Infectious diseases of these species can potentially pose health risks to domestic animals and humans. However, few studies have been conducted in which immune system cells in these species were phenotyped. The aims of this study were to determine the cross-reactivity of a wide anti-human panel of commercial antibodies on formalin-fixed and paraffin-embedded (FFPE) samples and to describe the distribution of roe and fallow deer main immune cell subsets in the lymph nodes and spleen. Twenty retromandibular lymph nodes (RLNs) and spleen samples were collected from 10 roe deer and 10 fallow deer and were tested by a panel of 12 commercial anti-human antibodies. The CD79a, CD20, CD3, Iba-1, MAC387, and AM-3K antibodies were successfully labeled cells in cervine tissue, while the Foxp3 and the CD68 did not show suitable immunostaining. This study supplies the first immunohistochemical description of immune cell subpopulations in non-pathological spleen and RLNs from roe and fallow deer and provides an easily repeatable manual IHC protocol to immunolocalize cervine B-, T-cells, and macrophages subsets in FFPE tissue samples.</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9654140/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40459574","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The present study sought to replace FBS and BSA by insulin-like growth factor 1 (IGF1), fibroblast growth factor 2 (FGF2) and epidermal growth factor (EGF). In Experiment 1, absence of FBS from maturation medium (MM) did not affect the rate of in vitro maturation, as assessed by the extrusion of the first polar body. However, when gonadotropins and FBS were removed from the MM, the maturation rate was significantly reduced even in the presence of growth factors. Therefore, gonadotropin-supplemented MM medium was established as the base medium for the defined maturation condition. In Experiment 2, the addition of growth factors to gonadotropin-supplemented MM medium supported similar maturation (~90%) compared to the undefined condition (FBS-carrying). In Experiment 3, the addition of growth factors to embryo culture medium showed similar in vitro competence compared to the undefined (FBS) control. In Experiment 4, completely defined conditions (absence of FBS and BSA during in vitro maturation and embryo culture) were tested. A higher cleavage was observed with FGF2 (86%) compared to EGF (77%) and the FBS control (77%), but similar blastocyst rates were observed for FGF2 (24%), EGF (19%) and the FBS control (25%). Embryo quality was similar among groups. Finally, post-thawing survival was higher for FGF2 (94%) compared to the FBS control (77%). Thus, we report a simple defined IVP system for bovine species that generates developmental outcomes and embryos of similar quality than those produced under conditions containing FBS.</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9654503/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40457233","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The Cyprus mouflon (<i>Ovis gmelini ophion</i>) is such a species, which is endemic to the mountain habitats of Cyprus. Recognizing water scarcity to be a major pressure to the mouflon, and with global warming projected to intensify hot and dry periods in the region, the Game and Fauna Service has been maintaining a network of locally designed watering troughs in Pafos Forest-the mouflon's stronghold-since 1997. This study describes the mouflon's use of the water troughs and examines whether visitation rates differed at the daily or weekly scale in response to environmental, climatic or anthropogenic parameters. Using camera traps, ten troughs were monitored from September 2017 to March 2018 (1,065 days; range 29-164 days per trough). Mouflon were detected at seven troughs (mean herd size 1.5 ± 1.2) during 373 independent detections (≥30 min interval between photographs), with visits peaking during late morning and midday hours. Generalized mixed-effect models showed mouflon visiting water troughs more frequently during hotter days, regardless of recent precipitation. Visits were also more frequent at water troughs located close to tar roads. Moreover, there was no evidence of mouflon avoiding water troughs used by predators (red foxes, feral dogs) at either daily or weekly scale, or during hunting days. The study supports the value of artificial water troughs for mediating, partially at least, the effects of climate change on mountain ungulates such as the Cyprus mouflon. Additional studies are proposed that will examine both mouflon drinking patterns across all seasons and ways of improving the effectiveness of the current water trough grid.