Primary empiric treatment for tuberculosis comprises a four-drug regimen: Isoniazid, Rifampicin, Pyrazinamide, and Ethambutol. The disease and its treatment may pose undernutrition due to the increased metabolic load and decreased food intake. Immunomodulators and micronutrients are consumed by patients, worldwide, to counteract nutritional insufficiencies. Immusante ® is a polyherbal formulation recommended as an immunomodulator in various immunocompromised states, including tuberculosis. This study aims to identify the pharmacokinetic interactions between the first-line antituberculosis (anti-TB) drugs, that is, AKT-4 tablets (a combination of isoniazid, rifampicin, pyrazinamide, and ethambutol), and Immusante ® . We developed a method of sensitive liquid chromatography with tandem mass spectrometry for the simultaneous quantification of isoniazid, rifampicin, pyrazinamide, and ethambutol in rat plasma and validated it as per the European Medicines Agency guidelines. The method was used to quantify all four drugs in rat plasma treated with a combination of AKT-4 and Immusante ® . No significant alterations in the values of various pharmacokinetic parameters such as V d , t 1/2 , CL, t max , C max , and AUC were observed, indicating that coadministration of Immusante ® does not influence the pharmacokinetic profiles of the anti-TB drugs. Chamaenerion angustifolium (Onagraceae), commonly known as “fireweed,” is a traditional food and medicinal plant in Europe. Current research has focused on comparative analysis of the aerial part of 15 fireweed samples collected in Ukraine and Lithuania using Gas chromatography–mass spectrometry (GC-MS) method. The overall amino acid composition in samples of C. angustifolium was similar. Sulfur-containing amino acids (cysteine and methionine) were absent in all samples. The alanine content in the samples was the highest, and in the samples from Kharkiv (Ukraine), Ivano-Frankivsk (Ukraine), and Plungė district (Lithuania), it ranked first among others (its content in samples was 2.350, 6.090, and 2.44 mg/g, respectively). The high amount of free amino acids was recorded in the sample from Ivano-Frankivsk (Ukraine). The results of chemometric analysis indicated L-alanine and L-phenylalanine could be used as potential quality markers for the evaluation of the plant quality. The results indicate the potential for further pharmacological studies of fireweed raw material. In this study, we investigated the effect of diosmin, a flavonoid on the CYP2E1-mediated metabolism of paracetamol and NAPQI formation in Wistar rats. For 28 days, animals were orally administered with paracetamol (300 mg/kg) with and without diosmin (25 and 50 mg/kg). The blood samples were collected on the 28th day and the plasma concentrations of paracetamol and NAPQI were determined using the reverse phase high-pressure liquid chromatography. Paracetamol combination with diosmin (25 and 50 mg/kg) showed a dose-dependent increase in the area under the plasma concentration-time curve (AUC 0–∞ )and the peak plasma concentration ( C max ) of paracetamol and a dose-dependent decrease of AUC 0–∞ and C max of NAPQI compared to paracetamol control ( p < 0.001). Simultaneously, serum biomarkers and histopathological studies were conducted to evaluate the effect of diosmin on the liver and kidneys. Interestingly, compared to the paracetamol control group, the combination with diosmin significantly reduced the elevated functional biomarkers of liver and kidney. In addition, diosmin ameliorated paracetamol-induced hepatotoxicity and nephrotoxicity.
