Li, Lin-gang Xu, Zhiqi Chen, Rui-Lin Wang, Hong Zhang","doi":"10.3969/J.ISSN.1003-0808.2010.03.010","DOIUrl":"https://doi.org/10.3969/J.ISSN.1003-0808.2010.03.010","url":null,"abstract":"Background Endothelin-1 (ET-1) is an active regulator of intraocular pressure.The ET-1 level in aqueous humor is elevated in primary open-angle glaucoma,normal intraocular tension glaucoma and the animal model of glaucoma.There is now accumulating evidence for a role of ET-1 in the pathogenesis of glaucoma.However,the effect of ET-1 on the phagocytic function in trabecular meshwork cells (TMCs) is unclear.ObjectiveThis study is to observe the effect of ET-1 on the phagocytic function in cultured human TMCs.Methods Human trabecular meshwork tissue was obtained from healthy donator and cultured and subcultured in vitro by the explant culture method.The third passage of human TMCs were incubated with fluoresent red-labeled latex beads for 0,4,8,12,24,48 and 72 hours.The phagocytic kinetics of human TMCs were continuously evaluated by counting the number of latex beads in TMCs using a fluorescence microscope.Depending on the concentrations of ET-1 in culture medium,the TMCs were divided into control group (without ET-1),low-dose ET-1 (10~(-9)mol/L) treatment group,middle-dose ET-1 (10~(-8)mol/L) treatment group and high-dose ET-1 (10~(-7) mol/L) treatment group.In addition,based on the addition of endothelin receptor (ETAR) antagonist,the TMCs were divided into control group (without ETAR antagonist),ET-1 (10~(-8)mol/L) treatment group,ETAR antagonist (1×10~(-7)mol/L BQ123+10~(-8)mol/L ET-1) treatment group and ETBR antagonist(1×10~(-7) mol/L BQ788+10~(-8) mol/L ET-1)treatment group.TMCs of each group were incubated with latex beads,and the numbers of latex beads in TMCs were counted under a fluorescent microscope.Results Cultured HTM cells showed positive reactions for FN,LN,NSE and negative response for FⅧRag.The phagocytic kinetics test revealed that the latex beads were detected 4 hours after incubation.The density of latex beads was gradually increased with the delay of incubation duration and peaked at 24 hours.The number of the latex beads saturated after 48 hours of incubation.However,the number of latex beads in TMCs was significantly reduced after the addition of ET-1 in a dose-dependent manner (F=28.91,P<0.05).The number of latex beads in the ET-1 group was less than that in the control group and the ETAR receptor antagonist group (q=13.7228,q=9.4312,P<0.05).No significant difference was found in latex beads number between the ET-1 group and the ETBR antagonist group (q=1.1600,P>0.05).Conclusion ET-1 inhibits the phagocytic function of human TMCs and ETAR plays a partial role in the phagocytic function of human TMCs. \u0000 \u0000Key words: \u0000trabecular human TMCs; meshwork cells; endothelin-1; phagocytosis; latex beads","PeriodicalId":186994,"journal":{"name":"Chinese Ophthalmic Research","volume":"59 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2010-03-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"121636803","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Detection of Toric intraocular lens misalignment by digital photograph","authors":"Haike Guo, Haiying Jin, Hongyang Zhang","doi":"10.3969/J.ISSN.1003-0808.2010.02.016","DOIUrl":"https://doi.org/10.3969/J.ISSN.1003-0808.2010.02.016","url":null,"abstract":"Background Toric intraocular lens (Toric IOL) is a well accepted method of correct of corneal astigmatism.Axial misalignment,including the error of intraocular lens alignment during surgery and rotation of Toric IOL,is the main barrier to the precise alignment of Toric intraocular lenses.Therefore,the angle of axial misalignment is a main index to evaluate the effectiveness of Toric IOL implantation.Objective The present study is to describe a method of precisely detecting Toric IOL misalignment using postoperative digital photograph.Methods A measuring template (polar coordinates) was constructed using Adobe Photoshop software.Minimum graduate to detect the Toric axis