They should be differentiated from other types of mesenchymal tumours. GISTs harbour specific activating mutations in the KIT or platelet-derived growth factor receptor a (PDGFRA) receptor tyrosine kinases, which makes them responsive to pharmacologic inhibitors. The advances in the identification of GISTs, by its molecular and immunohistochemical basis, have led to a better understanding of the rare gastrointestinal tract tumours with predictable behaviour and outcome, replacing the older terminologies like leiomyoma, schwannoma or leiomyosarcoma. There are two main varieties of β thalassaemia, β0 thalassaemia, in which no β globin is produced, and β+ thalassaemia, in which some β globin is produced, but less than normal. As the patient is diagnosed, regular blood transfusions and chelating treatment is given lifelong. In this study, we observed the haematological profiles of multi transfused thalassemia major patients with the accompanying complications. Aims and Objectives: To study the clinical features with RBC parameters, serum ferritin levels, chelation therapy and prevalence of blood transfusion related infection in multi transfused beta thalassemia major patients. Materials and Method: In this cross sectional study, total 90 diagnosed thalassemia major patients were taken with their complete history, physical examination and laboratory parameters such as-complete blood counts. All the patients were monitored for serum ferritin levels and chelation therapy along with screening for transfusion transmitted infections over a year. All the data were compiled and appropriate statistical analysis was done. Results: In present study out of 90 patients, 54 patients were males and 36 patients were females. Mean age of 15.26 ± 7.04 years with 68.9% (N=62) of cases required twice monthly transfusion, 30.0% (n=27) cases required once a month transfusion. 25 cases were found to be HCV+ (27.78%), 3 were HIV + (3.3%) and 62 cases were n on-reactive (68.89%). On first post transfusion CBC we found that 30 cases (33.33%) were found to be adequately transfused with levels > 10 gm%, where as in second results 34 cases (37.78%) were found to be adequately transfused with levels >10 gm%. Mean of average yearly average ferritin values in the study duration was found to be 4724 ± 287.65 ng/mL. The chelating agent dose was with mean 1040 mg and standard deviation 476.19. Micro RNA 21 (mi-RNA 21) has been identified as oncogenic RNA, implicated in etio-pathogenesis of various neoplastic lesions. Objective: (i) To assess the microRNA 21 expression in oral cancer patients with history of tobacco chewing habits and (ii) to carry out its clinicopathological correlation. Material and Method: The present study was carried out as a case-control study, for a two year period in which a total of 20 biopsy tissue specimen obtained from oral cancer patients having tobacco chewing habits were enrolled as cases and an equal number of biopsy tissue specimen obtained from age, and sex-matched patients of oral leukoplakia were enrolled as controls. Clinical details, personal habits were noted and histopathological assessment was done. miRNA21 was done using Taq Man microRNA assay (ABI) using Polymerase Chain Reaction (PCR).Real time PCR results was analyzed and expressed as relative miRNA expression of the threshold cycle (CT) values. Mean values were compared using independent samples ‘t’-test and ANOVA using SPSS 21.0 software. p-value less than 0.05 indicated a statistically significant association. Results: Mean age of cases was 52.65±7.82 years, majority of cases were males (85%). Among cases, majority (70%) had clinical stage I and were histo-pathologically well differentiated (75%). There were 3 (15%) cases with metastasis. Mean relative miRNA 21 expression were significantly higher in cases (10.53±3.52) as compared to that in controls (8.76±1.16) (p=0.029). Relative miRNA21 values showed a significant increasing trend with increasing clinical stage, histopathological grade and metastasis. We present a case of late melanoma recurrence that manifested as an in-transit metastasis in the iliac bone, occurring two years after the excision of the tumour. In the present case report; A 59 year female was reported to the outpatient department of the medical college & associated hospital. The patients past medical history stated that she is known case of foot melanoma diagnosis and was operated in 2019. In July 2020, the patient developed recurrence in July 2020. A wide local surgical excision to achieve a 2-centimeter margin with skin graft was undertaken with an associated sentinel lymph node biopsy. Upon follow-up, it was discovered that the patient had been disease-free for a full year. However, the lesion started to reoccur in the period of July 2022. A PET-CT of her whole blood was performed. The patient received IV fluids, antibiotics, analgesics, and other supportive care before being released once the hemodynamics were determined to be stable. A 10-year period of disease-free living cannot usually be regarded as a definitive cure for melanoma. Loss of caspase-3 expression and activity has been reported in primary human breast cancers. It is suggested that it might promote tumour development by preventing and reducing senescence. It was a retrospective observational pilot study aimed to evaluate the prognostic potential role of caspase-3 in breast cancer patients. Method: It was a retrospective observational pilot study in which fifty breast cancer patients were enrolled. Haematoxylin and eosin-stained slides of the cases were screened to obtain the best paraffin block for IHC, on which immunohistochemical expression of caspase-3 marker was done and studied. It was correlated with the known prognostic markers of breast cancer. Results: Out of 21 cases with moderate caspase-3 staining, 85.7% cases were grade III and 14.2% were grade II. From 16 cases with strong caspase-3 staining, 75% cases were grading III followed by 25% cases of grade II. There was no significant correlation between caspase-3 staining with histological grade, size of tumour, lymph node involvement, ER, PR and Her2/neu status (p=0.68, 0.24, 0.20, 0.68, 0.45 and 0.11 respectively). Correlation of survival was significantly associated with lymph node involvement (p=0.01) while it was insignificant for tumour size and histological grade (p=0.36 and 0.42 respectively). Conclusion: The survival of breast cancer patients was correlated only with lymph node metastasis. It was better with smaller number of lymph node involvement. While, Caspase-3 did not have any direct association with prognostic markers of breast carcinoma like tumour size, grade of tumour, lymph node and ER, PR and HER-2/neu status. Lung carcinomas are broadly divided into small cell carcinoma and non-small cell lung carcinoma (NSCLC). Non-small cell lung cancer accounts for 80% of lung cancers and lung adenocarcinoma is one of the main types of NSCLC. Here we studied the significance of the commonly used lung adenocarcinoma diagnostic markers, including TTF-1, Napsin A and CK7, in the diagnosis of lung adenocarcinoma using immunohistochemical (IHC) analysis. Aim: To evaluate and compare the significance of IHC markers, CK 7 and Napsin A in diagnosis of lung adenocarcinoma as far as specificity and reliability are concerned. Material and Methods: This study was conducted over a period of 5 years, from 2018 to 2022 which included all cases diagnosed as lung carcinoma on tissue biopsies and later subjected for IHC using relevant markers like CK7, TTF-1, Napsin-A, CK5/6, p63, Synaptophysin and Chromogranin A. 47 cases of lung adenocarcinoma were selected from the retrospective database of lung cancer. Results: Among the 47 cases of lung adenocarcinoma. IHC was done on 41 cases (87.23%). Immunocytochemistry (ICC) was done on 6 cases (12.8%). Lung adenocarcinoma IHC markers study shows CK 7 positive and Napsin-A negative in 21 cases (51.2%). CK 7 negative and Napsin-A positive only in 1 case (2.4%). Both are positive in 16 cases (39.0%) and both were negative in 3 cases (7.3%). ICC was studied in total 6 cases which show CK 7 positivity in 4 cases (66.7%). Only 2 case of ICC show Napsin-A positivity (33.3%).
