A high amount of punicalagin in the pomegranate peel allows the pomegranate to produce antiinflammatory, antioxidant, antiapoptotic effects. In this study, it was aimed to investigate the protective effects of PPE in LPS-induced uveitis model in rats.\u0000Material and Methods: 60 male rats were divided into;healthy,uveitis,LPS+DEX, LPS+PPE(100mg/kg),LPS+PPE(300mg/kg),LPS+PPE (500mg/kg). Analyzed PPE was given to rats by oral gavage for 7 days before LPS administration. One hour after the last extract, LPS was applied to rats in all groups except the healthy group. DEX was applicated intraperitoneally 1 mg/kg x three times, half an hour before, at the same time and half an hour after the LPS application. After LPS application, half of the rats in each group at the 3rd and the other half at the 24th hour was taken eye tissues of rats and were stored for histopathological, molecular and biochemical analysis.\u0000Results: PPE administration significantly decrease MDA levels and a significant increase in GSH levels and SOD activities depending on the dose, compared with the uveitis group. {"title":"CHARACTERIZATION of cox3 and rnl GENES ENCODED IN MITOCHONDRIA of Fusarium graminearum","authors":"Aylin Gazdağlı, G. Albayrak","doi":"10.23902/trkjnat.1144980","DOIUrl":"https://doi.org/10.23902/trkjnat.1144980","url":null,"abstract":"In this study, the phylogenetic relationship among Fusarium graminearum isolates was established for the first time based on mitochondrial cox3 and rnl genes’ variations. The cox3 and rnl were amplified from 45 isolates purified from Turkey and Iran together with 2 Korean strains by polymerase chain reaction. The amplicons were sequenced and nucleotide polymorphisms were detected by alignment. The phylogenetic relationship was constructed by using PAUP 4.0a with the maximum parsimony method. Fragments with 477 bp length, belonging to cox3, were obtained from 46 isolates; 1547 bp-amplicons of rnl were produced from 45 isolates. Sequence similarities were calculated as 30-100 % and 17-94 % for cox3 and rnl, respectively. Nucleotide variations within the rnl was found higher than within cox3. It has been shown that SNPs and in-dels, found in coding regions, cause a codon change and may alter more than one codon by causing frame shift without affect gene’ functions. Bootstrap values belonging to cox3 and rnl dataset was found ranging from 57 to 84 %, and 54 to 100 %, respectively. Parsimony analysis revealed that Korean isolates were in monophyletic relationship with Turkish and Iranian isolates. Development of an appropriate reconstructed epidermis model might overcome the limitations of monolayer culture systems. To that end, in the current study we used the immortalized human keratinocyte cell line, HaCaT, to develop an ex vivo 3D cell culture system for keratinocyte-based toxicity analysis. Mouse embryonic fibroblast-conditioned medium and Matrigel matrix-based 3D HaCaT cell culture systems expressed skin-related genes and proteins in culture. The 3D HaCaT cultures demonstrated a skin-like phenotype and response against selected test compounds. Reliable results were obtained compared to monolayer HaCaT cells which were exposed to selected chemicals for 1 h and 24 h. Gene expression profiles of 3D HaCaT cell cultures and monolayer cultures were completely different after administration of the test compounds. \u0000Overall, our results showed that a 3D HaCaT cell culture system generated in Matrigel matrix exerted a skin epidermis-like phenotype. In conclusion of the study, a new species of the genus Lathrobiuum Gravenhorst, Lathrobium gocmeni sp. n. (Edirne, Enez) is described, illustrated, and distinguished from congeners. Astenus thoracicus (Baudi di Selve, 1857), A. procerus (Gravenhorst, 1806), Lobrathium rugipenne (Hochhuth, 1851), Micrillus testaceus (Erichson, 1840), Medon fusculus (Mannerheim, 1830), Paederus littoralis, Gravenhorst, 1802 and, Scopaeus gracilis (Sperk, 1835) from Thrace and Lathrobium fulvipenne Gravenhorst, 1806 from the Marmara Region are reported for the first time. {"title":"Characterization of differential expression patterns of the extracellular purinergic enzymes in colorectal cancer","authors":"S. Additionally, tumor vs stromal cell expression of the target genes was analyzed from publicly available patient expression datasets. Finally, the correlation between CD39 and CD73 expression with patient prognosis was analyzed via TCGA datasets. \u0000Results: In CRC cell lines CD39 found to be not expressed at all while CD73 was expressed extensively in most cell lines via WB and RT-CR analyses. Patient microarray expression data confirmed the results from CRC cells lines that CD39 expression was very low in epithelial/tumor cells relative to other stromal cell types yet CD73 was expressed abundantly in every cell type within patient tumor samples. Interestingly, CD39 expression in patient tumors was correlated with favorable prognosis while CD73 expression was associated with worse prognosis. \u0000Conclusion: Although CD39 and CD73 are related enzymes involved in extracellular purinergic signaling, their expression patterns in tumor cells and prognostic effects in patients show opposing outcomes. Hücre içi trehaloz miktarının düzenlenmesinde trehalozun sentezi ve yıkımı önemlidir. Bu nedenle, TPS1 ve NTH1 gen ekspresyonları transkripsiyon ve translasyon sırasında sıkı bir şekilde düzenlenmektedir. Her iki genin promotor bölgesinde Stres Tepki Elementleri bulunduğundan stres koşullarında birlikte aktive olurlar. Ancak her iki genin promotor bölgesinde benzer düzenleyici elemanların bulunması bu genlerin transkripsiyon seviyesinde farklı bir regülasyona uğradıklarını göstermektedir. Çalışmamızda, besin açlığında TPS1 ve NTH1 genlerinin transkripsiyonel düzenlenmesinde SAGA kompleksinin rolü analiz edildi. Ayrıca yaban tip ve mutant maya hücrelerinde trehaloz seviyesi enzimatik olarak tespit edildi. TPS1 ve NTH1 genlerinin in silico promotor analizi sonucunda Msn2/4 transkripsiyon faktörlerinin bağlanması için gerekli olan STRE dizilerinin NTH1 promotorunda nükleozomlar tarafından kapatıldığı TPS1 promotorunda ise açıkta kaldıkları belirlendi. Ada1 proteininin yokluğunda, TPS1 geninde stres kaynaklı promotor aktivasyonu gözlenirken, NTH1 geninde promotor aktivasyonu gözlenmedi. Bu sonuçlara göre, Ada1 proteininin yokluğunda STRE dizilerini kaplayan nükleozomlar mobilize edilemediğinden Msn2/4 transkripsiyon faktörleri stres koşullarında promotora bağlanamayarak transkripsiyonu aktive edememiş olabilir. The description took into account the stages of formation of the vegetative component of the inflorescence, the most significant features of its structure, the number of flowers in the inflorescence, flowering dates. Abundance and level of flowering are estimated. In the research conditions generative individuals of R. typhina bloom annually. In this study, a novel ensemble technique was proposed for the prediction of PM2.5 concentration in cities with high traffic noise using traffic noise as an input parameter. Air pollutants concentration (P), meteorological parameters (M) and traffic data (T) simultaneously collected from seven sampling points in North Cyprus were used for conducting the study. The modelling was done in 2 scenarios. In scenario I, PM2.5 was modelled using 4-different input combination without traffic noise as input parameter while in scenario II, traffic noise was added as an input variable for 4 input combinations. The models were evaluated using four performance criteria including Nash-Sutcliffe efficiency (NSE), root mean square error (RMSE), correlation coefficient (CC) and bias (BIAS). Modelling PM2.5 with combined relevant input parameters of P, M and T could improve the performance of the model developed with only one set of the parameters by up to 12, 17 and 29% for models containing only P, M and T respectively. All the models in scenario II have demonstrated high prediction accuracy than the corresponding model in scenario I by up to 12% in the verification stage. After his Primary and Secondary education in Cyprus, he graduated from Agricultural Faculty of Ankara University in 1960. He completed his PhD. thesis on Forestry Entomology with the highest academic achievement, Magna Cum Laude at George-August University, Germany, in 1966. He worked as Adjunct Associate Professor in The University of Queensland, Gatton Campus, Australia since 1973. Professor Hassan retired in 2000 when in Queensland University. He was asked to continue to give lectures in the university with Honorary degree. He provided many lectures to undergraduate and postgraduate students, supervised thesis of many M.S. and Ph.D. candidates. He was also doing consulting at both national and international levels and was developing training workshops in applied entomology.\u0000He published many articles on Plant Extracts, Essential Oils, Biopesticides, and Integrated Pest Management, and received 608 citations. Professor Hassan named a parasitoid species new for science with the name of his wife in memory of their marriage for more than 50 years.\u0000He had a good relationship with Turkish colleagues, and contributed many of them with his scientific experience. \u0000He passed peacefully on June 27, 2022. He touched the lives of so many people in Turkey, and will be remembered forever. \u0000Editor-in-Chief note: Professor Errol Hassan started to contribute to Editorial Board of Trakya University Journal of Natural Sciences since 13 November 2017 in the fields of Entomology and Agriculture. He read all submitted manuscripts in these fields with an incredible devotion and without a sign of offence, and provided criticisms to improve all. When we made a contact with Professor Errol Hassan to propose him to become the guest editor of the special issue of “Biodiversity of Insects”, he replied in the affirmative with a kind manner. He continued to contribute all submissions both in the Editorial Board of the journal and as the guest editor role of the special issue until May 2022.
