The separation was carried out on Thermo Scientific AcclaimTM 120 C18 column with a mobile phase composed of methanol and water (containing 0.1% formic acid) in gradient mode at a flow rate of 0.6 mL/min. The limit of detection of 10-15 ng/mL and limit of quantitation of 29-45 ng/mL were determined. The method was evaluated in terms of recovery, precision, linearity, limit of detection, limit of quantification. The determined parameters are in the commonly acceptable ranges for that kind of analysis. The developed chromatographic method allows the rapid screening of urine metabolites, which can serve as potential markers of various tumor diseases as well as for the monitoring of treatment processes.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"43 1","pages":"1-8"},"PeriodicalIF":0.0,"publicationDate":"2018-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"88855527","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Its effect is bactericidal, and due its action, it is the most efficient first-generation cephalosporin against resistant microorganism (β-lactamase producers). Although this drug has been clearly studied and researched about its antimicrobial activity, pharmacokinetics and pharmacodynamics, there are a few studies in literature regarding the development of analytical methodology for this cephalosporin. The aim of this work was to develop and validate a new method of analysis, using high performance liquid chromatography, resulting in an innovative method, quick and using solvents of low toxicity, minimizing, in this way, its toxic actions to the operators and leavings in the environment. The method developed and validated for the quantification of sodium cephalothin in lyophilized powder for injectable solution used high performance liquid chromatography (HPLC). The mobile phase consisted in water with 0.7% of glacial acetic acid and ethanol (70:30 v/v), wave-length of 237 nm, Zorbax Eclipse Plus C18 AgilentTM column and room temperature of 25°C, retention time of 4,20 minutes. The method was linear in the concentrations of 20, 40, 60, 80 e 100 μg/mL, selective, accurate and robust towards these modifications: ethanol brand, water source, mobile phase rate, glacial acetic acid proportion, flow rate, room temperature and wave length. The dosing for CET was of 106.72%.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"13 1","pages":"1-8"},"PeriodicalIF":0.0,"publicationDate":"2018-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"85698255","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

There are a number of concerns on sample derivatization, method specificity, sensitivity, separation efficiency and long elution time, although a large number of methods using Liquid Chromatography-Mass Spectrometry (LC-MS) and Gas Chromatography-Mass Spectrometry (GC-MS) for determination of estrogens and metabolites have been published before. We successfully developed and validated an efficient Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) method for determination of fifteen estrogens and metabolites in human serum. The estrogens and metabolites were derivatized with dansyl chloride and analyzed by LC-MS/MS using positive Electrospray Ionization (ESI+) and a Multiple Reaction Monitoring (MRM) mode. A Phenomenex Synergi Hydro-RP 2.5 μm 80 Å column (100 × 2.0 mm) at 60°C was eluted with mobile phases consisting of acetonitrile, methanol, water and formic acid at a flow rate of 0.4 mL/min. All of the fifteen dansyl derivatives of estrogen and metabolites were separated by LC with retention time differences between peaks ≥ 0.2 min. The recoveries of the fifteen estrogens and metabolites from hydrolysis and derivatization were in a range of 74.4-95.6% at 30-801 pg/mL level. The interday accuracies of the dansyl derivatives were in a range of 91.7-109.6% with a precision of 2.7-12.6%RSD. The LC-MS/MS system had linear responses to these derivatives within the calibration range of 12-10980 pg/mL with regression coefficient r2 ≥ 0.9934. The Limits of Quantification (LOQ) of these derivatives were in a range of 5.3-71.1 pg/mL in human serum. In comparison to a typical published method, our method provided a more efficient LC separation using only one-third of the elution time, and a similar or slightly better sensitivity.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"12 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2018-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"78598367","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Optimal extraction conditions were ascertained by means of a multiple linear regression analysis and the generation of a response surface. Conditions to reach the equilibrium of VOC’s at the headspace were established to be 30 minutes of ultrasonic agitation at 50°C. The conditions for extraction were exposure of the fiber at room temperature for 30 minutes. Optimal extraction of volatile compounds released by the Oaxaca cheese allowed identification of 21 volatile compounds: 8 acids, 1 alcohol, 4 aldehydes, 4 ketones, 1 ester, and 3 lactones. Quantification of VOC’s was carried out using the technique of standard addition. The limits of quantification ranged from 0.0033 to 0.0311 ng/μg. Ten compounds were identified with the heaviest concentration and are listed as follows, from heaviest to lightest: propionic acid, acetoin, acetic acid, diacetyl, capric acid, caprylic acid, δ-decalactone, δ-dodecalactone, γ-dodecalactone, butyraldehyde. During storage of the cheese, the most significant changes in the concentration of VOC’s took place between day 1 and day 15. Based on the results obtained, it can be concluded that the optimization of the SPME makes it possible to isolate and quantify VOC’s of fresh cheeses in an efficient and reproducible manner.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"47 6 1","pages":"1-7"},"PeriodicalIF":0.0,"publicationDate":"2018-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"83549803","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The presence of heavy metals in water and wastewater causes toxic effects to the living beings and the environment. Compared to other heavy metals (such as Cr6+, Pb2+, Zn2+ etc.,) Chromium found in the tannery and electroplating industrial effluents causes serious effects to the humans and living organisms. It also contaminates the soil and groundwater. The excess amount of chromium affects the lungs and lead to respiratory disorders in the human beings. Due to a large number of tanneries and electroplating industries, the chromium contamination in the wastewater and aqueous solutions exceeds the tolerance limits. Many methods are used to remove the chromium from the aqueous solutions and industrial effluents. Adsorption is one of the cost - effective methods being widely used for the removal of heavy metals from industrial and commercial wastewaters. It has been established as an important and economically feasible treatment technology for removing heavy metals, particularly chromium. This review article presents the information about the application of different kinds of natural waste adsorbents for the removal of hexavalent chromium (Cr6+) from industrial effluent.","PeriodicalId":15534,"journal":{"name":"Journal of Chromatography & Separation Techniques","volume":"213 1","pages":"1-6"},"PeriodicalIF":0.0,"publicationDate":"2017-12-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"77503185","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
