A simplified sample preparation method was developed. Several types of commercially available mushrooms were analyzed for quantification of ergothioneine using a Cogent Diamond Hydride column (100 x 4.6 mm) at a flow rate of 1 mL/min, with a deionized water:acetonitrile:0.1% formic acid mobile phase using an aqueous normal phase (ANP) gradient. EGT was detected at 254 nm and was identified by matching retention times of the verified standard. The standard ergothioneine content ranged from 0.01 mg to 0.1 mg/mL with excellent linearity correlation (R2 = 0.99999). EGT recoveries were within 96.5–100.3%. It is the most employed technique for separating, identifying, and quantifying components of a mixture. While there are different types of chromatography, the success of all chromatography systems largely depends on the accuracy and precision of their instruments. For the length of my career, nearly every year ushers in giant steps in the systemic improvement of commercial systems’ accuracy, precision, and overall efficiencies. These “leaps” have frequently been attributable to the physical changes in our systems, such as materials, miniaturization, and chemistries. Today, we turn our attention to software, an essential component of chromatography instruments. To protect human health, agricultural and environmental soil require careful investigation, but preparing samples for gas chromatography–mass spectrometry (GC–MS) analysis is time-consuming, and mostly done manually. Accelerated solvent extraction (ASE) has been the preferred preparation method for the past few decades, and while we have seen advancements, the method remains manual. While beneficial for most applications, quantification of PFAS using low-resolution triple quadrupole instruments can be complicated by the presence of co-eluted interferences, which can result in false positives or inaccurate PFAS concentrations. They are used in a wide range of different industries because they repel water and oil, and they are heat-resistant. However, PFAS contamination has become a widespread global concern because of their ubiquitous occurrence, persistence in the environment and living organisms, and potential health risks. Governments around the world are continuously introducing and updating regulatory limits for PFAS in drinking water, other types of water, biota, and, most recently, food. Analyzing such a wide range of sample types for PFAS requires high sensitivity and selectivity, typically provided by methods based on advanced liquid chromatography–tandem quadrupole mass spectrometry (LC–MS/MS) instrumentation. However, one critical aspect to sampling and analyzing PFAS accurately is to avoid PFAS contamination during these processes. This paper emphasizes the significance of robust and reliable PFAS analysis to support efforts to mitigate the global PFAS crisis. Specifically, since 1987 (the year of the previous NAS report on this field), the number of faculty members readily identifiable as “separations researchers” has dropped by 40% in the top chemistry departments, and by 30% in the top chemical engineering departments. At the same time, national government funding organizations (such as the National Science Foundation [NSF]) have significantly reduced funding for separation science. All this occurs despite the continuing demand for separation scientists and engineers in the largest economic sectors of the United States. Because of the current ongoing global recession, we anticipated that many workers would be concerned about their current jobs and how to adjust to rising inflation. In this year’s LCGC Salary Survey, we sought to explore these topics. In addition to compiling information about the average salary trends and reported on how these trends compare to previous years, we focused this edition of our questionnaire on the current economy, asking respondents to offer their opinions on whether a recession is taking place where they reside, and what impact it is having on them mentally and financially.
