{"title":"Cannabinoid exposure: effects on development.","authors":"S L Dalterio","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The literature concerning the teratologic effects of cannabinoids is reviewed, and some methodological issues associated with perinatal cannabinoid exposure are discussed. The long-term consequences of perinatal cannabinoid exposure on brain, endocrine, immune, and hepatic functions are considered. In our studies, perinatal cannabinoid exposure affected the long-term development of body weight regulation, neuroendocrine function, and adult sexual behaviors. In addition, the immune system and hepatic cytochrome P-450 levels were also influenced in adult male mice perinatally-exposed to cannabinoids. It is hypothesized that these effects may be mediated by cannabinoid-induced alterations in the fetal and/or neonatal hormonal milieu. In addition, the possibility that perinatal cannabinoid administration affects the subsequent ability of the exposed offspring to adapt to the environment is discussed. Finally, possible mechanisms of cannabinoid action in altering development are evaluated. It is concluded that the evidence to date indicates that cannabinoids can be embryocidal, affect gestational length and labor, induce maturational delays, and that these substances affect a myriad of physiological processes in the developing offspring, including effects on behavioral parameters, not only in laboratory animals, but also in the human neonate. This article examines the interaction between perinatal drug research and the law. The reactivity of the law in assimilating research on exposure of the developing organism to a broad range of neurotoxins, including both licit and illicit drugs, is illustrated. Outcomes of some cases depend to a great extent upon legal concepts invoked rather than upon the scientific evidence involved. The types of behavior surveyed include activity, maze learning, steady state performance, and passive and active avoidance. Hormonal and neurochemical changes are discussed as well. Two opposing hypotheses to account for deficits following fetal drug exposure are presented and evidence for their validity is discussed. This study was designed to determine whether a lower blood ethanol concentration (BEC) or a brief exposure during the brain growth spurt in the rat might cause permanent brain damage. Two doses of ethanol (4 g/kg body weight daily during days 6-16 after birth and 6 g/kg on day 6 only after birth) were used. Significant differences in whole brain weight, a disproportionately decreased cerebellar weight, altered balancing ability, and a decreased number of cerebellar cells were observed on days 17 and 70 postnatally in both ethanol-treated groups as compared with controls. Cerebellar weight a both ages were more markedly affected in animals treated only on day 6 postnatally than in animals treated throughout the major portion of the brain growth spurt. This antinociception has been shown to be unrelated to neuromotor impairments and is modulated by environmental factors such as novelty of the test situation. Furthermore, behavioral observations revealed that TEL induced a profile of behavior during hot-plate testing which was unlike that produced by an equianalgesic dose of morphine (10 mg/kg). While morphine-injected rats were hypokinetic and remained in one location the rats injected with TEL vigorously explored the apparatus. It was subsequently found that manipulation of test variables such as familiarity with the apparatus and test environment could modify TEL's antinociceptive effects. For example, repeated exposure to a non-functional hot-plate as well as daily hot-plate testing attenuated the antinociception induced by TEL. Statistical analysis shows a significant negative relationship between the degree of in utero exposure to cadmium and lead and the child's motor and perceptual abilities. Any effect on memory or verbal skills was not statistically significant. Two learning tasks, one given during development and the other at maturity, also revealed no differences among groups. In addition, two activity level measures, time-sampled observations and the open field task, were taken on five occasions. These measures also failed to demonstrate any effects of food coloring. Blood lead levels of the mothers were less than 1, 24.4, and 37.4 micrograms/100 g blood, respectively, while those of the offspring were substantially higher, at least in the early stages of development. At the age of 12-15 months the animals were tested for group activity levels in an unfamiliar environment. No substantial lead-related alterations of activity occurred neither for group activity nor for the activity of individual animals. There were, however significant dose-related impairments of pattern discrimination learning set formation, while in simple discrimination learning during the early training phases deficits were seen in the high lead group only. Emotional alterations of these animals may account for this result, whereas true cognitive deficits are likely to underly the impairment of learning set formation seen in the low lead group.
