Materials and Methods: A total of 110 GA patients hospitalized from January 2016 to September 2019 were selected as observation group, with 55 at remission stage (observation group I) and 55 at acute attack stage (observation group II). Another 55 healthy volunteers were selected as control group. The correlations of IL-1β and COX-2 with uric acid (UA) were explored by Spearman’s analysis. IL-1β and COX-2 expression levels were compared at different time points after treatment. Results: IL-1β, COX-2 and UA levels of patients were significantly higher in observation group I and II than those in the control group, and higher in the observation group II than those in observation group I. IL-1β and COX-2 were positively correlated with UA (P<0.05). ROC curve analysis showed that the optimal cut-off values of IL-1β and COX-2 for predicting GA were 18.23 and 9.12 pg/mL, and AUC values were 0.702 and 0.714 (P<0.001), respectively. On admission and after treatment for 3, 7, and 10 days, IL-1β and COX-2 levels of observation group II significantly exceeded those of observation group I (P<0.05). Compared with the efficacy group, after treatment for 14 days, IL-1β and COX-2 levels in serum and synovia significantly increased in the non-efficacy group (P<0.05). Inflammatory response variables in urine and blood tests can help rule out other disorders and support a clinical diagnosis of acute appendicitis (AA), but there is no specific blood test that can diagnose AA. In order to address this, our study sought to establish the value of (NLR) as a prognostic and severity predictor in patients with AA. Material and method: We conducted an observational retrospective study of 356 patients who had undergone surgery for AA, assessing NLR as an outcome metric. The NLR was estimated using the differential WBC (white blood count) reported on admission; the data obtained were then recorded in a database and statistically analyzed. Results: The NLR medians correlated (p<0.001) in uncomplicated AA, AA with localized peritonitis, and AA with generalized peritonitis (Kruskal-Wallis test). The ROC curve identified 81.4 % sensitivity, 62.5 % specificity, and a 73 % AUC in localized peritonitis. The NLR value was associated with generalized peritonitis with a sensitivity of 63.7 %, specificity of 64.3 %, and AUC of 68.2%. The NLR-patient death association had 100% sensitivity, 80.3% specificity, and 93.5 AUC. Conclusion: NLR determination in individuals with acute appendicitis may be useful in predicting complications. NLR values greater than 7.86 are usually related with generalized peritonitis, while values larger than 12.9 may predict an increased risk of patient death. MC3T3-E1 cells were randomly divided into control, strain loading, LF and strain loading + LF groups. Alkaline phosphatase (ALP) activity was measured. Cytoskeletal morphology was measured by rhodamine-phalloidin staining. Formation of mineralized nodules was observed by alizarin red staining. The expressions of differentiation-related genes type I collagen (COL-1), interleukin-6 (IL-6) and osteocalcin (OCN) were determined by RT-PCR, and those of p-Runx2 and p-ERK1/2 proteins were detected by Western blotting. The number of ALP positive cells and expressions of OCN, COL-1 and IL-6 were significantly elevated (P<0.05). The optical density of strain loading + LF group was higher than those in strain loading and LF groups after incubation for 4 and 7 days (P<0.05). The cell volume and extension range were elevated in strain loading + LF group compared with those in strain loading group. The amount of mineralized nodules in strain loading + LF group was significantly higher than those in strain loading and LF groups, while it was slightly higher in LF group than that in strain loading group. The expressions of p-ERK1/2 and p-Runx2 in strain loading + LF group exceeded those in strain loading and LF groups (P<0.05). Application of next-generation sequencing technology generated a massive amount of information on the gut microbiome composition used to understand its role in the healthy state and in various diseases. We aimed to provide information on the gut microbiota composition of Romanian subjects diagnosed with ankylosing spondylitis, an immune-mediated arthropathy linked to a genetic predisposition and gut dysbiosis. Methods. Stool samples collected from 25 patients with ankylosing spondylitis and 16 healthy controls were investigated using high-throughput DNA sequencing of 16S rRNA amplicons from seven different hypervariable regions and Ion Torrent PGM instrument. Microbial composition of metagenomic data was analyzed with QIIME software and differential abundance analysis of taxa encompassed linear discriminant analysis effect size (LEfSe). Results. Overall, 14 phyla, 114 families, 114 genera, and 275 species were identified across the 41 samples, the aggregated data revealing as most abundant the phyla Bacteroidetes, Firmicutes, and Proteobacteria, the families Bacteroidaceae, Prevotellaceae, and Ruminococcaceae, the genera Bacteroides, Prevotella, and Faecalibacterium, and Prevotella copri species. Using various cutoffs for abundance and prevalence, core taxonomic members were identified which in general were shared between the patients and controls. However, evidence was gained that the diversity in the microbiomes from the former cohort was lower than for controls and that certain taxa had significantly different abundance between the two groups. Conclusion. Acute kidney injury (AKI) is one of the common clinical symptoms, it is not clear whether complement system-mediated signaling pathway is