We here develop a biomathematical model of the disease course by translating known biological mechanisms of virus-host interactions and immune responses into ordinary differential equations. We explicitly describe the dynamics of virus population, affected alveolar epithelial cells, and involved relevant immune cells comprising for example CD4+ T cells, CD8+ T cells, macrophages, natural killer cells and B cells. We also describe the humoral response dynamics of neutralising antibodies and major regulatory cytokines including CCL8 and CXCL10. The model is developed and parametrized based on experimental data collected at days 2, 3, 5, and 14 post infection. Pulmonary cell composition and their transcriptional profiles were obtained by lung single-cell RNA (scRNA) sequencing analysis. Parametrization of the model resulted in a good agreement of model and data. The model can be used to predict, for example, the time course of the virus population, immune cell dynamics, antibody production and regeneration of alveolar cells for different therapy scenarios or after multiple-infection events. Although the role of seasonality was emphasized previously, this aspect is rarely considered yet, and it remains elusive how this impacted hydroclimate during winters and summers across Central Europe. Here, we coupled biomarker-based δ<sup>18</sup>O and δ<sup>2</sup>H from Bergsee in southern Germany to reconstruct deuterium excess as a proxy for evaporation history from the Bølling-Allerød to the Preboreal. We compared this dataset with other biomarker isotope records in Central Europe. They are all lacking a strong isotopic depletion during the Younger Dryas, which is best explained by the summer sensitivity of the biomarker proxies: As Younger Dryas summers were relatively warm, there is an absence of the strong winter cooling signals recorded in annual water isotope records like Greenland or Lake Steißlingen. Lake evaporation at Bergsee together with other paleohydrological reconstructions draw a coherent picture of the Late Glacial hydroclimate, with strong evidence for warm and dry Younger Dryas summers. Rather than a southward shift of the Westerlies during winter, we suggest that a recently proposed feedback mechanism between North Atlantic sea ice extend, strong winter cooling and summer atmospheric blocking serves as a suitable explanation for summer dryness. Recent studies have highlighted that non-coding RNA (ncRNA) dysregulation is a notable feature in patients with SLE. As a result, this research was designed to investigate lncRNA CASC2 and miR-155 levels as non-invasive diagnostic biomarkers in SLE patients, including those with and without nephritis, and to investigate their effectiveness in assessing disease severity and predicting LN. Our study included 60 patients with SLE who were subclassified into (30 non-LN and 30 LN groups), along with 30 control subjects. Quantification of lncRNA CASC2 and miR-155 in serum samples from the Egyptian population was carried out with real-time polymerase chain reaction (RT-PCR). The disease activity index (SLEDAI) for SLE was evaluated, and the analysis of the receiver operating characteristic (ROC) curve was implemented. Increased levels of lncRNA CASC2 were observed in SLE patients compared to healthy controls, with even higher levels observed in the LN group versus the non-LN patients' group. Conversely, miR-155 was noted to be down-regulated in SLE patients relative to controls, and its levels were lower in the LN group relative to the non-LN patients' group. The elevated expression of lncRNA CASC2 and reduced expression of miR-155 were both correlated to the severity of the disease. The current conventional verification method for nowcasting uses a binary classification event verification method, which exists with double punishment, leading to low scoring issues. In order to make up for the shortcomings of conventional verification methods and explore the potential value of forecasting, based on the characteristics and requirements of strong convective weather nowcasting operations, this paper proposes a neighborhood verification method that considers spatial scale, time scale, and intensity error information simultaneously, based on the spatial neighborhood fraction skill score (FSS) verification method. The paper designs an intensity neighborhood correction scheme, a time neighborhood correction scheme, and a comprehensive correction scheme that considers both intensity and time neighborhoods. By introducing spatial neighborhood probability, the strict spatial matching requirement is weakened. Based on the Gaussian membership function in fuzzy logic, the value of forecasting grid below the threshold is explored. Time neighborhood adaptive weighting strategy is adopted to solve the problem of early and delayed forecasts.
