{"title":"Effects of melatonin on oxidative stress and the expression of ICAM-1 on retina in diabetic rat","authors":"吴昌凡, 薛嘉睿, 郎平","doi":"10.3969/J.ISSN.1003-0808.2010.04.010","DOIUrl":"https://doi.org/10.3969/J.ISSN.1003-0808.2010.04.010","url":null,"abstract":"目的 观察褪黑素对糖尿病(DM)大鼠氧化应激和视网膜组织中细胞间黏附分子-1(ICAM-1)表达的影响.方法 36只Wistar大鼠随机取10只作为正常对照组;其余26只制成DM模型,将造模成功的20只大鼠随机分为褪黑素组和DM组,每组10只.褪黑素组大鼠每日左下腹腔注射10mg/kg褪黑素进行干预治疗,正常对照组和DM组大鼠不进行治疗.12周后测各组大鼠的体重、血糖、血清一氧化氮(NO)及血清超氧化物歧化酶(SOD)值,并观察大鼠视网膜形态以及ICAM-1表达的不同.结果 12周后,与正常对照组比较,DM组和褪黑素组大鼠体重均明显下降(t=5.15,t=3.62;P<0.05),血糖均升高(t=69.03,t=45.78;P<0.05),而DM组和褪黑素组体重(t=1.88,P=0.09)和血糖(t=-1.41,P=0.19)比较,差异均无统计学意义.与正常对照组比较,DM组和褪黑素组大鼠血清中SOD、NO水平均明显下降(P<0.05);与DM组比较,褪黑素组大鼠血清SOD值、血清NO水平均明显升高(t=9.45,t=2.11,P<0.05).组织病理学检查显示,DM组和褪黑素组大鼠视网膜结构紊乱,白细胞黏附于血管内皮;免疫组织化学检测显示2组大鼠视网膜标本中ICAM-1颗粒表达于视网膜血管内皮细胞,但褪黑素组较DM组表达明显降低.结论 褪黑素可显著提高DM大鼠体内抗氧化能力、降低炎症反应,对DM引起的视网膜损伤有一定的保护作用。","PeriodicalId":186994,"journal":{"name":"Chinese Ophthalmic Research","volume":"348 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2010-04-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"123328206","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Zuo, Wei-li Geng, Ying Jin, Xiaoyan Yang, Li-qing Liu","doi":"10.3969/J.ISSN.1003-0808.2010.03.019","DOIUrl":"https://doi.org/10.3969/J.ISSN.1003-0808.2010.03.019","url":null,"abstract":"Background Laser in situ keratomileusis (LASIK) is frequently performed to reduce or eliminate myopic refractive error.Some patients complain of a loss of visual function after surgery even when they have 20/20 visual acuity.One of the reasons is the change of asphericity of the cornea.Objective This study attempts to investigate the short-term changes of the anterior and posterior corneal asphericity after LASIK.Methods One hundred and seven myopic eyes of 54 subjects with the equivalent spherical diopter of -1.30 to -7.50 D who have received LASIK were enrolled in this prospective study.The Q-values of the posterior corneal surface for different corneal diameters (6mm,7mm,8mm and 9mm) were measured with Pentacam preoperatively and 1 month postoperatively.The correlations between Q-value,Q change (△Q),and the mean preoperative spherical equivalent refraction (SE),central corneal thickness (CCT),central ablation depth (AD) and residual bed thickness were investigated.Written informed consents were obtained from all the subjects prior to the clinical trial.Results The Q-values of the anterior and posterior corneal surfaces gradually decreased to negative values with the increase of corneal diameter in myopic eyes.Weak correlations were found between the asphericity of the anterior and posterior corneal surfaces from diameters of 6mm,7mm,8mm and 9mm (r=0.227,0.288,0.303,0.389;P=0.019,0.003,0.002 and 0.000,respectively).No statistically significant correlation was found between Q-value and the diopter of refractive error (P>0.05).Both the anterior and posterior corneal Q-values varied toward the positive direction except that in the 9mm area of the posterior corneal surface at postoperative 1 month(t=-1.495,P=0.138).The increase of the anterior corneal asphericity (△Q) was more obvious than that of the posterior corneal surface and showed a positive correlation with ablation depth and a negative correlation with residual bed thickness.However,no statistically significant correlation was seen between △Q and these two parameters in the posterior corneal surface (P>0.05).Conclusion The shape of the anterior and posterior corneal surface shows more prolateness as the increase of corneal diameter in myopic eyes.Both the anterior and the posterior corneal surfaces have an oblate shift within the ablation zone 1 month after LASIK. \u0000 \u0000Key words: \u0000myopia; excimer laser; asphericity; Q-value","PeriodicalId":186994,"journal":{"name":"Chinese Ophthalmic Research","volume":"11 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2010-03-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131477946","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Mo, Wen-ji Wang, Hou-yan