involved. This study demonstrated that the expressions of complement C5a and C5a receptor (C5aR) protein in a mouse model with glycerol induced AKI were significantly increased, and the expression of inflammatory cytokines, such as IL-1β, IL-6 and TNF-α, were significantly higher than those in the blank control group. While C5aR antagonist (C5aRa) was added, western analyses for C5a and C5aR were reduced, meanwhile, qPCR and ELISA data showed that inflammatory cytokines also decreased significantly. In addition, preliminarily explored, the Mitogen-activated protein kinases (MAPKs) can be activated by the C5a/C5aR pathway in an AKI mouse model which showed that the C5a/C5aR pathway in a mouse model group activated ERK1/2 and p38, and the protein expression decreased when C5aRa was added. Methods: A total of 120 eligible patients were selected as VAP group, and 60 other patients without VAP were selected as control group. According to APACHE II scores, VAP group was divided into low-, medium-, and high-risk groups. The correlations of PCT, STREM-1, and CPIS with APACHE II score were analyzed. The values of PCT and STREM-1 combined with CPIS for evaluating prognosis were analyzed using ROC curves. Results: In the VAP group, serum PCT and STREM-1 levels and CPIS after mechanical ventilation significantly exceeded those of the control group and before mechanical ventilation (P<0.05). These values were higher in the high-risk group than in medium- and low-risk groups (P<0.05). They were positively correlated with APACHE II score (P<0.001). These values of the death group were significantly higher than those of the survival group 1, 3, 5, and 7 d after treatment (P<0.05). AUC of PCT and STREM-1 combined with CPIS for assessing prognosis was 0.892 (P<0.001). Conclusions: Serum PCT and STREM-1 levels and CPIS significantly increase in elderly patients with VAP, which rise with aggravation. In this study, we aimed to investigate the effects of resveratrol on the sphingosine-1-phosphate (S1P) and oxidative stress biomarkers in hearth ischemia-reperfusion (I/R). Materials and Methods: The biochemical and histopathological effects of RSV on cardiac ischemia-reperfusion injury were investigated through ELISA- and light microscope. Results: We observed statistically significant differences between the treatment group and the control group in terms of malondialdehyde (MDA) level, catalase (CAT) and superoxide dismutase (SOD) activities (p<0.05). Histopathologically, we also observed decreased Polymorphonuclear Leucocyte (PMNL) infiltration, myocardial edema, miyositolysis in the treatment group compared to the I/R and sham groups. Materials and Methods: Gastric carcinoma and adjacent normal tissues were harvested from 60 patients. Claudin-1 expression was detected by RT-qPCR. The expressions in human gastric carcinoma MKN45, SGC7901 and MKN28 cells and immortalized human gastric epithelium GES-1 cells were determined by RT-qPCR and Western blotting. Claudin-1 was overexpressed in SGC7901 cells by lentiviral transfection, and they were divided into Control (untransfected), normal control (NC) (transfected with lentiviral vector) and Claudin-1 (transfected with Claudin-1 overexpression lentivirus) groups. The proliferation, invasion and migration of gastric carcinoma cells were detected through cell counting kit-8, Transwell and wound healing assays, respectively. The effects of Claudin-1 on the expressions of epithelial-mesenchymal transition (EMT) marker proteins E-cadherin and N-cadherin were detected by Western blotting. Ten 4-week-old male BALB/c nude mice were subcutaneously injected with lentivirus-treated SGC7901 cells to establish the transplanted tumor model, and the effect of overexpression of Claudin-1 was explored. Results: The expression of Claudin-1 in gastric carcinoma tissues was significantly lower than that in adjacent tissues (P<0.05). Overexpression of Claudin-1 significantly inhibited the proliferation, invasion and migration of SGC7901 cells, increased the expression of E-cadherin, and decreased that of N-cadherin (P<0.05). Overexpression of Claudin-1 in the mouse model significantly inhibited the growth of subcutaneous transplanted tumors (P<0.05). Conclusion: Claudin-1 has low expression in gastric carcinoma tissues. Roman","doi":"10.2478/rrlm-2021-0033","DOIUrl":"https://doi.org/10.2478/rrlm-2021-0033","url":null,"abstract":"Abstract Introduction: Although obesity and its biomarkers have been intensively studied, little is known about the metabolomic signature of visceral adiposity independent of insulin resistance that frequently accompanies increased levels of visceral fat. Our study aimed to investigate specific changes in amino acid (AA) levels as biomarkers of increased visceral adiposity independent of insulin resistance, in healthy subjects. Methods: Forty-two adult women were included in this cross-sectional study. Serum samples were analyzed by AAs targeted metabolomics according to their visceral fat area (<100 cm2 and ≥100 cm2). Results: By corrected t-test and supervised partial least-squares discriminant analysis (PLS-DA) we identified 4 AAs that were significantly higher in the group with higher visceral fat: proline (variable importance in the projection [VIP] predicted value: 1.97), tyrosine (VIP: 2.21), cysteine (VIP: 1.19), isoleucine (VIP: 1.04; p-values <0.05). Also, glycine was significantly lower in the group with higher visceral fat (VIP: 1.65; p-value <0.05). All AAs identified were associated with visceral fat independent of homeo-static model assessment for insulin resistance (p-value for regression coefficients <0.05).
