Objective: To evaluate whether the addition of curcumin to the diet of calves at different phases (pre- and post-weaning) has a positive effect on metabolic profile, performance, and anti-coccidian action. Methods: Thirtythree Holstein calves were selected at various phases of development: Experiment 1 (E1: n=10) 18±7 (pre-weaning), Experiment 2 (E2: n=11) 64±4 (pre-weaning) and Experiment 3 (E3: n=12) 95±8 (post-weaning) days of life. The calves were separated in three groups according to their phase of development. In each experiment, animals were divided into two sub-groups: control and curcumin. The curcumin groups received 200 mg of additive per animal/day either in milk (pre-weaning) or concentrate (post-weaning). Fecal collections were performed on days 0, 10 and 15 of the experiment to count Eimeria oocysts per gram of feces and to perform fecal score analysis. Complete blood counts, oxidant and antioxidant profiles, protein metabolism markers, lipid levels, glucose levels, and animal weights were measured. Analyses of digestibility and composition of the diet used in Experiment 3 (post-weaning) were also performed. Results: Independent of phase, animals that received curcumin had greater weight gain on days 0 to 15 (E1, E2 and E3 p=0.04, 0.001 and 0.001, respectively), probably due to the increased digestibility of hay and concentrate at 72h (p=0.03 and 0.02, respectively). The supplemented calves had lower level of oxidants (thiobarbituric acid reactive substances –TBARS- and reactive oxygen species –ROS-), indicating that free radical levels in serum and lipid peroxidation were lower. This was probably due to increased enzymatic antioxidants gluthatione S-transferase (E1, E2 and E3 p=0.001, 0.001 and 0.02, respectively), catalase (E1 p=0.001) and superoxide dismutase (E3 p=0.001) in treated animals at day 15. Furthermore, calves receiving curcumin had lower numeric number of Eimeria infection during the experimental period, and the difference was significant in day 15 (E1 and E2 p=0.02, and 0.001, respectively). Objective: To evaluate the transcriptional response promoted by biofloc on shrimp ( Litopenaeus vannamei ) under a recirculating aquaculture system (RAS). Methods: Quantitative real-time RT-PCR was used to monitor seven key genes related to the immune system in shrimp post-larvae, reared in a RAS with and without biofloc (BF and noBF). In addition, we present for the first time nucleotide sequences of ADP-ribosylation factor 4 (LvArf4) from Litopenaeus vannamei . Results: Transcripts for penaeidin3 (Pen3), penaeidin4 (Pen4), crustin, and Toll receptor (LvToll) genes were upregulated between 3 and 24 h in both systems, and tumor necrosis factor receptor-associated factor 6 (TRAF6) in no-BF as an early response. Regarding differential expression between treatments, 13 occurrences were encountered. Nine that were higher in BF than in no-BF and four higher in no-BF than in BF. In some sample times, expression of Pen3, crustin, LvToll, TRAF6, IMD, and LvArf4 was higher in BF than in no-BF and in others, expression of Pen3, Pen4, and TRAF6 was higher in no-BF than in BF. Conclusions: BF modulates the transcription of genes related to the immune response in shrimp as an early response. However, the RAS with no-BF promotes a similar response. Resumen Antecedentes: Los cultivos de camaron estan evolucionando de sistemas semi-intensivos a hiper-intensivos con biofloc y con recirculacion. Objetivo: Evaluar la respuesta transcripcional promovida por el biofloc en un sistema acuicola con recirculacion (SAR). Metodos: Monitoreamos mediante RT-PCR cuantitativo siete genes relacionados con el sistema inmune en postlarvas de camaron cultivadas en un SAR con y sin biofloc (BF y no-BF). Ademas, presentamos por primera vez la secuencia de nucleotidos del factor de ribosilacion 4 de ADP (LvArf4) de Litopenaeus vannamei . Resultados: Los genes penaeidina3 (Pen3), penaeidina4 (Pen4), Crustina y Toll (LvToll) se sobre-expresaron entre las 3 y 24 h en ambos sistemas, y el factor 6 asociado al factor de necrosis tumoral (TRAF6) en BF como una respuesta temprana. Con respecto a la expresion diferencial entre los tratamientos, se presentaron 13 ocurrencias. Nueve donde el BF fue mayor que sin-BF y