</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9656673/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40457235","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, the current dataset of chicken faces based on accurate data is scarce, detection models possess low accuracy and slow speed, and the related detection algorithm is ineffective for small object detection. To tackle these problems, an object detection network based on GAN-MAE (generative adversarial network-masked autoencoders) data augmentation is proposed in this paper for detecting chickens of different ages. First, the images were generated using GAN and MAE to augment the dataset. Afterward, CSPDarknet53 was used as the backbone network to enhance the receptive field in the object detection network to detect different sizes of objects in the same image. The 128×128 feature map output was added to three feature map outputs of this paper, thus changing the feature map output of eightfold downsampling to fourfold downsampling, which provided smaller object features for subsequent feature fusion. Secondly, the feature fusion module was improved based on the idea of dense connection. Then the module achieved feature reuse so that the YOLO head classifier could combine features from different levels of feature layers to capture greater classification and detection results. Ultimately, the comparison experiments' outcomes showed that the mAP (mean average Precision) of the suggested method was up to 0.84, which was 29.2% higher than other networks', and the detection speed was the same, up to 37 frames per second. Better detection accuracy can be obtained while meeting the actual scenario detection requirements. Additionally, an end-to-end web system was designed to apply the algorithm to practical applications.</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9655765/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40457229","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Although head, pelvis and limb movements have been described, less is known about the temporal relationships between them. This information is needed to understand how the movements change with lameness. This is particularly relevant in gaited horses, such as the Icelandic horse that perform gaits such as tölt and pace, which are challenging to evaluate. This study used inertial measurement units to investigate head, withers and pelvis motion relative to limb movements in Icelandic, Warmblood and Iberian horses. Limb movements, together with vertical movements and lowest/highest positions of the head, withers and pelvis were calculated, and the relative timing of the events was compared across breeds. Additionally, data for tölt and pace were collected and evaluated in ridden Icelandic horses. For all gaits except walk and pace, the lowest/highest positions of the head/withers/pelvis were closely temporally related to midstance and hoof-off, respectively. Pelvic and withers total range of motion differed between all breeds. The Icelandic horses showed shorter stride duration and smaller movements of the upper body than the other breeds at trot, which may explain why lameness evaluation in this breed is challenging. Abstract Knowledge of vertical motion patterns of the axial body segments is a prerequisite for the development of algorithms used in automated detection of lameness. To date, the focus has been on the trot. This study investigates the temporal synchronization between vertical motion of the axial body segments with limb kinematic events in walk and trot across three popular types of sport horses (19 Warmbloods, 23 Iberians, 26 Icelandics) that are known to have different stride kinematics, and it presents novel data describing vertical motion of the axial body segments in tölting and pacing Icelandic horses. Inertial measurement unit sensors recorded limb kinematics, vertical motion of the axial body at all symmetrical gaits that the horse could perform (walk, trot, tölt, pace). Limb kinematics, vertical range of motion and lowest/highest positions of the head, withers and pelvis were calculated. For all gaits except walk and pace, lowest/highest positions of the pelvis and withers were found to be closely related temporally to midstance and start of suspension of the hind/fore quarter, respectively. There were differences in pelvic/withers range of motion between all breeds where the Icelandic horses showed the smallest motion, which may explain why lameness evaluation in this breed is challenging.","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9657284/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40457230","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The material was taken from non-glandular (the blind ventricular sac) and glandular parts (the plicated edge margin/cardiac part, the body of stomach and the pyloric part) of the stomach. It was preserved and prepared according to the standard protocol. The histological slides were stained (H-E, Masson-Goldner and PAS). The analyses were performed using the light microscope. All measurements were statistically elaborated. The crown-rump length growth rate was estimated as isometric. The blind ventricular sac growth rate was lower than CRL (negative allometric) and the partition of stomach mucosa into non-glandular and glandular part occurred in the 1st age group. The plicated edge margin/cardiac part and the pyloric part shoved similar tendencies. Only