{"title":"~(125)I-β-CIT imaging study of striatal dopamine transporters in mice model of Parkinsonism","authors":"Weng Zhongfang","doi":"10.3760/CMA.J.ISSN.0253-9780.2001.02.008","DOIUrl":"https://doi.org/10.3760/CMA.J.ISSN.0253-9780.2001.02.008","url":null,"abstract":"Objective To detect the activity of striatal dopamine transporters (DAT) in lesions of different order of severity of MPTP-induced mice model of parkinsonism by autoradiography with 125I-β-CIT and to evaluate the clinical use of the β-CIT imaging for DAT detection. Methods With regard to the different duration (days) of MPTP treatment, the C57BL mice were randomly divided into 5 groups, that is MPTP 1,3,5 and 7day groups and control group treated with normal saline instead of MPTP. Two hours after intravenous administration with 125I-β-CIT of 148 kBq, the brain tissue sections were imaged by autoradiography. The levels of dopamine (DA) and its metabolites were measured by high performance liquid chromatography and electrochemical detection (HPLC-ECD). The tyrosine hydroxylase (TH)-positive cells and fibres in the substantia nigra and striatum of the mice were observed by means of immunohistochemical technique. Results As compared with control group, the radioactivity ratios of striatum to cortex (ST/CX) in 4 MPTP-treated groups were significantly reduced,by 20%, 42%, 45% and 52%,respectively. The concentrations of DA in the striatum of 4 MPTP-treated groups were remarkably decreased,by 47%, 75%, 95% and 95%,respectively. The gradual loss of DA neurons and fibres in the substantia nigra and striatum in 4 MPTP-treated groups was observed under microscopy. Conclusions The functional abnormality of DAT paralleled the changes observed in neurochemistry and neuropathology studies in the lesions of different order of injury of the MPTP-treated mice. Zhao, Xiangrui Ji, Hua Zhang, Tianfu Jiang","doi":"10.3760/CMA.J.ISSN.0253-9780.2001.02.023","DOIUrl":"https://doi.org/10.3760/CMA.J.ISSN.0253-9780.2001.02.023","url":null,"abstract":"Objective To investigate the biological effects and the effecting mechanisms of rare earth elements La,Gd and Ce on cultured rat cells.Methods The biological effects of La 3+ on cultured rat cells and the subcellular distribution of La and Gd and Ce, and the inflow of 45Ca 2+ into the cells and total cellular calcium were measured by isotopic tracing, Proton Induced X Ray Emission Analysis (PIXE) and the techniques of biochemistry and cellular biology.Results La 3+ at the concentration of 10 -10(or 10 -9)~10 -6mol/L significantly increased quantity of incorporation of 3H-TdR into DNA, total cellular protein and the activity of succinic dehydrogenase of mitochondria. The cell cycle analysis showed that the proportions of cells in S phase were accordingly increased acted by La 3+ at above range of concentration. But these values were significantly decreased when concentration of La 3+ raised to 10 -4~10 -3mol/L. It was further discovered that La,Gd,and Ce distributed mostly in the nuclei, and then in membranes. Methods 17 patients with various breast tumors underwent RII. Results 8 of 9 patients with primary breast cancer gave positive results of RII, with the sensitivity of 88.89%. Ipsilateral lymphatic metastasis was detected in 4 of 5 patients with primary breast cancer plus axillary lymph node involvement and for the remaining 1 patient contralateral lymph node metastasis was detected; the patient received mastectomy 5 years ago due to breast cancer of that side. RII results of the remaining 7 patients were negative, they were 6 patients with benign breast tumour and 1 followedup patient who underwent mastectomy 7 years ago due to breast cancer.
