EFFICIENCY OF FEED ADDITIVE NIKAMAX® FOR NATURAL INFECTION OF BROILER CHICKENS WITH EIMERIA SPP. 饲料添加剂尼卡玛斯®对肉鸡自然感染埃默氏菌的效果

Perig","doi":"10.36359/scivp.2023-24-2.18","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-2.18","url":null,"abstract":"The article presents the results of the study of the effectiveness of the combined feed additive NIKAMAX® (powder for oral use), produced by JSC Biopharm (Ukraine), the active pharmaceutical ingredients of which are the polyester ionophore antibiotic maduramycin and the synthetic compound nicarbazine, in the treatment of coccidiosis in broilers naturally infected with coccidia genus Eimeria (E. tenella, E. maxima and E. acervulina). The main criteria for demonstrating the effectiveness of the feed additive NIKAMAX® were: the number of Eimeria oocysts in feces; assessment of intestinal damage; poultry mortality and morbidity associated with coccidiosis. Additional indicators of the effectiveness of the feed supplement were: average live weight; average daily increase in live weight of poultry; average daily feed consumption, the ratio of the amount of feed consumed to growth; data of bird health monitoring during the experiment. The results of the study showed that the feed additive NIKAMAX® when added to feed at a dose of 0.5 g per 1 kg of feed, or 3.75 mg of maduramycin ammonium and 40 g of nicarbazin per 1 kg of feed, causes a decrease in the degree of infestation of broilers with coccidiosis, reduces damage of the intestine of a sick poultry, reduces its mortality and morbidity associated with coccidiosis. The combination of nicarbazine and maduramycin, what are the active ingredients of the feed additive NIKAMAX®, contributed to an increase in the growth of broiler chickens and a decrease in the feed conversion ratio in the group of birds to which the additive was added to the compound feed, compared to the infected birds to which it was not administered. In the complex treatment of infectious diseases of reptiles, a fairly effective antibiotic is ceftifur, which belongs to the third-generation cephalosporins, which are widely used in veterinary medicine. To study the effect of cephalosporins on the body of reptiles, a histological and ultra-structural examination of the kidneys and liver of Red-eared slider (Trachemys scripta elegans) was performed, which were administered intramuscularly ceftifur (at a dose of 2.2 mg/kg) every 24 hours. Experimental animals were removed from the experiment on the 7th day. Histological sections were made using a sledge microtome and cryostat microtome, stained with hematoxylin and eosin, Sudan III. Electron microscopic research was carried out using an electron microscope PEM-100-01. As a result of the histological examination and transmission electron microscopy of the kidneys, dyscirculatory changes were revealed, characterized by the expansion and overflow of blood mainly in venous vessels, desquamation of endothelial cells in the vessel lumen, as well as dystrophic changes in the epithelium of the renal tubules. Structural changes in the epithelium of renal tubules were accompanied by damage to mitochondria, expansion of the tubules of the smooth endoplasmic reticulum and the accumulation of cytosomes, lysosomes, and autophagolysosomes in the cytoplasm. In the liver, a decrease in the density of melanomacrophage complexes was recorded, as well as a decrease in the number and change in the shape of fatty inclusions in the cytoplasm of hepatocytes, decrease in the number of glycogen granules. According to the results of coprological studies, the animals of the experimental and control groups were found to have spontaneously infected dogs with endoparasites, the causative agents of digestive tract nematodes: Toxocara canis, with an intensity of invasion (II) of 100 to 190 eggs in one gram of feces (ЕGF) with an average intensity of invasion of 140.9 ± 4.07 ЕGF and Ancylostoma caninum with an intensity of invasion of 36 to 54 eggs in 1 g of feces with an average intensity of invasion of 44.9 ± 1.45 ЕGF, as well as coccidia of the alimentary canal of the species Cystoisospora canis with an intensity of invasion of 580 to 750 oocysts in 1 g of feces (OGF) with an average intensity of invasion of 677.5 ± 4.07 OGF. Clinically, nematodes of the alimentary canal in dogs and puppies were manifested by: loss of appetite, depression, pallor of the mucous membranes, and in some animals vomiting and mild diarrhea, and in cystoisosporosis - depression, motility, weight loss, during defecation - feces are liquid, sometimes with impurities of mucus. After the use of the drugs \"Profilain coccid\" and \"Prokoks@ for dogs\", coprological studies were carried out with the establishment of indices of parasitological damage and determination of the intensive effectiveness (IE) and extensive effectiveness (EE) of the drugs. On the basis of the obtained results, it was established that for a spontaneous toxocarosis infestation of dogs on the 7th day after deworming, the intensity of the drug \"Profilin coccid\" was 96.2 %, and the reference drug \"Prokoks@ for dogs\" was 96.9 %. On the 14th day after deworming the animals, the intensive and extensive effectiveness of both drugs was 100%. Helminthological studies established that for spontaneous hookworm infestation in dogs 7 and 14 days after the use of the researched drug and the reference drug in the recommended doses, the intensive and extensive effectiveness of veterinary drugs was 100 %. In case of spontaneous cystoisosporosis infestation in dogs, it was established that on 3 and 7 days after oral administration of the drugs to the animals, the intensity of the studied drug was 53.0 and 85.2 %, and the intensity of the reference drug was 56.0 and 87.3 %, respectively. On the 14th, 21st, 28th, and 35th day after the use of drugs in the form of a suspension, the intensive and extensive effectiveness for cystoisosporosis in dogs was 100 %. Based on the fact that farms can accumulate a significant amount of unusable straw, the problem of effective utilization of this spoiled biomass arises. From an economic and ecological point of view, the method of recycling spoiled straw of various origins into biofertilizer by fermentation using various symbiotic conglomerates of microorganisms is effective. Regulated fermentation improves the mineralization of organic waste and increases the efficiency of straw use in bioconversion units. The problem of the effect of different doses of the domestic biodestructor on the content of microelements in composted oat, wheat and pea straw is not sufficiently studied. Samples of spoiled straw were taken for the experiments, which were fermented in the control group (without introduction of the microbiological preparation) and with introduction of the preparation in doses of 7.0; 14.0 and 28.0 cm3/t. The content of Manganese, Ferrum, Copper, Zinc, and Cobalt was determined in the straw of fermented cereals and legumes. As a result of composting of pea straw, an increase in the content of Ferrum in the biomass was found. With the use of the largest dose of the biodestructor, an increase in the Ferum content by 9.1% compared to the control one was established. The use of a biodestructor affects the growth of Zinc content in pea straw. Fermentation of this biomass at biodestructor doses of 14.0 and 28.0 cm3/t contributes to an increase in its Zinc content, respectively, by 20.9 and 25.5 % relative to the control group. With an increase in the dose of the biodestructor in the straw during its composting, the Manganese content increases in comparison with the indicator in the control group. The content of Cuprum in pea straw is affected by the dose of biodestructor during composting. The use of a biodestructor at a dose of 28.0 cm3/t leads to an increase within the limits of statistical significance in the content of Cuprum in pea straw. It was established that in experimental groups I, II and III, the Cobalt content in fermented pea straw was higher than in the control one by 5.5, 16.6 and 27.7%, respectively. With the use of biodestructor in doses of 14.0 and 28.0 cm3/t, a tendency to increase the Ferrum content in wheat straw has been established. The content of Zinc in composted wheat straw biomass from II and III experimental groups increased by a statistically significant value compared to the control one and the indicator in unfermented wheat straw. Fermentation with the participation of a biodestructor contributed to the increase of Manganese in wheat straw. Manganese content in fermented wheat straw has been increased by a statistically significant value relative to the indicator in the control one. Pazderska","doi":"10.36359/scivp.2023-24-2.28","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-2.28","url":null,"abstract":"The main field of activity of the National Reference Laboratory of Veterinary Drug Residues Control is the expert evaluation of honey safety based on the parameter of antimicrobial drug residues in honey bathes exported from Ukraine to European and American countries. The content of streptomycin and its derivative dihydrostreptomycin is one of the safety parameters for honey, which is investigated by enzyme-linked immunosorbent assay (ELISA). Since 2002, we have developed and proposed the clear scheme to control export honey batches on the content of residual amounts of these aminoglycoside antibiotics by means of Ridascreen® Streptomycin R3104 test kit from R-Biopharm (Germany). The test kit has been validated at 2.0 and 5.0 µg/kg taking into account the minimum required performance limit (MRPL) or the set reference point for action (RPA) according to European Commission Decision 2002/657/EC and 2021/808/EC. The fitness-for-purpose of the methods was confirmed based on the establishing of main validation parameters for screening methods (technical threshold and cut-off factor) using control (blank) honey samples and spiked with streptomycin standard solutions at the level of ½ MRPL by means of “added – found” approach. The preparation of samples is based on the preliminary purification by the method of solid-phase extraction using C18 columns, which facilitates the purification the sample extract from the influence of matrix components as well as the additional concentration of the analyte. The article also presents the results of the verification and validation of the Streptomycin NA01904H test kit from Kwinbon Biotechnology (China). The results of the assay of control (blank) honey samples and spiked with streptomycin standard solutions at the level of 2.0 and 5.0 μg/kg are given. Ensuring a high level of production of livestock products largely depends on the quality of feed, which is determined, first of all, by toxicological control. C-phycocyanin is a protein complex that belongs to phycobiliproteins and can be used as one of the potential natural components to improve the feeding of commercial fish. In order to establish the safety of the use of active feed components for feeding and ready-made feed into which these components are introduced, first of all, it is necessary to study their effect on the organisms of protozoa and fish, which are used for laboratory research. The high sensitivity of ciliates to the appearance of toxic substances in their environment and changes in their concentration led to the use of these organisms for testing the most diverse chemical elements and compounds. The article presents the results of studying the effect (harmlessness) of C-phycocyanin on the bodies of protozoa (Paramecium caudatum) and guppy fish. When studying the effect of C-phycocyanin on the body of protozoa (Paramecium caudatum), it was established that its use for 48 hours in doses of 5, 10, 25, 50, and 100 mg/l did not cause the death of ciliates. When studying the effect of C-phycocyanin on the body of guppy fish, it was established that its use in doses of 5, 10, 25, 50, and 100 mg/l did not cause the death of fish. So, on the basis of the conducted research, it was established that C-phycocyanin belongs to class 4, and is a non-toxic substance for Paramecium caudatum ciliates. Mamchur","doi":"10.36359/scivp.2023-24-2.25","DOIUrl":"https://doi.org/10.36359/scivp.2023-24-2.25","url":null,"abstract":"Some aspects of feeding dairy cows and factors affecting the production of high-quality milk suitable for the production of cheeses are considered, in particular, the study of the possibility of full or partial replacement of corn silage with silage from one-year forage crops in the rations. The study was conducted on cows of the Ukrainian black and spotted dairy breed using the method of periods. In the first period (duration 36 days), in addition to the main diet, the animals were fed 25 kg of corn silage, in the second (29 days) - 17 kg of hay from one-year forage crops, and in the third (35 days) - 12 kg of corn silage + 10 kg hay one-year forage crops. The main ration consisted of cereal and legume hay, winter wheat straw, fodder beets, a mixture of grain concentrates and premix. Density, content of dry matter, fat, total protein and casein and its fractional composition, lactose, degree of bacterial contamination, coagulation time of milk under the action of rennet enzyme were determined in milk. At the end of each period, batches of cheese were produced, the acidity, content of dry matter, fat, protein, degree of maturity were determined in the samples, and their expert evaluation was carried out. Since for the production of high-quality cheeses, milk must meet certain organoleptic parameters, chemical composition, bacterial contamination, the article provides requirements for its quality, as well as recommended rations and composition of premixes. Insecticidal efficiency was determined on groups of cattle of different sex and age groups. The research was carried out during the period of maximum intensity of infestation by ectoparasites. Counting of insects on the body of animals was carried out before their treatment, as well as on the 2nd, 7th, 15th and 30th day. According to the obtained data, in cows treated with a preparation based on cyfluthrin, the extent of bovicolosis invasion decreased already on the 2nd day and amounted to 23.3%, in the group of heifers of parous age, on the 2nd day after treatment, the extent of invasion was 22.2%, in the group of young animals, they noted a decrease in the extent of