was 1° (maximum error 0.5°).The clinical data of 24 eyes undergone Toric IOL implantation was retrospectively analyzed.Corneal astigmatism was detected by rotating Scheimpflug camera (Pentacam) or slit scan (Orbscan).Digital retro illumination picture was photographed after mydriasis.The picture was analyzed by our derived template.The steepest corneal axis and the axis of Toric IOL were then determined.The misalignment of Toric IOL was determined as the angle between the steepest corneal axis and the axis of Toric IOL.Misalignment of Toric IOL and its distribution were analyzed by statistical means.Results A method using polar coordinates and digital photo to precisely detect Toric IOL misalignment was derived.Mean Toric IOL misalignment was 10.93±6.90 degrees (0-24 degrees) in the 24 cases.The axial misalignment in all of cases were within 25 degrees,including clockwise in 14 eyes(58.33%),anticlockwise in 10 eyes (41.67%).In this study,the misalignment angle of 4 eyes were within 5 degrees;11 eyes were 5 to 10 degrees;1 eye was 10 to 15 degrees;6 eyes were 15 to 20 degrees;2 eyes were 20 to 25 degrees.Conclusion The method is feasible to the direction of Toric IOL misalignment in clinic. \u0000 \u0000Key words: \u0000digital photograph analysis; Toric intraocular lens; misalignment; corneal astigmatism","PeriodicalId":186994,"journal":{"name":"Chinese Ophthalmic Research","volume":"35 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2010-02-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"132848651","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

{"title":"Electrophysiological evaluation of injection of tissue plasminogen activator into optic nerve","authors":"Cai-hui Jiang, Mao‐nian Zhang, Kamei Motohiro","doi":"10.3969/J.ISSN.1003-0808.2010.02.007","DOIUrl":"https://doi.org/10.3969/J.ISSN.1003-0808.2010.02.007","url":null,"abstract":"Background Retinal vein occlusion (RVO) is a common vasculoretinopathy in ophthalmology.No an effective treating way is confirmed so far,and some new approach is in exploration.Objective The present study aims to investigate the safety and feasibility of injection of tissue plasminogen activator into optic nerve for the treatment of RVO using flash visual evoked potential (F-VEP) and flash electroretinogram(F-ERG).Methods The recording electrodes for F-VEP were implanted into the skull of rabbits to investigate the safety and feasibility of injection of tPA into the optic nerve.25μg of tPA or 12.5μg tPA (0.1 mL) was injected in pars plane via the center of optical disc respectively in different groups (6 eyes in each group),and 0.1 mL of BSS was injected at the same method in control group.Other 6 eyes were as normal control group.Slit lamp biomicroscope,indirect ophthalmoscope,F-VEP and F-ERG examinations were performed in 1 day,3,4,14 and 28 days after injection.All experiments were conducted in accordance with the Association for Research in Vision and Ophthalmology statement on the use of animals in ophthalmic and Vision Research.Results No obvious evidence of optic nerve or retinal toxicity or damage was found by the examination of ophthalmoscope,F-VEP,and F-ERG after injection of tPA.No significantly differences were found in the mean latency of P_1 wave of F-VEP (P=0.411),the mean amplitude of P_1 wave of F-VEP (P=0.065),the mean latency and amplitude of ERG a-waves (P=0.627,P=0.725) as well as the mean latency and amplitude of ERG b-waves (P=0.257,P=0.096) among 25 μg of tPA group,12.5 μg tPA group,BSS group and control group.Conclusion The injection of tPA into optic nerve is a safe and simple procedure.This therapy may is a potential approach to the treatment of RVO. \u0000 \u0000Key words: \u0000tissue plasminogen activator; optic disc; retinal vein occlusion","PeriodicalId":186994,"journal":{"name":"Chinese Ophthalmic Research","volume":"10 6 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2010-02-10","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"127078459","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