cuatro donde el no-BF fue mayor que el BF. La expresion fue mas alta en BF que en no-BF en Pen3, Crustin, LvToll, TRAF6, IMD y LvArf4. En contraste, la expresion fue mayor en no-BF en Pen3, Pen4 y TRAF6. In horses, several species of the genus Malassezia have been reported in different areas of the skin and ear canal. Objective: Isolate, characterize and identify the different species belonging to the genus Malassezia isolated from the ear canal and skin of equine patients with no dermatological lesions that were referred to the large animal clinic of veterinary teaching hospital at the National University of Colombia. Methods: 22 horses were evaluated and sampled. Eighty-two samples were obtained by swabbing either the ear canals (left and right), skin areas of prepuce, mammary gland and inguinal region. The samples were examined by cytological evaluation and were cultured on modified Dixon’s agar and phenotypic and molecular identification were performed for yeast colonies. Results: Fourteen yeast isolates were obtained from the 82 samples. Biochemical identification determined that 50% (n=7) were Malassezia spp., 35.7% (n=5) were identified as Candida spp. and 14.3% (n=2) as Cryptococcus spp. Using molecular tests, the Malassezia species were M. slooffiae (28.6%) and M. nana (57.1%); only one isolate was classified as Trichosporo asahii. Conclusion: M. nana and M. slooffiae were identified as part of the normal ear canal and skin microbiota in the evaluated horses. The observed prevalence of Malassezia spp. was 18.2% (n=4/22) in this study sample. Keywords: culture; ear canal; horses; identification; Malassezia ; normal microbiota; PCR; skin; yeast. Resumen Antecedentes: Las levaduras del genero Malassezia hacen parte de la microbiota normal cutanea de humanos y animales. En equinos se han reportado diferentes especies de Malassezia aisladas de varias regiones de piel y canal auditivo externo. Objetivo: Aislar, caracterizar e identificar las especies del genero Malassezia spp. a partir de canal auditivo externo y piel de equinos sin lesiones dermatologicas, remitidos a la Clinica de Grandes Animales de la Facultad de Medicina Veterinaria y de Zootecnia de la Universidad Nacional de Colombia. Metodos: Se evaluaron 22 equinos, a partir de los cuales se obtuvieron 82 muestras entre hisopados de canal auditivo externo (izquierdo y derecho) y diferentes regiones de piel (prepucio, glandula mamaria e ingle). Las muestras fueron procesadas mediante examen directo y cultivo en agar Dixon modificado. A partir de los aislamientos en los que se observaron colonias morfologicamente compatibles con Malassezia spp. se realizo la identificacion fenotipica y molecular. Resultados: De las 82 muestras procesadas se obtuvieron 14 aislamientos de levaduras, de las cuales (n=2) a Cryptococcus spp. Luego mediante pruebas moleculares se identificaron las especies del genero Malassezia como: M. slooffiae (28,6%) y M. nana (57,1%); y un aislamiento correspondio a Trichosporon asahii. Objective: To assess the evolution of the structure and genetic diversity of the Romosinuano breed in Mexico, through pedigree analysis. Methods: Pedigree data was obtained from Asociacion Mexicana de Criadores de Ganado Romosinuano y Lechero Tropical (AMCROLET). The ENDOG program (4.8 version) was used to analyze two datasets, one that includes upgrading from F 1 animals (UP) and the other with only straight-bred cattle (SP). For both datasets, three reference populations were defined: 1998-2003 (RP1), 2004-2009 (RP2), and 2010-2017 (RP3). The pedigree included 3,432 animals in UP and 1,518 in SP. Demographic parameters were: Generation interval (GI), equivalent number of generations (EG), pedigree completeness index (PCI), and gene flow among herds. Genetic parameters were: Inbreeding ( F ) and average relatedness ( AR ) coefficients, effective population size ( N ec ), effective number of founders and ancestors, and number of founder genome equivalents. Results: The GI varied from 6.10 to 6.54 for UP, and from 6.47 to 7.16 yr for SP. The EG of the UP and SP improved >63% from RP1 to RP3. The PCI increased over time. No nucleus or isolated herds were found. For RP3, F and AR reached 2.08 and 5.12% in the UP, and 2.55 and 5.94% in