The plasma samples were extracted with methanol and chromatographic separation was achieved on phenomenex C18 (250 x 4.6mm, 5μ) analytical column with a mobile phase of methanol: 0.2% TEA (pH adjusted to 3 with orthophosphoric acid) in the ratio of 78:22% v/v and detection at 227nm. Calibration curve was linear in the range of 100-600ng/ml and the r2 value was found to be 0.9906. The extraction efficiency of RGE from rat plasma at the concentration of 50, 100 and 150% was found to be 99.6%, 99.9% and 98.05%. The Intra-day precision and interday precision was found to be 0.14 - 0.777% and 0.0070% - 0.0572% respectively. The maximum concentration (Cmax) obtained for two brands ( selected brand 1 & 2) formulation are 57.04 and 51.99 ng/ml respectively. The half life (t1/2) of two brands are calculated as 1 h. Area under the curve AUC 0- t of brand 1 and brand 2 is calculated as 343.235 and 310.1775ng h/ml and AUC 0-∞ is calculated to be 465.3665 and 501.4235ng h/ml respectively. The current study intends to investigate the phytochemical and physicochemical analyses of Vaividanga Choornam, a traditional Siddha polyherbal composition. This Vaividanga choornam contains the ingredients like Vaividangam (Embelica ribesBurm.), Milagu (Piper nigrumL.), Chukku (Zingiber officinale Rosc.), Omam (Trachyspermum ammi (L) Sprague), and Nilavaagai leaves (Cassia angustifolia Vahl. - syn. Cassia senna). It treatspitha diseases, constipation, indigestion, flatulence, burning sensation, bronchial asthma, and Kapha diseases. It is also an effective medicine for treating intestinal helminths caused by heavy worm burdens in young children. This study aims to estimate the quality of Vaividanga Choornam by performing pharmacognostic studies, preliminary phytochemical analysis, physiochemical parameters like ash value, extractive value, and loss on drying as per pharmacopoeial laboratory for Indian medicine guidelines, and also with the help of tests done with High-Performance Thin Layer Chromatography (HPTLC), FTIR Spectroscopy, and UV -Vis spectrometric analyses. The results obtained fromthose analyses showed the presence of various phytochemicals. The majority of herbal remedies and the products that are derived from them are frequently made from unrefined plant extracts, which include a complex mixture of various phytochemical components (plant secondary metabolites). The chemical characteristics of these components vary greatly between species. An intriguing device for determining the concentration of various active constituents in plants utilised in the cosmetic, medicine, the agriculture or medical sectors is the GC-MS method employed for the assessment of the extracted samples. The entire plant methanol extract of Tinospora was used in this work to identify bioactive elements employing gas chromatography and mass spectrometry (GC-MS). The GC-MS results suggest several different chemicals, including (9E,11E)-Octadecadienoic acid , 9-Octadecenoic acid, (E)-, 9-Octadecenoic acid, Octadecanoic acid, .gamma.-Sitosterol, n-Hexadecanoic acid, 9-Octadecenoic acid (Z)-, 2,3-dihydroxypropyl ester, Methyl cis-4,7,10,13,16,19-Docosahexaenoate and 2-Hydrazino-2-imidazoline in the Tinospora methanolic extract. As a result, the bioactive compounds in the methanol extracts of this herb may have chemopreventive, antitumor, anti-microbial, antioxidants, and hypoglycaemic potential. Polymers play an important role as excipients in designing the dosage form and influence drug release without any side effects. Natural polymers are of growing interest in the utilization of mucosal drug delivery as a novel technique due to their ability to interact with the mucosal layer of the body, improve the effectiveness of the drug, and prolong the residence time of the dosage form at the site of absorption. The bioadhesive polymers can exert control over the rate and extent of drug release and are selected in the formulation of mucoadhesive drug delivery systems. The crude extract was obtained by soaking the sample with methanol, following concentration using a rotary evaporator. Trypan blue exclusion test was performed in five concentrations of extract to detect the anti-cancer characteristic of Camellia cuongiana. Morphological changes in the treatment cell were observed using a phase-contrast microscope. After 72 hours of treatment, the extracts inhibited cell proliferation, with IC50 values of 72.26±5.75µg/mL. Increasing the extract concentration decreased the number of living cells. The most frequently observed abnormal morphology was cell shrinkage. Infection with the dengue virus can cause different febrile symptoms, such as dengue fever (DD) and dengue haemorrhagic fever (DHF), with or without shock. The purpose of this study is to obtain a new compound from Zingiber officinale that is expected to have potential bioactivity against DENV-2 NS2B/NS3 serine protease. A computational approach was applied in this study; which began with docking of compounds into protein targets, followed by density functional theory, drug-likeness, and ADMET analysis. According to the calculation, it was determined that compound 9 has binding interactions with the active triad through amino acids His51, Asp75, and Ser135. Additionally, drug-likeness and ADMET analysis for compound 9 showed that it has optimal lipophilicity and, when administered orally, can achieve good