Hormone and chemotherapy are the standard of care for advanced prostate cancer, but most patients develop resistance. Therefore, new therapeutic strategies are needed to improve the clinical management of prostate cancer. Low circulating levels of the secosteroid hormone vitamin D, and reduced expression of its receptor in prostate epithelial cells correlate with prostate cancer severity. However, the mechanisms underlying the anticancer effects of vitamin D are poorly understood. Our recent work demonstrates the therapeutic effects of a vitamin D analog on the tumor microenvironment in Pten<sup>(i)pe-/-</sup> mice, a mouse model of prostate cancer. In addition, we have shown that the combination of a vitamin D analog with the chemotherapeutic agent docetaxel, overcomes chemoresistance in primary prostate cancer spheroids, and in xenografts derived from a patient with prostate cancer resistant to docetaxel and to androgen deprivation. Therefore, therapeutic strategies based on the use of vitamin D analogs represent new avenues for advanced prostate cancer.</p>","PeriodicalId":18205,"journal":{"name":"M S-medecine Sciences","volume":"41 2","pages":"154-159"},"PeriodicalIF":0.6,"publicationDate":"2025-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"143542493","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The MITOMICS project thus contributes to the consolidation of a French \"mitochondrial medicine\", a notion that deserves to be examined. With the upcoming launch of the fourth national plan for rare diseases, it is an example of the study and management of rare and ultrarare diseases in France. This article traces the emergence of mitochondrial medicine since the early 1960s. It presents its main characteristics (genocentrism, strong techno-dependence), as well as its major technical and theoretical limitations, with a view to developing personalized mitochondrial medicine for the years to come.</p>","PeriodicalId":18205,"journal":{"name":"M S-medecine Sciences","volume":"41 2","pages":"173-179"},"PeriodicalIF":0.6,"publicationDate":"2025-02-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"143542480","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Many therapeutic approaches aim to reactivate the expression of the fetal hemoglobin genes. To this end, the CRISPR/Cas9 system has recently been used to genetically modify patients' hematopoietic stem/progenitor cells ex vivo and reactivate fetal hemoglobin expression in their erythroid progeny. More than 70 patients with severe β-thalassemia and sickle cell disease have been treated with the Casgevy® therapy. Most have achieved a significant improvement of clinical phenotype, with high editing efficiency in hematopoietic cells associated with normal or near normal hemoglobin levels. While the long-term safety and efficacy of this powerful approach still need to be evaluated, new strategies are being developed to further improve therapeutic outcomes, reduce potential genotoxicity and lower the costs of therapy.</p>","PeriodicalId":18205,"journal":{"name":"M S-medecine Sciences","volume":"41 1","pages":"33-39"},"PeriodicalIF":0.6,"publicationDate":"2025-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"143066724","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":4,"RegionCategory":"医学","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