the SP. For RP3, N ec was 57 in the UP and 45 in the SP. Genetic diversity losses were attributed mainly (>66%) to genetic drift, except for RP3 in the SP (44%). Conclusions: A reduction of the genetic diversity has been occurring after the Romosinuano breed association was established in Mexico, and this is mainly due to random loss of genes. However, in countries like Brazil, this trait is still not selected on a large scale, and no genetic parameters are reported in the literature. Objective: To estimate the variance components and genetic parameters for somatic cell score, milk yield, fat yield, protein yield, fat percentage, and protein percentage in Holstein cows. Methods: Records from 56,718 animals were used to estimate variance components, heritability, and genetic correlations using a multi-trait animal model by the REML method. Results: The heritability estimates were 0.19 for somatic cell score, 0.22 for milk yield, 0.26 for fat yield, 0.18 for protein yield, 0.61 for fat percentage, and 0.65 for protein percentage. The estimates of genetic correlations among analyzed traits ranged from -0.50 to 0.82. Conclusion: The low heritability observed for somatic cell score indicates that selection for this trait should result in benefits related to animal health and milk quality, but only in the long term. The low correlation between productive traits and somatic cell score indicates that inclusion of somatic cell score in animal breeding programs does not interfere negatively with the genetic selection for milk yield or solids. Resumen Antecedentes: El conteo de celulas somaticas es un parametro importante para predecir la calidad de la leche y la salud de las vacas. Sin embargo, en paises como Brasil, esta caracteristica aun no se selecciona a gran escala y no se reportan parametros geneticos en la literatura. Objetivo: Estimar los componentes de varianza y parametros geneticos para el conteo de celulas somaticas, produccion de leche, produccion de grasa, produccion de proteina, porcentaje de grasa y porcentaje de proteina en vacas de la raza Holstein. Metodos: Se usaron registros de 56.718 animales para estimar los componentes de la varianza, heredabilidad y correlaciones geneticas usando un modelo animal multicaracteristico por medio del metodo REML. Resultados: Las estimaciones de heredabilidad fueron 0,19 para el conteo de celulas somaticas, 0,22 para la produccion de leche, 0,26 para la produccion de grasa, 0,18 para produccion de proteina, 0,61 para el porcentaje de grasa y 0,65 para el porcentaje de proteina. Las estimaciones de correlacion genetica entre las caracteristicas analizadas variaron entre -0,50 a 0,82. Conclusion: La baja heredabilidad encontrada para conteo de celulas somaticas demostro que la seleccion para esta caracteristica podria resultar en beneficios para la salud animal y calidad de la leche, pero solo a largo plazo. Objective: To evaluate serum concentrations of triiodothyronine (T3) and thyroxine (T4) hormones in newborn calves from mothers supplemented with barium selenate during prepartum. Methods: Six black Frisian pregnant cows were supplemented with barium selenate subcutaneously during the last two months of gestation, until calving. Six cows were used as controls. All cows were subjected to a low Se diet, consisting of hay from natural pasture and commercial concentrate lacking Se. The Se balance was measured through the activity of erythrocyte glutathione peroxidase (GPx). Serum concentration of T3 and T4 in calves was determined by electrochemiluminescence. Results: Se supplementation during prepartum increased GPx activity in cows from day 45 post-supplementation (p<0.05). Calves from supplemented mothers showed higher average serum Se concentration than calves from non-supplemented mothers. The average concentration of T3 in the calves from supplemented mothers was lower in the first hour of life (p<0.05) compared with calves from mothers of the non-supplemented group. A decrease (p<0.05) in T4 serum concentrations was observed in both groups at seven days of age. Additionally, chemotherapeutic growth promoters are being substituted for safer alternative strategies. Objective: To evaluate the effect of feeding oleoresins from chili pepper (Capsicum annuum) and turmeric (Curcuma longa L.) on the performance, survival rate, and the Productive Efficiency Index of broilers from 1 to 21d challenged by Eimeria sp. Methods: A total of 700 male 1-dayold chicks (Cobb 500) challenged with Eimeria sp, were assigned to a completely randomized design with four treatments: (1) negative control diet, without anticoccidial agent or growth promoters; (2) control diet+0.05% salinomycin; (3) negative control diet+100g/ton; and (4) negative control diet +140g/ton of test product (Curcuma longa L. plus Capsicum annuum oleoresins). Results: Body weight, weight gain, feed conversion and Productive Efficiency Index did not differ between broilers fed anticoccidial chemotherapeutics and oleoresins. In addition, there was no difference in feed intake and survival rate between dietary treatments. During this period there is great variation in hematological and biochemical parameters. Many times these parameters are misinterpreted as being obtained from nonpregnant animals with unknown parasite status. Objective: To characterize the hematological, biochemical, and parasitological values within the peripartum period of Santa Ines sheep naturally infected. Methods: For this purpose, 45 ewes were used, with an average body weight of 50 kg. Data were collected during pregnancy, birth and, lactation. Measurements included body weight, body condition score (BCS), evaluation of conjunctive staining, and collection of blood and feces. Results: All variables changed throughout physiological stages. Weight and BCS were different between periods (p<0.05). Fecal egg counts (FEC) were higher in the peripartum period, with a predominance of Haemonchus ssp, 92.85%. Mean hematological and biochemical values varied throughout the study, (p>0.05). Conclusion: The influence of peripartum on the parameters studied should be considered and its use as a reference value. Objective: To determine the effect of adding two types of urea at two concentration levels to sugarcane silage on the nutritional value and fermentative characteristics of the silage. Methods: A completely randomized 2×2+1 factorial design was used, based on two types of urea (conventional and protected), two urea concentration (0.5 and 1.0% of the natural matter), and a control (sugarcane silage with no additives). Six replicates were used. The mixture was placed in PVC silos and opened after 60 days. Results: There was no difference in DM content or effluent losses by the addition of urea. In regards to neutral detergent fiber, acid detergent fiber, and lignin, decreased levels were observed compared to the control, regardless of urea type. Urea type and concentration had an effect on ammoniacal nitrogen content, with higher values for conventional urea, but not affecting the pH. The addition of urea to sugarcane silage reduced gas losses and raised the levels of crude protein, ether extract, and effective degradability. Objective: To evaluate the influence of fetal bovine serum (FBS) and bovine serum albumin (BSA) concentrations on the recovery and in vitro maturation (IVM) of bovine oocytes. Methods: The study was divided into Stage 1 (oocyte recovery), and Stage 2 (IVM). In the first stage, three experiments were conducted according to the recovery (R) medium used: (R1) 10 vs. 20% FBS; (R2) 5 vs. 10% BSA; and (R3) the best results from R1, R2, and the combination of FBS+BSA (5+5%). Within the second stage, the maturation medium was supplemented according to three experiments: (M1) 5 vs. 10% FBS; (M2) 0.4 vs. 0.8% BSA; and (M3) better results of M1, M2, and the combination of FBS+BSA (5+0.8%). Results: In Stage 1 (R1 and R2), the media with 10% FBS and 10% BSA showed better oocyte quality results and were defined for experiment R3. In R3, the 10% FBS and the combination of FBS+BSA (5+5%) allowed recovery of better-quality oocytes. In Stage 2 (M1 and M2), media with both levels of FBS (5 and 10%) and 0.8% BSA were defined as better according to the maturation and viability rates of cumulus cells, so they were defined for experiment M3. In M3, no difference was noted among the supplements. Conclusions: For oocyte recovery, 10% FBS and the combination of FBS+BSA (5+5%) can be used to obtain immature oocytes.
