Eight weeks old Swiss strain male mice ( Mus musculus ) were divided into five groups: (A) control, (B) nephrolithiasis, (C) Calcusol TM , (D) nephrolithiasis & Calcusol TM , and (E) nephrolithiasis & Calcusol TM simultaneously. Nephrolithiasis was induced by applying porang tuber ( Amor-phophallus muelleri ) flour 0.06 mg/g of body weight during 3 months. Calcusol TM is traditional medicine, made of tempuyung leaves extract with S accharum lactis as additional substance. The dosage for Calcusol TM treatment was 3.3 mg/g of body weight. After 3 months treatment, the mice were killed by neck-dislocation, the kidneys were isolated and prepared for paraffin histology. CuZnSOD was analyzed by immunohistochemistry (IHC), using rabbit policlonal antibody anti-SOD1 (Bioss, bs-1079R) as primary antibody. Tissues were observed under Olympus BX51, 400x magnification. Images were documented with Olympus Digital Camera DP20. The histology images were analized in Immunoratio software online (URL: http://153.1.200.58:8080/immunoratio/) to receive the percentage of number of cells expressing CuZnSOD. The result showed that Calcusol TM administration could decrease number of cells expressing CuZnSOD in kidney significantly (P<0.05). It was supposed that antioxidant content in Calcusol TM could scavenge ROS directly with no induction of CuZnSOD production in cell.","PeriodicalId":31368,"journal":{"name":"Journal of Biological Researches","volume":"53 1","pages":"26-31"},"PeriodicalIF":0.0,"publicationDate":"2014-10-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"74985602","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Leaves of 40 individuals were collected from 4 soursop populations in Central Java and East Java, include : Sukoharjo, Karanganyar (Central Java), and Ngawi, Pacitan (East Java). Genomic DNA was extracted from the leaves by the CTAB extraction procedure with some modifications. A total of 15 RAPD primers were purchased from commercial source and tested to find specific diagnostic markers for each individuals by RAPD-PCR. The measurement of soursop population genetic distance was based on similarity coefficient using method of Group Average Clustering and Unweight Pair Group Method Arithmetic (UPGMA) of NTSYS program version 2.02i. Results showed that each soursop population collected from different localities seemed have variability in RAPD profiles by using different primers. Four RAPD polymorphic primer was selected from 15 RAPD primers, namely A18, A20, P10 and P11. A total of 58 bands produced, varying from 9 to 20 bands per primer. The selected four RAPD primers produced 57 polymorphic bands, whereas polymorphism for each primer ranged from 95 % to 100 %. Dendrogram indicated that four soursop populations tend to segregate form two separated clade. The sample collected from Sukoharjo formed a separate cluster while the sample collected from Ngawi, Pacitan and Karanganyar grouped together in other cluster and diverged from population Sukoharjo.","PeriodicalId":31368,"journal":{"name":"Journal of Biological Researches","volume":"18 1","pages":"15-19"},"PeriodicalIF":0.0,"publicationDate":"2014-08-08","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"75713605","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The data for the alpha-patchouli alcohol isomer (CD521903, CD442384, and/or CD6432585) Pogostemon Herba were explored from the pubchem database. Molecular interaction studies with COX-1 and COX-2 from mouse were done using the molecular docking tools Hex 6.12 and LeadIT2 Bisolve. The analysis of the alpha-patchouli alcohol compounds of patchouli oil showed that alpha-Patchouli alcohol (CD521903) binds to COX-1 at active sites including: LEU223B, ASP228B, LEU237B, ARG 332B, TRP 138A, GLU 139A, SER 142A, ASN 143A, and the interaction to COX-2 at active site including: GLN 289B, GLU 290B, ARG 222B, LYS 211B, THR 212B, HIS 214B, ASN 382B, HEM682B, GLN 454B, HIS 386B, TRP 387B, HIS 388B, VAL 274B, GLN 203B, VAL 291B, VAL 295B. Protoplasts are cells that have eliminated the cell wall, but the cell membranes and organs can still function properly. Pichia manshurica is one of indogenous yeast that derived from Dahlia’s plants. The success rate protoplast isolation was determined by various fac tors, include the age of the culture and the used of lytic enzymes. The purpose of this research is to get the perfect age of yeast culture that is ready to be harvested and also to get the appropriate concentration of Glucanex lytic enzymes which used for protoplast isolation. The yeast of Pichia manshurica grown on YPD broth medium and growth observed in turbidimetry. Observation of the growth of yeasts performed every 6 hours for 42 hours. Glucanex lytic enzyme concentration used for the isolation of protoplasts is 0 mg / mL (L0 = control), 2 mg / mL (L2) and 4 mg / mL (L4).