Song","doi":"10.3969/J.ISSN.1003-0808.2010.03.012","DOIUrl":"https://doi.org/10.3969/J.ISSN.1003-0808.2010.03.012","url":null,"abstract":"Background The vitreoretinal traction plays a critical role in the formation of macular hole and cystoid macular edema.Enzymatic vitreolysis has potential in relieving vitreoretinal traction as a simple and less invasive method in comparison with pars plane vitrectomy.ObjectiveThis study is to investigate the effects of plasmin mutant with kringle domains deficiency(Plm△K)on vitreoretinal interface in new Zealand white rabbits.Methods Plm△K was prepared through activating plasminogen mutant with Kringle domains deficiency (Plg△K) by tissue plasminogen activator (tPA).100μL of Plm△K at the dose of 0.5,1.0 and 1.5μmol/min was injected respectively into the vitreous of 48 New Zealand white rabbits and 16 eyes for each dose.B-scan and optical coherence tomography (OCT) were performed to detect the structure variety at the vitreoretinal interface in 1 day and 7 days after injection.The gross anatomy analysis with triamcinolone acetonide fine particle suspension,as well as histopathological examinations by scanning electron microscopy,was performed in the different time points mentioned above.Results Two peptide chains were determined with the relative molecular weight about 26000 and 5000 by the gel imaging analysis of reduced SDS-PAGE.Separation of the posterior vitreous cortex from retina was found after intravitreous injection under the B-scan and OCT.The ultrastructure change of vitreoretinal interface as well as the examination of fine particle suspension by triamcinolone acetonide demonstrated the same outcome.The degree of remnants of vitreous cortex showed the negative correlation with the dosage of Plm△K (r=-0.9516,P=0.048).No significant correlation was found between the degree of remnants of vitreous cortex and the action time(r=-0.720,P=0.470).There was no obvious morphological difference in outer layer of retina between control eyes and Plm△K-treated eyes.No drug-related adverse event was found after intravitreous injection of Plm△K.Conclusion Intravitreous injection of Plm△K alone can induce complete separation of vitreous from retina.This procedure is safe and effective. \u0000 \u0000Key words: \u0000vitreoretinal interface; plasmin; Kringle domain; deficiency mutant","PeriodicalId":186994,"journal":{"name":"Chinese Ophthalmic Research","volume":"389 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2010-03-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"131753429","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Objective This study is to investigate the effects of hypoxia on the expression of AQP-4 in Muller ceils in vitro. Methods Muller cells were isolated from New Zealand white rabbits and primarily cultured in DMEM containing 20% fetal bovine serum by the explant culture method. The cells were identified by immunostaining for the glial fibrillary acidic protein( GFAP). Generation 2 of cells was cultivated with the chemical hypoxia inducer, CoCl2 ,for 24 hours in the hypoxic group and only with DMEM in the control group. The expression of the AQP-4 protein in Mtiller cells was detected by immunocytochemistry. The expression of AQP-4 mRNA was determined by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results About 90% of Mtiller cells (generation 2) presented a positive immunoreactivity for GFAP, showing a brown staining in the cytoplasm. Cultured cells displayed the presence of intermediate filaments, microvillus and various cellular organs. The Integral absorbanee of the AQP-4 protein in Mtiller cells was markedly increased 24 hours after incubation with CoC12 in comparison with the control group (t = 6.74 ,P 〈 0.05 ). The expression level of AQP-4 mRNA in MUller cells was significantly enhanced 24 hours after incubation with CoC12 in comparison with the control group (t = 21.79, P 〈 0.05 ). Conclusion Hypoxia enchances the expression of AQP-4 in Mutiller cells and further increases fluid accumulation in the retina.
