The pathogenesis of atherosclerosis is tightly linked to chronic inflammation and dysregulated lipid metabolism, processes that are also implicated in other inflammatory diseases like rheumatoid arthritis and psoriasis. Monoclonal antibodies (mAbs) have emerged as a promising therapeutic strategy, offering targeted intervention against key molecular drivers of atherosclerosis. This review summarizes recent advances in the development and clinical application of mAbs targeting both lipid-lowering pathways-such as low-density lipoprotein (LDL) and proprotein convertase subtilisin/kexin type 9 (PCSK9)-and inflammatory mediators, including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-17 (IL-17). Notably, anti-PCSK9 antibodies like alirocumab and evolocumab have demonstrated significant reductions in LDL-C levels and cardiovascular events in large-scale clinical trials. Similarly, antibodies targeting inflammatory cytokines have shown efficacy in reducing vascular inflammation and associated risks. The review also discusses the advantages and limitations of therapeutic mAbs, such as their high specificity, potential for adverse immune responses, and challenges related to tissue penetration and cost. Overall, monoclonal antibody therapy represents a significant advancement in the management of atherosclerosis, with ongoing research aimed at optimizing efficacy, safety, and accessibility. Sandwich Elisa technique was used to determine the concentration of cytokines. Phenolic contents and other active compounds of thyme were analyzed by Gas Chromatography-Mass Spectrometry (GC-Mass). In this study 15 male adult albino rats were divided into 3 groups (n=5), group one (G1) was the control group which fed on basal diet. Group two (G2) was the Rheumatoid arthritis (RA) group in which the rats were inoculated with 0.1 ml of CFA (Complete Freund's Adjuvant) yet fed on basal diet. Group three (G3) was the treatment group in which rats were inoculated with CFA along with the administration of thyme extract orally for 22 days. The results show that treatment with thyme extract significantly reduced proinflammatory cytokines IL-1 (interleukin-1), IL-6 (interleukin-6) and TNF- α (tumor-necrosis factor-alpha). Anti-inflammatory IL-10 (interleukin-10) showed a significant increase in the thyme-treated group. CD4 T (cluster of differentiation-4) cell levels showed a significant difference, while sCD14 (soluble cluster of differentiation-14) levels were non-significant in the thyme group compared to the RA group. Inflammatory markers (C-reactive proteins) CRP and (anti-cyclic citrullinated peptide) Anti-CCP antibodies were both significantly elevated in RA and significantly reduced by thyme treatment. Although body weight changes were statistically non-significant, they were visibly prominent. Paw edema was significantly decreased in the thyme-treated group. This study aimed to investigate the effects of different growth durations and salt concentrations on the production of bioactive compounds by these plants. This experiment was conducted under natural conditions and the plants were subjected to salt stress at different stages of growth. This analysis focused on assessing the production of polysaccharides, flavonoids, ergosterol, adenine, adenosine, hypoxanthine, and guanosine by UV spectrophotometer and high-performance liquid chromatography (HPLC) in both plants. The results showed that Z. officinale exhibited the highest polysaccharide content at 20-d of growth with 3mM salt, whereas G. glabra showed slightly lower polysaccharide content. Similarly, Z. officinale had higher flavonoid content at 25-d of growth with 5 mM salt compared to G. glabra. Additionally, Z. officinale demonstrated higher concentrations of ergosterol, adenine, adenosine, hypoxanthine, and guanosine than G. glabra, particularly at 25-d of growth with 5 mM salt. This study investigated the relationship between HbA1C, glucose levels, trace elements (lead, zinc, selenium, magnesium), vitamin A, and lipid profiles (TC, TG, HDL, LDL, VLDL) in type 2 diabetes (T2D) and DR patients, considering the impact of smoking. The study, conducted at Nasiriya General Hospital and the Center for Endocrinology and Diabetes (May 2023-March 2024), included three age groups (15-35, 35-55, and 55-75 years) divided into smokers and non-smokers. Results showed significantly higher glucose, HbA1C, lipid and selenium levels in T2D and DR patients compared to controls (p≤0.05), with smokers exhibiting greater lead levels. Zinc and vitamin A were significantly lower in DR patients, particularly among smokers. However, published data on HER2 expression in colorectal cancer (CRC) remain controversial. This study investigates the immunohistochemical and molecular expression of HER2 in primary CRC and evaluates its clinicopathological and prognostic significance in Tunisian patients. A retrospective analysis was conducted on 144 CRC patients. HER2 status was assessed by immunohistochemistry and tissue microarray analysis, following the diagnostic criteria for gastroesophageal adenocarcinoma. CRC cases with ambiguous results underwent chromogenic in situ hybridization. The mean patient age was 61.9 years (male-to-female ratio: 1.18:1). Tumors were classified as colonic (74.3%) or rectal (25.7%), with 45.8% located in the left colon. Stage III disease was identified in 37.5% of cases, and distant metastases were present in 13.9%. HER2 expression results were as follows: negative (score 0/1+) in 142 cases (98.6%), equivocal (score 2+) in one case (0.7%), and overexpressed (score 3+) in one case (0.7%). No HER2 gene amplification was detected, and none of the metastatic CRC cases showed HER2 immunostaining. These findings suggest that HER2 overexpression and amplification in CRC are rarer than previously reported, highlighting the need for multicenter Tunisian studies to validate these results. The total alkaloids were extracted in vitro from Caulis Mahoniae, and cervical cancer HeLa cell lines were used as experimental objects. The half inhibitory concentration (IC50) of total alkaloids on HeLa cell lines was detected via the preliminary experiment, the influences of total alkaloids at different concentrations on the proliferation of HeLa cell lines were detected via methyl thiazolyl tetrazolium (MTT) assay, and the cell growth curve was plotted. Moreover, the cell cycle