It was found that protein partitioning was influenced by different coexisting effects, such as hydrophobic and electrostatic interactions, related to the system parameters and physicochemical/surface properties of proteins. Moreover, a rigorous analysis and optimization of the significant forces for protein partitioning in PEG-salt ATPS was performed by design of experiments (DoE). Initially, KPP/NaCit, NaCl and PEG concentrations were considered as statistically significant factors by a fractional factorial design. Subsequently, the optimal values of the significant factors were determined by a central composite face-centered design coupled with response surface methodology. Furthermore, linear and quadratic models were obtained in the experimental designs, respectively, and evaluated by statistical regression analysis and analysis of variance. Finally, a validation of the quadratic model confirmed a good correlation between predicted and experimental results, thus verifying the validity of the model.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"11 1","pages":"1-16"},"PeriodicalIF":0.0,"publicationDate":"2017-12-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84307651","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

In the present study, a cystatin like a thiol proteinase inhibitor was isolated from chickpea using ammonium sulfate fractionation (40-60%) and gel filtration chromatography on a Sephacryl S-100 HR column with 21.09% yield and fold purification of 78.89. It gave a molecular mass of about 25.3 kDa as determined by SDS-PAGE, Dynamic Light Scattering (DLS), and from its gel filtration behavior. The inhibitor was found to have very low carbohydrate content (0.34%) and meager amount (1.45%) of sulfhydryl content. The Stokes radius, frictional, diffusion and sedimentation coefficients of CPC were 4.299 × 10-8 g/s, 22.82 A, 12.68 × 10-7 cm2 s-1 and 2.64 S, respectively. Kinetic analysis of CPC with thiol proteinases (papain, ficin, bromelain) revealed the reversible and competitive mode of inhibition with CPC showing the highest inhibitory efficiency against papain (Ki=0.82 nM) followed by ficin (Ki=17.6 nM) and then bromelain (Ki=41.3 nM). CPC possessed about 25.34% α-helical content as assessed by CD spectroscopy. UV-Visible and fluorescence spectra indicated that upon formation of papain-CPC complex there is a significant conformational change in one or both the proteins of the complex.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"14 1","pages":"1-13"},"PeriodicalIF":0.0,"publicationDate":"2017-11-07","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"88775827","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The chromatographic peaks of the eight organic acids were successfully identified by comparing their retention times and UV spectra with reference standards. Method validation was performed in terms of linearity, sensitivity, selectivity, accuracy, and precision. The limits of detection (LODs) for the instrument employed in these experiments ranged from 11 to 8,026 μg/kg, and the limits of quantification (LOQs) ranged from 40 to 26,755 μg/kg. The average recoveries of the eight organic acids ranged from 76.3 to 99.2%. Method replication resulted in intraday and interday peak area variation of <5.3%. The method developed was specific and reliable and is therefore suitable for the routine analysis of organic acids in animal feed.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"6 1","pages":"1-5"},"PeriodicalIF":0.0,"publicationDate":"2017-10-23","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"85415943","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The analysis of polyphenol profile and content was performed by HPLC-DAD/UV-Vis and in vitro bioassay of 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS•+) method was used to evaluate the radical scavenging activity. The HPLC analysis showed a great diversity of compounds. E. arborea presented high contents in flavonols (52%) and flavanols (25%) and the average content of antiradical activity higher than 85%. The polyphenols identified are often associated by literature as having preventive properties and protective effect against of degenerative and inflammatory processes, thus, our findings confirm the empirical health properties often associated to this endemic plant. Therefore, based in our study the E. arborea, can be used to extract and purify phytochemicals with potentially beneficial effects on health.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"19 1","pages":"1-5"},"PeriodicalIF":0.0,"publicationDate":"2017-09-05","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84593213","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