the body of stomach demonstrated a higher growth rate than CRL (positive allometric), which can be explained due to the strongest development of fundic glands. Moreover, comparing the adult reference group to the three parts of the foetal period, all metric values were lower than those achieved prenatally. The blind ventricular sac was covered with the multiple plane epithelium. The glandular parts of stomach that formed the superficial concave areas were covered with the simple columnar epithelium in the 1st age group, which developed to the cardiac, fundic, and pyloric glands in the 2rd and 3rd age groups. The <i>propria mucosae</i> was built with the mesenchyme, which differentiated later to the loose connective tissue. The muscular layer of mucosa was not clearly distinguishable in the 1st age group. The muscular layer of the stomach wall was formed with myoblasts in the 1st age group and later in the 2nd and the 3rd age groups built with fusiform myocytes divided into internal and external layers. The non-differentiated cells of glandular epithelium transformed into the parietal and chief cells. The first were visible in the gastric glands of the 2nd age group. Both of them were present in the 3rd age group gastric mucosa. The PAS staining proved a moderate PAS-positive reaction in the 2rd age group, while it was estimated as intense Pas-positive in the gastric glands in the 3rd age group and was comparable to postnatal observation (the adult reference group).</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9656738/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40466182","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In this study, our aims were twofold: to observe and compare the spontaneous behaviour of companion cats and dogs in the same novel environment, and to habituate them (if needed) to the novel environment in the presence of their owners and an unfamiliar experimenter. The behaviour of 62 family cats, 31 experienced (which had the opportunity to meet strangers and explore unfamiliar places) and 31 inexperienced cats, and 27 family dogs was compared in an unfamiliar room. The subjects' behaviour was coded during the first five minutes in the presence of two passive persons, their owners, and an unfamiliar experimenter. Then, based on a set of rules, first the owner, and then the experimenter tried to initiate interactions with the subjects and the subjects' willingness to interact was evaluated. To pass the habituation test, subjects were expected to play with the experimenter or accept food from them. All dogs passed the test on the first occasion, while almost 60% of the cats failed. The cats' experience did not play a significant role, in fact, younger cats passed more easily. We found marked differences between dogs and cats in all behavioural variables; compared to dogs, cats spent more time crouching and close to their box, while less time exploring or close to the owner. We did not find a difference in the cats' behaviour based on their experience. Our results support the hypothesis that unlike dogs, cats need extensive habituation in a novel environment. This could partially be explained by the difference in the ecology and/or domestication process of the species, although developmental effects cannot be excluded. Our findings have fundamental consequences not only for the considerations of the testability of cats and on the interpretation of their data collected in the laboratory, but in a more general sense on developing comparative experiments.</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9657355/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40466181","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The home range of Swan Geese was estimated to be an area from Yubu Island in the south to Janggu Bay in the north; however, the core home range and habitat use characteristics differed significantly between daytime and nighttime (Day: 59.9 km<sup>2</sup>, Night: 40.3 km<sup>2</sup>, on average, 100% MCP). During the day (08:00-18:00), Swan Geese mostly spent time resting or feeding on tidal flats, especially those around tidal channels or paddy fields near Janggu Bay, whereas they mostly rested on sand dunes near Yubu Island along with the mudflats at Janggu Bay at night. Our results provide practical information on the habitat use of wintering Swan Geese population over time and indicate that Yubu Island is an important resting place. Hence, these results can contribute to evaluating threats to Swan Geese and establishing management and protection strategies for the Seocheon Tidal Flat, a major wintering site for the Far East Russian population of Swan Geese.</p>","PeriodicalId":519482,"journal":{"name":"Animals : an Open Access Journal from MDPI","volume":" ","pages":""},"PeriodicalIF":3.0,"publicationDate":"2022-11-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://www.ncbi.nlm.nih.gov/pmc/articles/PMC9658776/pdf/","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"40466183","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