bovicolosis infestation on the 2nd day after treatment - 77.8%. The extent of invasion due to siphunculatosis in the group of cows treated with the drug on the 2nd day was 20.0%, in the group of heifers of mating age - 22.2%, in the group of young calves - 88.9%. The rate of extensibility for the treatment of groups of cows, heifers of mating age, as well as young animals on the 30th day of the experiment was 90.0, 91.5, and 91.1%, respectively. In cows treated with cyfluthrin, the intensity of invasion due to bovicolosis decreased already on the 2nd day and amounted to 43±3.1 exps/100 m2 of the skin of the animal’s body, in the group of heifers of mating age, the intensity of invasion was 46±2.1 exps/100 m2, in the group of young animals – 32±3.3 copies/100 m2. Intensity efficiency for processing groups of cows, heifers of mating age, as well as young animals on the 30th day of the experiment was 87.5%, 84.3% and 90.7%, respectively. The conducted set of studies established that when determining the effectiveness of cyfluthrin against bovicolosis and syfunculatosis, the intensity of treatment of groups of cows infested with hair-eaters on the 30th day of the experiment was 87.5%, among heifers of mating age, as well as young animals - 84.3 and 90.7 % respectively. The intensity efficiency indicator for the treatment of groups of cows, heifers of mating age, as well as young animals infested with lice, on the 30th day of the experiment was 87.0, 91.8 and 84.5%, respectively. The article presents data on the development of an ecologically safe method of sanitation treatment of the water supply system of poultry houses. For sanitation treatment of the water supply system, the alkaline detergent and disinfectant \"Sandez\" was used, which is an aqueous solution of alkali, does not contain anti-corrosion substances and compounds that create a potential threat of negative impact on the environment. The active substance of the drug \"Sandez\" - sodium hydroxide causes denaturation of protoplasmic proteins of microbial cells. In the experiment, the water supply system of the poultry house was filled with a 1.5% solution of \"Sandez\" disinfectant and left filled with this solution for after standing for 1 hour, the aqueous solution was drained and the pipelines were washed with clean tap water. In the control, sodium hypochlorite with a concentration of 75–100 mg/dm3 of active chlorine was used to disinfect the water supply network. Environmentally safe disinfectant \"Sandez\" ensures compliance of water quality and safety indicators with the requirements of state sanitary standards and regulations. \"Sandez\" product has an excellent washing effect with water hardness up to 8 mg-eq/l, has a low corrosive effect on metal structures of technological equipment. And S-methoprene is an analogue of the juvenile hormone, which leads to the arrest of the development and reproduction of insects. These active substances are used in insect-acaricidal drops to destroy and prevent the reproduction of lice, fleas and ticks in pets. Aim of the work was to develop a method for the identification and simultaneous quantification of fipronil and S-methoprene in drops for external use. The method was developed and validated by indicators of selectivity, linearity and suitability parameters of the chromatographic system. Insecto-acaricidal drops for external use were used as a sample-object for development. The standard sample and the test sample were dissolved in acetonitrile to fipronil concentration – 100 μg/ml and S-methoprene – 90 μg/ml. The samples were separated on a Dionex Ultimate 3000 chromatograph equipped with an Acclaim C18 chromatographic column 150×4.6, 5 μm. The mobile phase was a mixture of acetonitrile and water in a volume ratio of 90:10. Fipronil and S-methoprene were detected at an absorption wavelength of 270 nm. Under the above-mentioned conditions, it was possible to completely separate fipronil and S-methoprene (retention time of the chromatographic peaks - 2.3 min and 7.6 min) and other components of the studied drug. At the same time, the suitability parameters of the chromatographic system did not exceed the limits specified in the recommendations of the USA Food and Drug Association for HPLC methods. For fipronil and S-methoprene peaks the efficiency of the chromatographic system was 8300–13000 theor. plates The relative standard deviation (RSD) for the peak areas of the active substances was ± 0.50%, and the peak separation ratio (RS) of fipronil and S-methoprene and other drug components was 27.6. The symmetry coefficient of the fipronil peak was 1.4, and that of S-methoprene was 1.2. The calibration curves were linear in the recommended DFU 2.0 range (80–120% of the nominal concentration of the corresponding active substance).
