Ivanova","doi":"10.15414/JMBFS.3609","DOIUrl":"https://doi.org/10.15414/JMBFS.3609","url":null,"abstract":"Conjugated linoleic acid (CLA) is a family of more than 28 isomers of linoleic acid wherein the isomers cis-9, trans-11 (rumenic acid) and trans-10, cis-12 are the most abundant. It  is associated with a number of potential health benefits for human organism. Many foods are a good source of it but it is mosty found in meat and dairy products, derived from ruminants. Dairy products contain CLA in different amounts. The enrichment of these products with CLA is appropriate given the lower CLA content in these products in comparison with the recommended health intake. Modification of CLA concentration can be done by specific animal feeding and diet modification, by direct CLA supplementation in the form of oils or by addition of specific starter culture. The influence of technological treatments on the stability of the final product during storage and maturation is still not completely elucidated. There is a need for further studies on the physiological effects of CLA isomers on humans. Ďúranová","doi":"10.15414/JMBFS.2584","DOIUrl":"https://doi.org/10.15414/JMBFS.2584","url":null,"abstract":"Detection and tracking of microorganism’s behavior in natural environments and under stressed conditions have been nowadays accelerated by the requirement for the risk assessment associated with environmental contamination. Luminescence-based techniques, for they cost efficiency, fastness, reliability and reproducibility are particularly appropriate for such studies. We used bioluminescence sensing assay to determine ATP levels in the yeast Schizosaccharomyce pombe. Determination of adenosine concentration per cell was assessed in the yeast cells counted prior to the experiment. To test the methodology, model organism was exposed to life-threatening concentration of heavy metals cadmium and nickel for three hours. Afterwards, changes in the ATP levels were detected and compared to the control. Strikingly, our results revealed that such conditions lead to considerable increase in the production of ATP by mitochondria compared to untreated cells. Normal level of ATP in the control increased from the basal 9.92x10-16 M to 2.90x10-15 M after Cd exposure and to 7.58x10-15 M upon Ni treatment. The methodology was subsequently used for assessment of cleanness of margarine producing company devices after regular sanitation procedure. Validation characteristics (precision, diagnostic and analytic specificity, diagnostic sensitivity, relative linearity) were defined both at the beginning of the release of diagnostic set and the expiration date (as the element of stability study). The values of diagnostic sensitivity and specificity, defined via the in-process set of serums (20 positive and 50 negative) comprised 100%. The presence of other classes of Chlamydia antibodies in samples didn’t influence the results of ELISA for specific IgM antibodies. Linearity of the method was sufficient. The intra-assay variation of results is between 3.1% to 6.4%, and intra-assay precision was from 1.5% to 8.4%, staying acceptable (≤ 10%) both at the moment of release and during expiration date. ISO7932 method was used for enumeration of B. cereus, afterwards a PCR was performed to confirm the presence of cereulide toxin (ces) gene. In addition, the effect of different concentrations of chitosan and chitosan nanoparticles (CNPs) (0.05, 1%) on B. cereus was studied in pasteurized milk.The shelf life was evaluated by clot on boiling test and Ph value. Their pathological effect was detected by orally administration to experimental rats to investigate their adverse effect on rats liver and intestine using histopathological examination. The most prominent potent bactericidal effect was observed inCNPs 1%, B. cereus count reached to undetectable level at 5th day of refrigerated storage. The pasteurized milk inoculated with B. cereus, chitosan and CNPs showed some pathological lesions in rats treated with B. cereus,while the chitosan 0.5% had antibacterial activities without cytotoxic effect on rat tissues, alternatively to CNPs 1%, which exist edematous tips of intestinal villi and mild kupffer cells activation in liver.Therefore, our results contribute data that are primary to indicate the risk of food poisoning due to B. cereus and trials to control that in food by careful using of nanotechnology. We investigated effect of extract consisting mainly of polyphenols, brown algae and plant polysaccharides on the reproduction potential of male rabbits (New Zealand white breed) during 60 days long dietary experiment. The rabbits were divided into three groups. Control was fed a basal diet, whereas the second and third group were supplemented with seaweed and polyphenols mixture: T1 = 0.3% and T2 = 0.6% respectively. We observed that sperm concentration in both experimental groups increased in comparison to the control group. Results of the CASA analysis showed enhanced motility (C = 85.09±7.53%, T1 = 87.21±8.25%, T2 = 89.38±8.02%) and progressive motility (C = 74.28±12.6%, T1 = 79.07±13.89%, T2 = 81.28±11.37%) in experimental groups supplemented with combination of algae and polyphenols in comparison with the control group. While monitoring ferric reducing ability of plasma (FRAP), we found the highest value in T1 group, changes were insignificat. An increase in GPx activity was measured in experimental groups in comparison with the control group with major difference in T1 group. In experimental groups, we determined an increase in activity of superoxide dismutase (SOD) in comparison with control group, the difference was significant in T2 group (C = 0.26±0.11 U/mg TP; T2 = 0.31±0.09 U/mg TP). Traditionally it is processed through natural convective drying or sun drying of ripe mango pulp. Mango leather was produced through sun, convective (hot air), freeze, and microwave drying. Drying kinetics was studied with the help of nine empirical models used extensively in food industries. Root mean square error (RMSE), correlation coefficient (R2), and the sum of square errors (SSE) these four statistical measures were examined for nine different models to learn the best-fitted model. The Fourier-transform infrared spectroscopy study was conducted for compositional analysis of differently dried samples in the wavelength range of 400-4000 cm-1 at ambient temperature. For sensory analysis, the test panel was constructed as per ISO 8586-1. ISO 4120:2004, ISO 5496:2005, ISO 10399:2004 standards were maintained for selecting panel members. The 30 isolates analyzed in this study all belonged to clonal complexes (CC) CC1 (3; 10%), CC5 (3; 10%), CC15 (16; 53.33%) and CC152 (8; 26.66%), respectively. Of antibiotic resistance-associated genes reported; Twenty out of 30 isolates (66.66%) carried the beta-lactamase operon (blaZ/I/R) and the tetracycline resistance gene (tetK), respectively. Also, the macrolide resistance gene (ermA/msrA) were detected in ten isolates (33.33%), the multidrug efflux pump resistance gene (sdrM) was detected in seven isolates (23.33%) and one isolates (3.33%) harboured the chloramphenicol resistance gene (cat). None of the isolates harboured genes conferring methicillin resistance. In terms of genes encoding enterotoxins; seb was detected among all isolates of CC15 while the enterotoxin gene cluster egc and a distinct variant of the enterotoxin A (sea-N315) were detected among isolates of CC5 and CC152, respectively. Two CC152 isolate was positive for genes encoding the Panton-Valentine leukocidin (lukF-PV+lukS-PV). In addition, all the isolates harboured the genes encoding for intracellular adhesion proteins (icaA/C/D) and the biofilm-associated genes (clfA and clfB) except for CC1 isolates. The capsular genes cap5 and cap8 were detected in isolates of CC5 and CC15 while only cap5 was present in CC152 isolates.