and apoptosis after treatment with total alkaloids at different concentrations were detected via flow cytometry, and the caspase-3 gene and protein expressions were detected via reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting. The IC50 of total alkaloids in Caulis Mahoniaeon HeLa cell lines was 12.5 μg/mL. With the gradual increase of concentration of total alkaloids in the treatment of cervical cancer cells, the inhibitory rate on cancer cells was gradually increased, and the proportion of cells in the G0/G1 phase was gradually decreased, while that in S and G2/M phases was gradually increased. Besides, with the increase in the concentration of total alkaloids, the apoptotic rate of cervical cancer cells was gradually increased, and both caspase-3 gene and protein expressions were also gradually increased. For this aim, E. coli were collected for bacterial identification and drug sensitivity testing from bloodstream infections in elderly patients in the hospital from January 2016 to December 2019. ESBLs positive strains were assayed for genotypes and their integron carriage rates by PCR amplification. The characteristics and differences of various genotype rates were compared and analyzed. Results showed that a total of 230 E. coli strains were isolated. The detection rate of ESBLs-producing bacteria was 37.39 %. ESBLs-producing E. coli showed a high rate of resistance to cefepime, levofloxacin, cotrimoxazole, and ticarcillin/clavulanic acid (>40%). The resistance rate of 230 strains of E. coli to meropenem, minocycline, amikacin, gentamicin and cefoxitin was less than 20%. Among the ESBLs-producing E. coli in bloodstream infections in elderly patients, CTX-M-9 accounted for 27.91%, CTX-M-2 for 17.44%, and SHV for 13.95%. The detection rate of type I integrated genes was 41.30%, and type II and III integrated genes were not detected. ESBLs-producing genotyping-positive bacteria were detected with more than 50% of type I integrated genes. Overall survival is related to clinical stage: more advanced stages show lower survival rates; therefore, they need to be monitored regularly with new, less invasive and more specific biomarkers. The concentration and integrity index of circulating cell-free DNA (ccfDNA) have been proposed as potential diagnostic and prognostic biomarkers for CRC, however, inconsistent results are still observed in different reports. Here we analyze these potential CRC biomarkers in a Mexican population. In this study, 124 patients with sporadic CRC and 37 healthy individuals were examined as a reference group. The ccfDNA was isolated from plasma samples of all included subjects. The ccfDNA concentration was determined by fluorometry and the integrity index (ALU247/ALU115 ratio) by quantitative PCR amplification (qPCR) of ALU sequences. The results show that ccfDNA concentration was higher in CRC patients than in the reference group (P=0.001). The integrity index showed no significant differences between these groups (P=0.258), except for histological type (P=0.012). A higher ccfDNA concentration was also associated with patients younger than 50 years (P=0.030). The ccfDNA concentration showed significant discriminatory power (AUC: 0.854, C.I.: 0.78-0.92, P=0.001) between patients and the reference group and between tumor-node-metastasis (TNM) stages. The researchers reviewed ten cases of PASCC and summarize their clinical outcomes, pathological morphological traits, immunophenotypes, treatment plans and the corresponding follow-up data. Results showed that the pathological morphology revealed complex reticular structures, where numerous tracts of anastomose, and lacunar structures lined with atypical neoplastic cells, which resembles the histopathological appearance of angiosarcoma. Particularly, we observed pathologic patterns that resemble Sclerosing Epithelioid Fibrosarcoma (or Myxoid Fibrosarcoma) in the patients who suffered a relapse. All cases present negative results for vascular markers (CD31, ERG) and positive results for epithelial markers (CK-pan, p40). The average age of the participants is 60 years old (range: 48-79), relative aged, and there is no significant difference between male and female participants (6 men and 4 women). The locations of neoplasms involve face (n=3), upper limbs (n=1), waist(n=1), cervix uteri (n=1), lungs (n=2), thyroid (n=1), and breasts (n=1). All participants had received clinical follow-ups that range from 4 to 47 months, during which the researchers observed Lymph Node Metastases developed in three participants (out of 10; 30%); Distant Metastases in five participants (out of 10; 50%); two local recurrences at the site of surgical resection; and four deaths due to disease (out of 10; 40%), with 9.5 months estimated median survival time and 9 months mean survival time. It was concluded that PASCC presents the tendency for recurrence and metastasis. Accurate pathological diagnosis and standardized medical procedures are crucial to the treatment of PASCC. This study investigated the fundamental molecular function of the TALNEC2 in the pathogenesis of cerebral infarction to provide insights on the potential novel therapeutic agents in cerebral infarction. RT-qPCR measured expression of TALNEC2 and JNK in human neural cell line SH-SY5Y. Cell transfection upregulated or silenced the genes with MTT assay examining cell viability. RT-qPCR detected cell death in the apoptosis biomarker caspase-3, inflammation in the biomarkers C-reactive protein (CRP) and IL-6 and verified cell proliferation via the ki67 and PCNA markers. Luciferase assay was performed to see the luciferase activity and western blotting determined the protein expression of JNK in proliferation, inflammation. The results demonstrated that TALNEC2 was highly expressed after OGD/R treatment in nerve cells after cerebral infarction. In addition, TALNEC2 silencing prevented apoptosis and inflammation of nerve cells after cerebral infarction. TALNEC2 directly interacted with miR-19a-3p to regulate JNK protein expression. Lastly, miR-19a-3p inhibitor abolished the protective effect of si-TALNEC2 against OGD/R induced damage in vitro. In summary, this study has demonstrated that TALNEC2 is a positive moderator for pathogenesis of cerebral infarction. Furthermore, our conclusions provide further insights on the interplay among TALNEC2, miR-19a-3p and JNK in cerebral infarction.
