The bacteria was grown in medium containing minerals and vegetable oils (virgin olive oil, virgin coconut oil, mustard oil and sunflower oil) as sole carbon sources. The bacteria growth was measured as optical density at =600 nm. The lipase production was determined as lipase activity of crude extract toward ρ-nitrophenyl palmitate as a substrate. The results showed that the vegetable oils used provided the good growth and lipase production of L.garvieae. Even though, olive oil had a great inductive effect on bacterial productivity. The addition of 1% olive oil improved the lipase activity by 135% compared to without olive oil. Crude extract with lipase activity of 56.790 66.571 U/mL produced by L. garvieae when cultivated in medium containing 1 5% olive oil for 20 hours. Enzyme activity was measured based on the amount of reducing sugar that produced by the DNS (Dinitrosalycilic Acid) method on a wavelength of 540 nm spectrophotometer. The results showed that the best substrate for cellulase production on coconut fibers substrate with an incubation period of 7 days which had the highest enzyme activity value of 1.340 U / ml. Coconut fibers contain the highest cellulose content compared to the other substrates. The lowest activity was shown by coconut fibers substrate at day one incubation time of 0.660 U / ml with the reducing sugar content of 0.594 mg/ml and protein content of 0.147 mg/ml. At present the productivity of Indonesian oil plam plantation, especially small holderd is relatively low. Efforts to increase productivity can be done through biochemicals approach in this case enzymes that responsible for oil production. One of the bottle neck for oil production is the step converting acetyl co-A to malonyl co-A by acetyl co-A carboxylase (ACCs). ACCs in human is known to be activated with certain amino acid and organic acids, and divalen metals. For this reason this study was conducted to separate and study the activation of ACCs by those substances. Activity of ACCs was measured developed using HPLC method. Oil palm ACCs had 50 kDa of Biotin Carboxylase (BC) subunit dan 31 kDa of Carboxyl Transferase (CT) subunit. Separation of crude extract ACCs using dialysis and sephadex G-25 column chromatography showed the higher activity of enzyme. Besides, they also produce honey, which is very beneficial for human health. Therefore the health of honeybees is essential to note. One aspect that contributes to the health of honeybees is the balance of the gut microbiota. Lactic acid bacteria (LAB) are known to give importance to intestinal health in their hosts. This study aimed to study gut-associated LAB in Apis nigrocincta (Smith), a species of a cavity-nesting honeybee endemic to Sulawesi and Mindanao islands. The method used to obtain information about the presence of lactic acid bacteria was metagenomic profiling of the honeybee gut microbiome by targeting the V3V4 region of 16S rRNA genes using the Illumina MiSeq. In total, 83,018 reads of the microbiome comprising 749 bacterial operational taxonomic units (OTUs). Some genera belong to lactic acid bacteria, among others are Bifidobacterium, Enterococcus, Fructobacillus, Lactobacillus, Lactococcus, Leuconostoc, and Vagococcus. It may have been considered that binding specificity of lectins to some carbohydrates provokes to produce many unique biological activities, including cell agglutination, mitogenic activity, antibacterial activity and antitumor activity. The aim of this study was to determine the antibacterial effect of crude lectins fractions from red macro algae collected from the southern coast of Java island, Gunung Kidul Regency, Yogyakarta Indonesia. The.Crude lectin fraction showed no antibacterial activity against gram-negative bacteria because most of bacteria cells surface contain of mannan glycoprotein and the sample has no specificity to bind this glycoprotein type.
