Endophytic fungi associated with medicinal plants become promising sources because of their ability to produce medical bioactive. This study aimed at bioprospecting fungal endophytes from an ethnobotanical medicinal plant, Pometia pinnata J. R. Forst & G Forst for their antimicrobial activity. Endophytic fungi isolated from leaf, stem, and root aseptically. Ten selected endophytes were cultured and then extracted using ethyl acetate. Screening antibacterial activity of ethyl acetate extract carried out by paper disk diffusion assay against Escherichia coli ATCC 25922. TLC-bioautography assay was used to study the type of secondary metabolite which has antibacterial activity. Identification of endophytic fungi based on morphology and ITS sequence using ITS1 and ITS4 primers. The result showed that two isolates have antibacterial activity but only one isolate exhibits strong antibacterial activity with a diameter of inhibition zone 21 mm. The isolate was identified as Aspergillus minisclerotigenes with 100% identity of the ITS sequence using BLAST software on NCBI. Wedhastri","doi":"10.2991/absr.k.210810.036","DOIUrl":"https://doi.org/10.2991/absr.k.210810.036","url":null,"abstract":"An osmotolerant rhizobacteria (Enterococcus flavescens), isolated from the weed rhizosphere, has been subjected to osmotic stress in broth media supplemented with varied concentrations of glucose and sodium chloride (NaCl). The aims of this work was to evaluate the cells’ response and obtain a comprehensive view of metabolites profiles under osmotic stress. Cells were grown in rich medium (Luria Bertani, LB) supplemented with NaCl at varying concentrations (0.5 M – 1.8 M NaCl) and glucose (5% – 20%), both as a single or double stress condition. Physiological response was observed by measuring rhizobacterial growth and analysing metabolite synthesised using GC-MS. The results of this study demonstrated that different concentration of salt and glucose resulted in different metabolite profiles. It was also observed that cells cultivated in LB + 1,6 M NaCl and LB + 1,8 M NaCl medium resulted in only two different compounds detected, i.e amide and fatty acid. Under double stress, however, concentration of hexadecanoic acid, hexadecenoic acid, proline and glycerin was found increased, while under single-salt and glucose stress, hexadecanoic acid and 9octadecenoic acidmethyl ester were detected. Those two acid were absent from the cells’ metabolites under double stress. The results of this study thus suggested that osmotic stress provokes different pathway of fatty acid metabolism. The metabolome profile of the rhizobacteria under stress condition may also reveal specific mechanism of microbial tolerance to osmotic stress under different condition. Bacillus bacteria were isolated from GIT (duodenum, jejunum, and ileum) on Trypticase Soya Agar (TSA+5% NaCl). The dilution was heated at 80°C for 20 min to kill other bacteria and induced Bacillus spore. Screening method based on the ability to against pathogens, then microbiological and biochemical characteristics. Inhibition test against pathogen using a well diffuse method. Thirteen Bacillus isolates then continued for testing against Escherichia coli FNCC 0091 for bacteriocin production purposes. Biochemical identification using conventional identification. The selected Bacillus bacteria, which had the highest inhibition zone, were isolates 2, 3B, and 11A (23.7, 18.6, and 19.9 mm). Biochemical and microbiological identification revealed that the isolate 2 isolated from duodenum was mixed culture (Bacillus subtilis, and Bacillus sp.), isolate 3B isolated from jejunum was mixed culture (Bacillus subtilis, Bacillus sp., and Bacillus sp.) and isolate 11A isolated from ileum was pure culture (Bacillus subtilis). But nowadays, the use of AGP as a growth promoter has been banned because it has negative impacts such as antimicrobial resistance. Campylobacter jejuni is a pathogenic bacterium that often infects birds, especially broilers. This research aims to investigate the role of synbiotic composed of a prebiotic, and Saccharomyces sp. and Lactobacillus sp. as probiotics (commercial products) in performance of broilers challenged with C. jejuni as a substitute for AGP. Twenty-one Day Old Chicks (DOC) of Cobb-strain broilers were divided into three groups, each consisting of 7 chicks. Group I was given basal diets, group II was given basal diets and AGP (with 250 g/ton of enramycin), and group III was given basal diets and synbiotic (100 gram/100 kg). C. jejuni challenge test was carried out at the 3 week. The results suggested that at week 4, group III showed more and significantly different body weight from that of group I, but it did not differ significantly from that of group II. Similarly, at week 4 the weight gain of group III was higher and significantly different compared to that of group I, but did not differ significantly from that of group II. Also, the carcass percentage at week 5 did not differ significantly among the groups, but group III had a higher percentage of carcasses than groups I and II.
