The causative agent was identified by cytopathology, routine mycological methods, and DNA sequencing of the internal transcribed spacer (ITS) region of rDNA. Apart from COVID-19 complications and the corticosteroid therapy, no underlying condition was diagnosed. The symptoms suggesting the fungal infection were shown two weeks after being discharged from COVID-19 hospital stay. Magnetic resonance of the brain showed a multi-focal central nervous system infection. However, the polymicrobial biofilm of both microorganisms has not been fully characterized.</p></div><div><h3>Aims</h3><p>To characterize the polymicrobial biofilm of <em>C. albicans</em> and <em>S. aureus</em> in terms of microbial density, synergy, composition, structure, and stability against antimicrobials and chemical agents.</p></div><div><h3>Methods</h3><p>Crystal violet assay was used to measure the biofilm formation. Scanning electron microscopy and confocal microscopy were used to analyze the structure and chemical composition of the biofilms, respectively.</p></div><div><h3>Results</h3><p>Supplemented media with fetal bovine serum (FBS) decreased the biofilm formation of <em>S. aureus</em> and the polymicrobial biofilm. For <em>C. albicans</em>, depending on the culture media, the addition of glucose or FBS had a positive effect in biofilm formation. FBS decreased the adhesion to polystyrene wells for both microorganisms. Supplementing the media with glucose and FBS enhanced the growth of <em>C. albicans</em> and <em>S. aureus</em>, respectively. It seems that <em>C. albicans</em> contributes the most to the adhesion process and to the general structure of the biofilms on all the surfaces tested, including a catheter model. Interestingly, <em>S. aureus</em> showed a great adhesion capacity to the surface of <em>C. albicans</em> in the biofilms. Proteins and β-1,6-linked polysaccharides seem to be the most important molecules in the polymicrobial biofilm.</p></div><div><h3>Conclusions</h3><p>The polymicrobial biofilm had a complex structure, with <em>C. albicans</em> serving as a scaffold where <em>S. aureus</em> adheres, preferentially to the hyphal form of the fungus. Therefore, a correct identification is important when evaluating the impact of such species in aspergillosis.</p></div><div><h3>Aims</h3><p>We aimed to describe the frequency, clinical and microbiological characteristics, and the outcomes of those cases of aspergillosis caused by cryptic species in a tertiary hospital.</p></div><div><h3>Methods</h3><p>We retrospectively identified all microbiologically documented cases of aspergillosis between January 2013 and December 2018. Definitive species identification of clinically significant isolates was achieved via sequencing methods. The polymerase chain reaction (PCR) products were sequenced, and the results obtained were compared to sequences deposited in GenBank. Antifungal susceptibility testing was performed using the Sensititre® YeastOne® panel.</p></div><div><h3>Results</h3><p>A total of 679 <span><em>Aspergillus</em></span> isolates were recovered from 489 patients, of which 109 were clinically relevant. Ten (9.2%) isolates were identified as cryptic species: <em>Aspergillus arcoverdensis</em> (2), <em>Aspergillus lentulus</em> (2), <em>Aspergillus ellipticus</em> (2), <em>Aspergillus alliaceus</em> (1), <em>Aspergillus nomius</em> (1), <em>Aspergillus tubingensis</em> (1) and <em>Aspergillus montevidensis</em> (1). Most patients already suffered some type of immunosuppression. Half of these patients had required intensive care before the infection showed up, and most of them had a pulmonary infection. Mortality at the 100-day follow-up was 40%. Antifungal susceptibility testing was performed on three of the isolates (<em>A. arcoverdensis</em>, <em>A. tubingensis</em> and <em>A. nomius</em><span>), which showed high minimum inhibitory concentrations (MIC) for azoles and amphotericin B.</span></p></div><div><h3>Conclusions</h3><p>The frequency of cryptic species in our centre was 9.2%. This issue highlights the urgent need for new and improved antifungal drugs with a novel target.</p></div><div><h3>Aims</h3><p>In order to evaluate whether juglone can be served as an alternative antifungal to cure drug-resistant <em>Candida</em> infections, we studied the <em>in vitro</em> susceptibility of juglone against fluconazole-susceptible and -resistance <em>Candida</em> isolates, alone and in combination.</p></div><div><h3>Methods</h3><p>Antifungal susceptibility testing was performed according to the CLSI (Clinical and Laboratory Standards Institute) guidelines.</p></div><div><h3>Results</h3><p>Juglone exhibited the highest minimal inhibitory concentration (MIC) values, followed by fluconazole and nystatin. Voriconazole showed significantly better antifungal activity than juglone, fluconazole, and nystatin, with MIC<sub>50</sub> and MIC<sub>90</sub> of 0.031 and 0.5<!--> <!-->μg/mL. There were significant differences in MICs of fluconazole (<em>p</em> <!--><<!--> <!-->0.001) and juglone (<em>p</em> <!--><<!--> <!-->0.0003) between <em>Candida</em> <em>albicans</em> and the rest of the species. Combination of juglone with fluconazole revealed insignificant effects against fluconazole-susceptible and -resistant <em>Candida</em> isolates. During hospitalization, the patient displayed episodes of bacteremia by <em>Staphylococcus haemolyticus</em> and a possible urinary tract infection by <em>T. asahii</em>. While the bacterial infection was successfully treated using broad-spectrum antibiotics, the fungal infection in the urinary tract was unsuccessfully treated with anidulafungin and persisted until the patient died.</p></div><div><h3>Conclusions</h3><p>With the evolving COVID-19 pandemic, invasive fungal infections have been increasingly reported, mainly after taking immunosuppressant drugs associated with long-term broad-spectrum antibiotic therapy. Although <em>Candida</em> and <em>Aspergillus</em> are still the most prevalent invasive fungi, <em>T. asahii</em> and other agents have emerged in critically ill patients.