bioavailability. Therefore, an alternative method is needed to help evaluate the treatment of TB patients. This study aimed to analyze the correlation of the Procalcitonin test with AFB and GeneXpert for evaluating the treatment of TB patients and the performance of Procalcitonin as a marker of TB patient treatment progress. A prospective cohort study was conducted from May to September 2022 at the West Nusa Tenggara General Hospital, Indonesia. Sputum and blood samples were collected from 36 patients who were confirmed positive for TB by GeneXpert MTB/RIF examination, then examined for procalcitonin and AFB before being given treatment and after the intensive phase of treatment. Procalcitonin tested with VIDAS Biomerieux and VIDAS BRAHMS PCT kit. Procalcitonin did not correlate with AFB (p=0.064, r= 0.327) and GeneXpert before treatment (p=0.169, r=0.245), but correlated with AFB (p=0.013, r=0.427) and GeneXpert MTB/RIF (p=0.020, r=0.405) after the intensive phase of treatment. Procalcitonin test with a cut-off value of 0.07 detected negative AFB cases after treatment with a sensitivity of 28.6 and a specificity of 96.2%. The procalcitonin cut-off value of 0.07 also detected negative Xpert MTB/RIF after treatment with a sensitivity of 16.7% and a specificity of 100%. The performance of Procalcitonin for detecting negative smear and negative Xpert MTB/RIF after the intensive phase of treatment is classified as having high specificity, but its sensitivity is still low. Alkaloids, tannins, and flavonoids found in cashew stembark exhibit broad-spectrum antibacterial activity, making them appropriate as an active ingredient in mouthwash. On the other hand, one of the key ingredients in mouthwash formulas is glycerin. It is also frequently used as humectant. This study aims to prove the effect of glycerin on the cashew stembark extract mouthwash on its antibacterial activity. This study used 4 formulas consisted of 7% concentration of cashew stembark extract added with 1% sorbitol, 1.5% peppermint, distilled water as solvent, and various concentration of glycerin for each formula. The clarity, color change, and taste were assessed at day 7. The growth inhibition of both gram positive (S. aureus) and gram negative (P. aeruginosa) bacteria was measured. The data obtained were tested using one-way ANOVA and LSD (α=0.05). All of the 4 formulas of the mouthwashes found to be relatively stable. Based on the results of the inhibition test, it can be concluded that the addition of glycerin affects the inhibition of S. aureus and P. aeruginosa bacteria. The aim of this study is evaluation the antihypertensive effect of dapagliflozin in hypertensive type 2 diabetic patients (controlled/inadequately uncontrolled), and study its effect on contributing factors in that effectiveness (ex. weight loss and SNS damping). Primary endpoints included the effect of dapagliflozin on hemoglobin A1C (HbA1c) and blood pressure. Secondary endpoints included the effect of drug on weight body, pulse rate and pulse pressure. Patients were randomly selected from the outpatient of the Endocrinology Department at Tishreen University Hospital in Syria, and divided into two groups: dapagliflozin group (n=45) received 5 or 10mg dapagliflozin once daily as an add-on therapy to oral anti-diabetic drugs (OAD)and antihypertensive drugs, and control group (n=38) received OAD and antihypertensive drugs. After 12 weeks follow-up, dapagliflozin-treated versus control-treated showed significant reduction in HbA1C (-0.7% vs -0.3%, p<0.05, mean seated SBP (-9.4 vs -1.2mmHg, p=0.0001) and mean seated DBP(-6 vs-0.3mmHg, p=0.0001). Dapagliflozin also reduced body weight compared with control (-2.6 vs -0.6kg, p=0.0001) andthere was a significant positive correlation between weight loss and BP reduction. The effect on pulse rate was modest and similar between two groups but dapagliflozin have lowered pulse pressure values. There was no significant dose-response with any studied parameter, too. 5mg dapagliflozin once daily improved glycemic control, reduced body weight, SBP more than DBP and thus improved pulse pressure. Silver nanoparticles were biosynthesized through Streptomyces clavuligerus and Curcuma extract in addition to chemical reduction method with trisodium citrate as reducing agent silver nitrate as the metal substrate. The syntheses of the silver nanoparticles were characterized using UV-Vis spectroscopy and scanning electron microscopy (SEM). The results of characterization revealed absorption peaks obtained at range from 217-222 nm for all AgNPs types. SEM photographs of biosynthesized AgNPs indicate a narrow size distribution with average 8 nm in ‎spherical with smooth surfaces‎ and serruond protein but, chemical AgNPs showed ‎ without serruond protein. Antibacterial activity of AgNPs was determined using agar well diffusion method against Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia and Pseudomonas aeruginos. Results show similar significant antibacterial activity of microbial and Phyto- biosynthesized silver NPs. On the other hand, the chemically synthesized AgNPs recorded no activity against all tested pathogens. The cytotoxicity of AgNPs was tested in vitro on lung tissue fibroblast (WI-38) using MTT with various concentrations. The results recorded IC50 4.6, 1.25 and 2.18 µgml-1 for microbially, Phyto- and chemically AgNPs respectively.
