine and ropivacaine) has no significant effect on IDO expression. In general, thoracic epidural block with complete intravenous anesthesia has no significant effect on oxygen saturation in ventilated lungs compared with intravenous anesthesia alone. Nevertheless, this combination significantly reduces postoperative pain and chills.
","PeriodicalId":520584,"journal":{"name":"Cellular and molecular biology (Noisy-le-Grand, France)","volume":" ","pages":"25-30"},"PeriodicalIF":1.6,"publicationDate":"2022-06-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"33504972","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
